Synthesis, characterization and crystal structures of Ni(II) and Cu(I) complexes with the condensation product of 2-(diphenylphosphino) benzaldehyde and 1-hydrazinophthalazine

A square-pyramidal Ni(II) complex (1) with the composition [NiL(HL’)]CIO4 center dot H2O was synthesized in the reaction of nickel(II) perchlorate and 2[2-(diphenylphosphino)benzylidene]-1-phthalazinylhydrazone (HL). The coordination surrounding the Ni(II) ion consist of a PNN coordinated deprotonated HL ligand and its corresponding oxidation product phosphine oxide HL’ coordinated via two nitrogen atoms of the hydrazinophthalazine moiety. The structure of Ni(II) complex 1 was determined by X-ray analysis, IR spectroscopy, elemental analysis and magnetic measurements. In the reaction of copper(II) perchlorate with HL, a binuclear phthalazine bridged Cu(I) complex (2) was obtained with the composition [Cu-2(HL)(2)](ClO4)(2)center dot C2H5OH. Three complexes containing 2-pyrazinecarboxylate (pzca(-)), including [Ni(pzca)(2)(H2O)(2)], [Co(pzca)(2)(H2O)(2)], and [Cu(pzca)(2)(H2O)(2)], have been synthesized and characterized using physicochemical and spectroscopic methods. Furthermore, the structure of each complex was determined by single-crystal X-ray diffraction. All three complexes have an octahedral geometry, where the metal ion chelated by two carboxylate oxygens, two nitrogen atoms belonging to pyrazinic acid molecules, and two oxygen atoms of two water molecules. The catalytic activities of these complexes were also investigated in the green synthesis of 2H-indazolo[2,1-b]phthalazine-triones by the reaction of hydrazine hydrate with an arylaldehyde, phthalic anhydride, and dimedone in acetic acid. A simple and highly efficient procedure has been described for the synthesis of 3-(3-aryl[1,8]naphthyridin-2-yl)-7-[1-[1,4-dioxo-1, 2, 3, 4-tetrahydro-6-phthalazinyl]-2, 2, 2-trifluoro-1-(trifluoromethyl)ethyl]-2-(3-aryl[1,8]naphthyridin-2-yl)-1,2,3,4-tetrahydro-1,4-phthalazinediones 3 by the condensation of 3-aryl-2-hydrazino-1,8-naphthyridines 1 with 4,4 ‘-(hexafluoroisopropylidene)diphthalic anhydride 2 in 2:1 molar ratio in the presence of catalytic amount of DMF under microwave irradiation/ in presence of catalytic amount of PTSA in the solid state at RT under grinding conditions. The products have been obtained in very good yields and in a state of high purity. The structures of the compounds 3 have been established on the basis of their spectral (IR and H-1 NMR) and analytical data. The compounds 3 have been screened for their antibacterial activity.

Synthesis of 1H-pyrazolo[1,2-b]phthalazine-5,10-dione derivatives: assessment of their antimicrobial, antituberculosis and antioxidant activity

A new series of pyrazolo[1,2-b]phthalazine derivatives (4a-p) bearing the 5-aryloxypyrazole nucleus was synthesized by one-pot, three-component, base-catalyzed cyclo condensation reaction of 3-methyl-5-aryloxy-1-aryl-1H-pyrazole-4-carbaldehyde (1a-d), malononitrile or ethyl cyanoacetate (2a-b) and 2,3-dihydro-1,4-phthalazinedione (3a-b) in ethanol containing an eco friendly base, NaOH, in good to excellent yields. All synthesized compounds (4a-p) were duly characterized by physico-chemical parameters, H-1 NMR, C-13 NMR, FT-IR and LCMS techniques. In vitro antimicrobial activity of the synthesized compounds was investigated against a representative panel of pathogenic strains. Compounds 4e, 4g, 4h, 4k and 4o exhibited excellent antimicrobial activity compared with first line drugs. In vitro antituberculosis activity was evaluated against Mycobacterium tuberculosis H37Rv, and compounds 4g and 4o emerged as the promising antimicrobial members with better antituberculosis activity. A brine shrimp bioassay was carried out to study the in vitro cytotoxic properties of the synthesized compounds. In vitro antioxidant activity was evaluated by the ferric-reducing antioxidant power method. Compounds 4c, 4d, 4g and 4h showed the highest antioxidant potencies.

