Homogeneous Composite Nonwoven Support for High Temperature-Resistant Separation Membranes

High temperature-resistant separation membranes are widely considered in various fields. However, incompetent support layer limits the performance of high temperature-resistant membranes severely. In this work, a homogeneous composite nonwoven support (HCNS) is developed by wet-laid and hot-pressing technology using undrawn polyethylene terephthalate fibers as bonding fibers. The significance of hot-pressing parameters on the performances of the HCNS is investigated, and quadratic regression models are established. The HCNS exhibits a smooth surface (roughness of 118 nm) and a porous structure (porosity of 21.06% and pore size of 6.548 mu m) with a mechanical property (tensile index of 45.80 N m g(-1)). The ultrafiltration membranes supported by the HCNS are prepared from poly(phthalazine ether sulfone ketone), which demonstrates high permeance, high rejection rate, and long-term stability at raised operation temperature, thereby suggesting the potential application of HCNS in high-temperature filtration.

Synthesis and evaluation of novel dimethylpyridazine derivatives as hedgehog signaling pathway inhibitors

We report herein the design and synthesis of a series of structural modified dimethylpyridazine compounds as novel hedgehog signaling pathway inhibitors. The bicyclic phthalazine core and 4-methylamino- piperidine moiety of Taladegib were replaced with dimethylpyridazine and different azacycle building blocks, respectively. The in vitro Gli-luciferase assay results demonstrate that the new scaffold still retained potent inhibitory potency. Piperidin-4-amine moiety was found to be the best linker between pharmacophores dimethylpyridazine and fluorine substituted benzoyl group. Furthermore, the optimization of 1-methyl-1H-pyrazol and 4-fluoro-2-(trifluoromethyl) benzamide by different aliphatic or aromatic rings were also investigated and the SAR were described. Several new derivatives were found to show potent Hh signaling inhibitory activity with nanomolar IC50 values. Among these compounds, compound 11c showed the highest inhibitory potency with an IC50 value of 2.33 nM, which was comparable to the lead compound Taladegib. In vivo efficacy of 11c in a ptch(+/)-p53(-/-) mouse medulloblastoma allograft model also indicated encouraging results. (C) 2018 Elsevier Ltd. All rights reserved.

Synthesis of hydralazine-N-lactose and its metabolites in vitro

Hydralazine-N-lactose is a main impurity in hydralazine tablets. Hydralazine and lactose were used as raw materials, and silica gel column and preparation column of octadecylsilane bonded silica were selected to prepare and purify hydralazine-N-lactose. Its in vitro metabolites were identified by utilizing SD rat liver microsomes. The results of samples in vitro were compared with blank group and the resulting metabolites structures were identified by LC-MS/MS. Hydrazine-N-lactose produced six main metabolites, including s-triazolo[3, 4-a] phthalazine, 3 -methyl-s-triazolo [3, 4-a] phthalazine, 3 -(1-hydroxy) methyl-s-triazolo [3, 4-a] phthalazine, 3 hydroxymethyl-s-triazolo[ 3, 4-a] phthalazine, hydralazine and dehydro-hydralazine-N-lactose after in vitro metabolism. This study will be valuable in hydralazine-lactose conjunctions metabolism research and hydralazine formulation safety.

Comparative study on the interaction of two binuclear Pt (II) complexes with human serum albumin: Spectroscopic and docking simulation assessments

