Phosphotungstic acid supported on SiO2@NHPhNH2 functionalized nanoparticles of MnFe2O4 as a recyclable catalyst for the preparation of tetrahydrobenzo[b]pyran and indazolo[2,1-b]phthalazine-triones

MnFe2O4@SiO2@NHPhNH2-phosphotungstic acid was synthesized by the reaction of diamine-modified silica coated manganese ferrite nanoparticles with H3PW12O40, the Keggin-type heteropoly acid. This is the first time that H3PW12O40 supported on diamine functionalized MnFe2O4 nanoparticles has been reported. The synthesized catalyst was characterized by various techniques, including Fourier Transform Infrared Spectroscopy (FTIR), Transmission electron-microscopy (TEM), X-ray Powder Diffraction (XRD), Scanning Electron Microscope (SEM), Energy Dispersive X-ray Spectroscopy (EDX), Inductively Coupled Plasma Atomic Emission Spectroscopy (ICP-AES), Thermogravimetric Analysis (TGA), and Vibrating Sample Magnetometer (VSM). The synthesized nanoparticle was examined as the heterogenous nanocatalyst for the synthesis of tetrahydrobenzo[b]-pyran derivatives and 2H-indazolo-[2,1-b]-phthalazine-triones under the solvent-free heating or ultrasonic irradiation conditions. This method offers several advantages including high yield, low reaction times, and the simple work up procedure due to the presence of the magnetic nanoparticles in this catalyst, so that, it can be easily recovered and reused for at least six successive times without the loss of its high catalytic activity, suggesting its great Potential for industrial applications. Synthesis of New Derivatives of 1,2,3-Triazole-Linked Phthalazine-1,4-dionein Water: Experimental Aspects and Molecular Docking Calculations

A new series of phthalazine-based 1,2,3-triazole derivatives are synthesized by the reaction of 2-methyl-3-(prop-2-yn-1-yl)-2,3dihydro- phthalazine-1,4-dione or 2,3-bis(prop-2-yn-1-yl)-2,3-dihydrophthalaz- ine-1,4-dione with aromatic azides via coppercatalyzed azide-alkyne cycloaddition reactions in the presence of metformine as a ligand. These reaction procedures have the advantages of high-to-excellent yields, short reaction times, mild experimental conditions, and operational simplicity. All the synthesized compounds are screened in vitro for their antibacterial activities against the three bacterial strains Micrococcus luteus, Pseudomonas aeruginosa, and Bacillus subtilis. Furthermore, the results of anti-bacterial activity of the synthesized compounds was investigated using molecular docking calculations.

Nanoscale liposomal formulation of a SYK P-site inhibitor against B-precursor leukemia

We report preclinical proof of principle for effective treatment of B-precursor acute lymphoblastic leukemia (ALL) by targeting the spleen tyrosine kinase (SYK)-dependent antiapoptotic blast cell survival machinery with a unique nanoscale pharmaceutical composition. This nanoscale liposomal formulation (NLF) contains the pentapeptide mimic 1,4-Bis (9-O dihydroquinidinyl) phthalazine/hydroquinidine 1,4-phathalazinediyl diether (C61) as the first and only selective inhibitor of the substrate binding P-site of SYK. The C61 NLF exhibited a very favorable pharmacokinetic and safety profile in mice, induced apoptosis in primary B-precursor ALL blast cells taken directly from patients as well as in vivo clonogenic ALL xenograft cells, destroyed the in vivo clonogenic fraction of ALL blast cells, and, at nontoxic dose levels, exhibited potent in vivo antileukemic activity against patient-derived ALL cells in xenograft models of aggressive B-precursor ALL. Our findings establish SYK as an attractive molecular target for therapy of B-precursor ALL. Further development of the C61 NLF may provide the foundation for therapeutic innovation against therapy-refractory B-precursor ALL.

