Synthesis and characterization of diazine-ring containing hydrazones and their Zn(II) complexes

Two new zinc(II) coordination compounds have been synthesized by the reaction of diazine-ring containing Schiff bases di(2-pyridyl) ketone phthalazine-1-hydrazone (HzDPK) and di(2-pyridyl) ketone 3-chloropyridazine-6-hydrazone (HpDPK) with zinc(II) salts in acetonitrile in the presence of triethylamine. The crystal and molecular structures of the complexes and that of the ligand HpDPK were determined by single-crystal X-ray structure analysis. In both complexes, zinc atoms are situated in distorted octahedral environments, formed by two meridionally coordinated NNN tridentate, mono-deprotonated ligands. Since the applicability of the coordination compounds depends on their thermal properties, the thermal decomposition of the ligands and their complexes was followed by simultaneous TG-DSC measurements. The desolvation process of the complexes is rather slow as a consequence of a restricted diffusion through the lattice and finishes similar to 200 A degrees C. The desolvated compounds are stable up to 340 A degrees C. In order to follow the solvent evaporation and to have a better insight into the decomposition mechanism of the compounds coupled TG-MS measurements were carried out.

In this paper, C-N bond formation between 2,3-dihydrophthalazine-1(4H),4-dione (phthalhydrazide) and alpha,beta-unsaturated esters was investigated and a new series of phthalazine derivatives was synthesized using an efficient and simple method under solvent-free conditions. An aza-Michael addition of phthalhydrazide to both acrylic and fumaric esters led to N-monosubstituted phthalhydrazides (as mono-Michael adduct) in the presence of tetrabutylammonium bromide as a high polar media, and 1,4-diaza-bicyclo[2,2,2] octane as an available organic base. In this reaction, the N1,N2-bis-Michael adduct was not observed at all. Also, reactions were performed at 90 degrees C and yields of products were good to excellent.

The structure of the most active catalyst, C@TiO2-SO3H-IL1, before and after the coating with the ionic liquid was investigated by FTIR, TGA, XRD, CHNS, EDX, SEM and HRTEM. The catalyst allows easy recovery and excellent reusability for up to five catalytic runs without considerable loss of activity.

Diiminoisoindoline (DII) is a crucial reagent for the synthesis of phthalocyanine as well as related macrocycles and chelates such as hemiporphyrazine and bis(iminopyridyl)isoindoline. In view of the anticonvulsant activity reported for phthalazine derivatives as non-competitive AMPA receptor antagonists, a new series of phthalazine-1,4-diones (2-12) were designed and synthesized. The neurotoxicity was assessed using rotarod test. The molecular docking was performed for the synthesized compounds to assess their binding affinities toward AMPA receptor as non-competitive antagonists. The molecular modeling data were strongly interrelated to biological screening data. Compounds 8, 7(b), 7(a), 10 and 3(a) exhibited the highest binding affinities as non-competitive AMPA receptor antagonists and also showed the highest relative potencies of 1.78, 1.66, 1.60, 1.59 and 1.29, respectively, as anticonvulsants in comparison with diazepam. The most active compounds 8, 7(b), 7(a), 10 and 3(a) were further tested against maximal electroshock seizure (MES). Compounds 8 and 7(b) and 3(a) showed 100% protection at a dose level of 125 mu gm/kg, while compounds 7(a) and 10 exhibited 83.33% protection at the same dose level. These agents exerted low neurotoxicity and high safety margin in comparison with valproate as a reference drug. Most of our designed compounds exhibited good ADMET profile.
