We report preclinical proof of principle for effective treatment of B-precursor acute lymphoblastic leukemia (ALL) by targeting the spleen tyrosine kinase (SYK)-dependent antiapoptotic blast cell survival machinery with a unique nanoscale pharmaceutical composition. This nanoscale liposomal formulation (NLF) contains the pentapeptide mimic 1,4-Bis (9-O dihydroquinidinyl) phthalazine/hydroquinidine 1,4-phathalazinediyl diether (C61) as the first and only selective inhibitor of the substrate binding P-site of SYK. The C61 NLF exhibited a very favorable pharmacokinetic and safety profile in mice, induced apoptosis in primary B-precursor ALL blast cells taken directly from patients as well as in vivo clonogenic ALL xenograft cells, destroyed the in vivo clonogenic fraction of ALL blast cells, and, at nontoxic dose levels, exhibited potent in vivo antileukemic activity against patient-derived ALL cells in xenograft models of aggressive B-precursor ALL. Our findings establish SYK as an attractive molecular target for therapy of B-precursor ALL. Further development of the C61 NLF may provide the foundation for therapeutic innovation against therapy-refractory B-precursor ALL.

Synthesis and evaluation of anti-proliferative activity of 1,4-disubstituted phthalazines

A series of new phthalazine derivatives 3a-i and 4a-c were synthesized via the reaction of 1-chlorophthalazine derivative 2 with either N-substituted-piperazines, primary or their secondary amines. The structure of the synthesized, new compounds were characterized by spectral data. The anti-proliferative activity on human breast cancer cell line MCF-7 of the synthesized compounds was determined. Anticancer activity assessment of two novel binuclear platinum (II) complexes

In the current study, two binuclear Pt (II) complexes, containing cis, cis-[Me2Pt (mu-NN) (mu-dppm) PtMe2] (1), and cis,cis-[Me2Pt(mu-NN)(mu dppm,) Pt((CH2)(4))] (2) in which NN = phthalazine and dppm = bis (diphenylphosphino) methane were evaluated for their anticancer activities and DNA/purine nucleotide binding properties. These Pt (II) complexes, with the non-classical structures, demonstrated a significant anticancer activity against Jurkat and MCF-7 cancer cell lines. The results of ethidium bromide/acridine orange staining and Caspase-III activity suggest that these complexes were capable to stimulate an apoptotic mechanism of cell death in the cancer cells. Using different biophysical techniques and docking simulation analysis, we indicated that these complexes were also capable to interact efficiently with DNA via a non-intercalative mechanism. According to our results, substitution of cyclopentane (in complex 2) with two methyl groups (in complex 1) results in significant improvement of the complex ability to interact with DNA and subsequently to induce the anticancer activity. Overall, these binuclear Pt (II) complexes are promising group of the non-classical potential anticancer agents which can be considered as molecular templates in designing of highly efficient platinum anticancer drugs. Then, the focused nanomaterial was handled as a productive and encouraging nanocatalyst for the establishment of 2H-indazolo[2,1-b]phthalazine-1,6,11(13H)-trione attendants through one-pot, multi-component contraction of aldehydes, phthalhydrazide, and dimedone under solventless status and the generality and practical tolerance of this useful and environmentally benign method are illustrated. The simple work-up, smooth reaction conditions, good to excellent yields, and reasonably short reaction times are the particular benefits of this protocol. Besides, the new nano-powdered ionic liquid was a successful and modest catalyst that could definitely be recycled and reused several times without apparent decrease in its catalytic activity. As planned, the designed uriedo-anilinophthalazines 7a-i showed superior binding affinity than their anilinophthalazine parents (IC50 = 0.083-0.473 mu M). In particular, compounds 7g-i showed IC50 of 0.086, 0.083 and 0.086 mu M, respectively, which are better than that of the reference drug sorafenib (IC50 = 0.09 mu M). (C) 2016 Elsevier Masson SAS.