Synthesis and evaluation of anti-proliferative activity of 1,4-disubstituted phthalazines

A series of new phthalazine derivatives 3a-i and 4a-c were synthesized via the reaction of 1-chlorophthalazine derivative 2 with either N-substituted-piperazines, primary or their secondary amines. The structure of the synthesized, new compounds were characterized by spectral data. The anti-proliferative activity on human breast cancer cell line MCF-7 of the synthesized compounds was determined. The results showed that six of the test compounds (3a, 3g, 3i, and 4a-c) displayed potent cytotoxic activity ranging from 1.4 to 2.3 mu mol.

Synthesis and characterization of Ni(II)-Vanillin-Schiff base-MCM-41 composite as an efficient and reusable nanocatalyst for multicomponent reactions

An efficient heterogeneous catalyst (Ni(II)-Vanillin-Schiff base-MCM-41) has been synthesized by immobilization of nickel(II)-vanillin complex on MCM-41 nanostructure and characterized by XRD, TGA, BET, EDS, SEM, ICP-OES, TEM and FT-IR spectroscopy. The catalytic activity and stability of the prepared compound has been studied in some multicomponent reactions with polyhydroquinoline, 1H-indazolo [1,2-b] phthalazine-trione and 2,3-dihydroquinazolin-4(1H)-one derivatives under mild condition reactions. All the reactions were carried out in the presence of water as the solvent and under solvent-free conditions. The simple work up, mild conditions, excellent yield, inexpensive and non-toxic catalyst; makes this protocol both attractive and economically viable. More importantly, this catalyst can be reused several times without any significant loss of its catalytic activity.

Preparation, Characterization, and First Catalytic Application of a Novel Phosphotungstic Acid-Containing Ionic Liquid Immobilized on CuFe2O4@SiO2 Magnetic Nanoparticles in the Synthesis of 1H-Pyrazolo[1,2-b]phthalazine-5,10-diones

A novel heterogeneous acidic ionic liquid based on functionalized imidazolium salt of phosphotungstic acid (H3PW12O40, denoted as PW), immobilized on CuFe2O4@SiO2 magnetic nanoparticles, denoted as CuFe2O4@SiO2@C-3-Imid-C4SO3-PW, was prepared and characterized using FT-IR, SEM, EDX, and VSM techniques. High activity of the prepared material as a novel catalyst was evaluated in one-pot synthesis of 1H-pyrazolo[1,2-b]phthalazine-5,10-diones by reaction of phthalhydrazide with an aromatic aldehyde and malononitrile under solvent-free conditions. The process gave high yields of the products over short reaction time. The catalyst was efficiently recovered by magnetic decantation and used repeatedly without significant loss of its activity.

Synthesis, biological evaluation and in silico studies of novel N-substituted phthalazine sulfonamide compounds as potent carbonic anhydrase and acetylcholinesterase inhibitors

The synthesis, characterization and biological evaluation of a series of novel N-substituted phthalazine sulfonamide (5a-1) are disclosed. Phthalazines which are nitrogen-containing heterocyclic compounds are biologically preferential scaffolds, endowed with versatile pharmacological activity, such as anti-inflammatory, cardiotonic vasorelaxant, anticonvulsant, antihypertensive, antibacterial, anti-cancer action. The compounds were investigated for the inhibition against the cytosolic hCA I, II and AChE. Most screened sulfonamides showed high potency in inhibiting hCA II, widely involved in glaucoma, epilepsy, edema, and other pathologies (K(i)s in the ranging from 6.32 +/- 0.06 to 128.93 +/- 23.11 nM). hCA I was inhibited with K(i)s in the range of 6.80 +/- 0.10-85.91 +/- 7.57 nM, whereas AChE in the range of 60.79 +/- 3.51-249.55 +/- 7.89 nM. ADME prediction study of the designed N-substituted phthalazine sulfonamides showed that they are not only with carbonic anhydrase and acetylcholinesterase inhibitory activities but also with appropriate pharmacokinetic, physicochemical parameters and drug-likeness properties. Also, in silico docking studies were investigated the binding modes of selected compounds, to hCA I, II, and AChE.