Design, synthesis, structure-activity relationship and mechanism of action studies of a series of 4-chloro-1-phthalazinyl hydrazones as a potent agent against Leishmania braziliensis

With the aim to identify a potential drug candidate to treat cutaneous leishmaniasis, a series of 1-phthalazinyl hydrazones were synthesized and tested against Leishmania braziliensis parasite, one of the main responsible of this disease in the world. A structure-activity relationship permitted to identify two phthalazines containing nitroheterocyclic moiety 31 and 3m as promising new lead compounds. These compounds showed a significant antileishmanial activity against promastigote form of L. braziliensis, with EC50 values in sub-micromolar and nanomolar ranges. The phthalazine 31 also displayed a selective and excellent activity against the clinically relevant intracellular amastigotes form, with a EC50 value in sub-micromolar range (0.59 mu M), without affecting the viability of the host cells. Oxidative stress was identified as the possible mode of action of the most active phthalazine. Considering their significant antileishmanial activity and ease synthesis, the phthalazine containing nitroheterocyclic represents a promising agent against Leishmania braziliensis for the rational design of new leads. Silver(I) complexes with quinazoline and phthalazine: synthesis, structural characterization and evaluation of biological activities

New silver.I) complexes with quinazoline (qz) and phthalazine (phtz), [Ag(NO3)(qz)](n) (1) and {[Ag(CH3CN)](2)(mu-phtz)(2)}[BF4](2) (2), have been synthesized and structurally characterized by using different spectroscopic and single-crystal X-ray diffraction techniques. The obtained results revealed that the reaction of AgNO3 with qz at room temperature in a 2 : 1 molar ratio led to the formation of the polynuclear complex 1. However, the reaction of AgBF4 with phtz under the same experimental conditions resulted in the formation of the dinuclear complex 2. The solution behaviour and air/light stability of these silver.I) complexes have been investigated. The complexes 1 and 2, along with the silver.I) salts used for their synthesis, were evaluated by in vitro antimicrobial studies against a panel of microbial strains that lead to many skin and soft tissue, respiratory, wound, and nosocomial infections. The obtained results indicate that all tested silver(I) compounds have good antibacterial activity with MIC values in the range from 1.5 to 15.6 mu g mL(-1) against the investigated strains. On the other hand, their antifungal activity against Candida albicans was moderate. In order to determine the therapeutic potential of 1 and 2, their antiproliferative effect on the normal human lung fibroblast cell line MRC5, hemolytic effect on red blood cells and embryotoxicity on zebrafish (Danio rerio) have also been evaluated.

Waste to wealth: conversion of nano-magnetic eggshell (Fe3O4@Eggshell) to Fe3O4@Ca(HSO4)(2): cheap, green and environment-friendly solid acid catalyst

Eggshell is a hazardous waste by European Union regulations, so that discarded thousands of tons per year. To convert waste (eggshell) to wealth (catalyst), nano-magnetic eggshell was prepared based on the nano-Fe3O4, and then the eggshell was converted to Ca(HSO4)(2) with organic acid, namely, chlorosulfonic acid. Based on the back titration, 5.18mmol SO4H group was loaded per gram of the nano-structure. Using this method eggshell was converted to cheap, green and environment-friendly solid acid catalyst. The prepared catalyst (nano- Fe3O4@Ca(HSO4)(2)) was characterized by Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), energy dispersive X-ray spectroscopy (EDX), field emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), vibrating sample magnetometer (VSM), and thermal gravimetric analysis (TGA). The activity of eggshell waste-derived catalysts was successfully evaluated in the synthesis of value-added products, namely indazolo[1,2-b]-phthalazinetrione derivatives as a benchmark multicomponent reaction. In addition, design of experiments shows that increase in amount of catalyst (and temperature), boost the reaction yield, especially with steeper slope at higher temperature.

Montmorillonite K-10 Supported 11-molybdo-1-vanadophosphoric Acid (H4PMo11V1O40/K-10) Catalysts for Environmentally Benign Synthesis of 2H-indazolo[2,1-b]phthalazine-triones Under Solvent-free Condition

A series of 11-molybdo-1-vanadophosphoric acid supported on montmorillonite K-10 catalysts were prepared and characterized by FT-IR spectroscopy, thermal analysis, XRD, BET and SEM analysis techniques. Characterization data reveals the chemical immobilization of H4PMo11V1O40 catalyst on the montmorillonite K-10 support. The catalytic performance of synthesized catalysts was investigated for the synthesis of 2H-indazolo[2,1-b]phthalazine-1,6,11(13H)-trione derivatives by one-pot three-component reaction of phthalhydrazide, dimedone and aromatic aldehydes under solvent-free conditions. Among different catalysts, 20% H4PMo11V1O40 supported on to montmorillonite K-10 showed the highest catalytic activity. Effect of reaction parameters such as catalyst loading, temperature and the nature of substituents on the aromatic ring of aldehydes were also evaluated. The protocol developed using H4PMo11V1O40/K-10 has several distinct advantages such as operational simplicity, short reaction time, high yield, reusable catalyst and preclusion of toxic solvent.
