A solution of 6-bromophthalazine-1(2H)-one (0.25 g, 1.11 mmol), potassium vinyl trifluoroborate (0.446 g, 3.33 mmol) and [0441] K2CO3 (0.46 g, 3.33 mmol) in DMSO (2 mL) was degassed with argon for 20 min at ambient temperature. PdCl2(dppf) (0.04 g, 0.055 mmol) was added at ambient temperature, and the reaction mixture was heated to 80 C. for 2 h. The reaction mixture was cooled to ambient temperature and filtered through celite bed under vacuum and washed with ethyl acetate. The reaction mixture was extracted with ethyl acetate and the combined ethyl acetate layer dried over Na2SO4 and concentrated under reduced pressure to afford the crude product. The crude compound was purified by column chromatography (SiO2, 100-200 mesh; 50% ethyl acetate/petroleum ether) to afford the title compound as a brown solid (0.12 g, 63%): 1H NMR (400 MHz, DMSO-d6) delta 13.61 (br, 1H), 8.33 (m, 1H), 8.19 (m, 1H), 8.01 (m, 2H), 6.97 (m, 1H), 6.15 (m, 1H), 5.56 (d, J=10.8 Hz, 1H); ESIMS m/z 172.93 ([M+H]+); IR (thin film) 1748, 1655, 3241 cm-1.

Under N2, acetic acid 2,6-dibromobenzyl ester (24.74 g, 80.3 mmol), 6-tert-butyl-8-fluoro-2H-phthazin-1-one (3.54 g, 16.1 mmol), Cs2CO3 (10.46 g, 32.1 mmol), CuI (4.61 g, 24.2 mmol) and N,N-dimethylethane-1,2-diamine (1.42 g, 16.1 mmol) were dissolved in DMF (70 mL). The reaction mixture was stirred at 150 C. for 4 h. The resulting mixture was cooled to room temperature and diluted with ethyl acetate (200 mL), and then 200 mL of water was added. The mixture was separated and the aqueous layer was extracted with ethyl acetate (200 mL¡Á2). The organic layers were combined and washed with water (200 mL) and brine (200 mL). The solution was dried over anhydrous sodium sulfate, filtered and concentrated. The resulting crude product was purified by silica gel chromatography (petroleum ether/ethyl acetate 4/1) to give 2-bromo-6-(6-tert-butyl-8-fluoro-1-oxo-1H-phthalazin-2-yl)-benzyl ester (3.21 g, 45%). 1H NMR (300 MHz, CDCl3): delta 8.17 (d, J=2.7 Hz, 1H), 7.71 (t, J=4.8 Hz, 1H), 7.50-7.45 (m, 2H), 7.36 (s, 1H), 7.34 (d, J=0.9 Hz, 1H), 5.17 (s, 2H), 1.94 (s, 3H), 1.42 (s, 9H). LC-MS calcd for C21H20BrFN2O3 (m/e) 446.06, obsd 447 and 449 [M+1]+.

5-Bromo-1, 3-isobenzofurandione (14. 8 g, 65.4 mmol) was suspended in acetic acid (150 mL), and treated with hydrazine hydrate at (10 mL) at 80 overnight. The mixture was cooled, filtered, and the residue was washed with methanol. The residue was triturated with 4M NAOH (ca. 250 mL), and filtered. The pale yellow filtrate was acidified with conc. HCL, and filtered. The residue was azeotroped with toluene (x4), and suspended in POC1s (75 mL). N, N-DIISOPROPYLETHYLAMINE (10 mL) was slowly added and the mixture was heated to reflux for 3 h and then concentrated under reduced pressure, suspended in chloroform and filtered. The filtrate was poured onto ice, and the organic phase was washed with sodium bicarbonate solution, followed by brine. The organic phase was dried (sodium sulfate) and filtered through a pad of silica, eluting with chloroform, which after evaporation gave an off-white solid (6.5 g, 36%)

Step 3: Preparation of 4-(4-fluoro-3-(4-(2-methyl-2-((5-(trifluoromethyl) pyridin-2- yl)oxy) propanoyl)piperazine-l-carbonyl) benzyl) phthalazin-l(2H)-one.To a solution of 2-methyl-2-((5-(trifluoromethyl)pyridin-2-yl)oxy)propanoic acid (0.204 g, 0.81 mmol) in dry DMF (6 mL) was added TBTU (0.289 g, 0.90 mmol) at room temperature under atmosphere of nitrogen. To this 4-(4-fluoro-3-(piperazine-l- carbonyl)benzyl)phthalazin-l(2H)-one (0.3 g, 0.81 mmol) and DIPEA (0.303 mL, 1.74 mmol) were added. The reaction mixture was stirred at room temperature for 2 h. The progress of reaction was checked by TLC by using mobile phase 5 % methanol in chloroform. The reaction mixture was diluted with ethyl acetate. The organic layer was washed with water, dried over anhydrous Na2S04 and solvents were evaporated on a rotatory evaporator under reduced pressure to crude solid which was purified by the flash column chromatography using eluent chloroform : methanol (98.3 : 1.7) to afford 4-(4-fluoro-3-(4-(2-methyl-2-((5-(trifluoromethyl) pyridin-2-yl)oxy) propanoyl)piperazine-l-carbonyl) benzyl) phthalazin-l(2H)-one as white solid (0.230 g, 47 %).

In a 25 mL container, 1-(3-bromo-2-formylphenyl)-1H-pyrazole-3-carbonitrile (50 mg, 181 mumol, Eq: 1.00), 6-tert-butyl-8-fluorophthalazin-1(2H)-one (79.8 mg, 362 mumol, Eq: 2) and copper (I) iodide (69.0 mg, 362 mumol, Eq: 2.00) were combined with DMSO (2.00 ml) to give a yellow suspension. Sodium bicarbonate (38.0 mg, 453 mumol, Eq: 2.5) was added to it. The mixture was heated in a microwave at 120 C. for 1 hr. The reaction mixture was poured into 25 mL sat NH4Cl and extracted with EtOAc (3¡Á25 mL). The organic layers were dried over MgSO4 and concentrated in vacuo. The organic layer was concentrated and purified through ISCO flash column chromatography using ethyl acetate (containing 5% methanol) in hexanes (5% to 80% linear gradient in 15 minutes, 12 g silica gel) to give the pure desired product which was triturated with ether in hexanes and filtered to obtain 1-(3-(6-tert-butyl-8-fluoro-1-oxophthalazin-2(1H)-yl)-2-formylphenyl)-1H-pyrazole-3-carbonitrile (19 mg, 25%).

The mixture was cooled to room temperature, a solution of (2-bromo-5- bromomethyl-4-fiuoro-phenyl)-methanol (203 mg, 0.68 mmol) in DMF (2 mL) was added and the mixture was stirred for 2.5 h at room temperature. Water (5 mL) was added, and the mixture was extracted with EtOAc (3 x 15 mL). The combined organic layers were dried (Na2S04), filtered, and evaporated. The residue was purified by preparative HPLC to give 2-(4-bromo-2- fluoro-5-hydroxymethyl-benzyl)-6-tert-butyl-8-fluoro-2H-phthalazin-l-one (59 mg, 10%). MS calcd. for CzoftoB^NzC^ [(M+H)+] 437, obsd. 436.8.