Human serum albumin (HSA) principally tasks as a transport carrier for a vast variety of natural compounds and pharmaceutical drugs. In the present study, two structurally related binuclear Pt (II) complexes containing cis, cis-[Me2Pt (mu-NN) (mu-dppm) PtMe2] (1), and cis, cis-[Me2Pt(mu-NN)(mu dppm) Pt((CH2)(4))] (2) in which NN = phthalazine and dppm = bis (diphenylphosphino) methane were used to investigate their interaction with HSA, using UV Vis absorption spectroscopy, fluorescence, circular dichroism and molecular dynamic analyses. The spectroscopic results suggest that upon binding to HSA, the binuclear Pt (II) complexes could effectively induce structural alteration of this protein. These complexes can bind to HSA with the binding affinities of the following order: complex 2 > complex 1. Moreover, the thermodynamic parameters of binding between these complexes and HSA suggested the existence of entropy-driven spontaneous interaction, which mostly dominated with the hydrophobic forces. The ANS fluorescence results also indicated that two binuclear Pt (II) complexes were competing for the binding to the hydrophobic regions on HSA. In addition, competitive displacement assay and docking simulation study revealed that complexes 1 and 2 bind to the drug binding sites II and Ion HSA, respectively. Furthermore, complex 2, with the higher binding affinity for HSA, shows more denaturing effect on this protein. Considering the protein structural damages in the pathway of harmful side effects of platinum drugs, complex 1 with the moderate binding affinity and low denaturing effect might be of high significance. A mesoporous SBA-15 silica catalyst functionalized with phenylsulfonic acid groups (SBA-15-Ph-SO3H) as a novel hydrophobic nanoreactor solid acid catalyst for a one-pot three-component synthesis of 2H-indazolo[2,1-b]phthalazine-triones and triazolo[1,2-a]indazole-triones

Phenyl sulfonic acid functionalized mesoporous SBA-15 silica (SBA-15-Ph-SO3H) was prepared through the silanization of activated mesoporous SBA-15 with diphenyldichlorosilane (DPCS) followed by silylation and sulfonation, and characterized by XRD, FT-IR, nitrogen adsorption analyses, BET theory and TEM. The sulfonic acid groups anchored to the silica surface of the pore walls were resistant to leaching in organic and aqueous solutions under mild conditions. This catalyst was found to be a recyclable heterogeneous catalyst for the rapid and efficient synthesis of various 2H-indazolo[2,1-b]phthalazine-triones and triazolo [1,2-a]indazole-triones. Also, this silylated mesoporous material containing phenylsulfonic acid groups has a greater stability of the sulfonic acid surface toward water than that of silica sulfuric acid (SSA) and sulfonated SBA-15.

Synthesis, crystal structure and antifungal activity of a Ni(II) complex of a new hydrazone derived from antihypertensive drug hydralazine hydrochloride

A new complex of nickel(II) with 1-(2-[(5-methylfuran-2yl)methylene)] hydrazono) phthalazine HL (1) has been synthesized and characterized by elemental analysis, IR, electronic spectra, TGA, H-1 and C-13 NMR spectroscopy. The crystal structures of the free ligand and its complex have been determined by single crystal X-ray diffraction techniques. In the complex the hydrazone ligand chelates to the nickel(II) through nitrogen atoms in a bidentate manner. The antifungal activity of the ligand and its nickel(II) complex 3 was studied against Aspergillus niger, Aspergillus flavus and Candida albicans. The results revealed modest activity of the complex (3) against the tested organisms with inhibition zones of 14, 14 and 13 mm, compared to the free ligand 1 with inhibition zones of 11 mm each for A. niger, A. flavus and C. albicans respectively. Highly efficient red phosphorescent organic light-emitting diodes based on solution processed emissive layer

Synthesis and Antitumor Activity of Novel 1,3-Disubstituted Pyridazinone Derivatives

In order to find more efficient and low toxicity antitumor drugs, a series of novel 1,3-disubstituted pyridazinone derivatives were synthesized and evaluated for their antiproliferative activities against four human cancer cell lines (MCF-7, PC-3, SW-620 and HGC-27) in vitro. The results showed that most compounds had good antiproliferative activities, especially 2-(4-(4-bromophenyl)-1-oxo-tolylazine-2(1H)-yl)-N-(2-fluorophenyl)acetamide (5g) exhibited better antiproliferative activities with IC50 value of 6.01 mu mol/L. In a nutshell, this work provided clues to discover antitumor agent based on the quinazoline scaffold.

Synthesis of new pyrrolo[2,1-a] phthalazine derivatives via multicomponent reaction of phthalazine with 1,1-dicyanoalkenes and alkyl isocyanides

Multicomponent reactions between phthalazine, alkyl isocyanides, and 1,1-dicyanoalkenes in acetonitrile-water mixture produced high yields of substituted pyrrolo[2,1-a]phthalazines, which were converted upon treatment with 10% hydrochloric acid to pyrrolo[2,1-a]-phthalazines unsubstituted at position 1.
