We describe here our investigation of the asymmetric Diels-Alder reaction of chiral 3-(acyloxy)acryloyl oxazolidinones as dienophiles in various Lewis-acid promoted reactions with cyclopentadiene. The resulting highly functionalized cycloadducts are useful intermediates for the synthesis, particularly for the optically active synthesis of 6-5-5 tricyclic hexahydro-4H-3,5-methanofuro[2,3-b]pyranol (3) with five contiguous chiral centers. This stereochemically defined crown-like heterocyclic derivative is an important high affinity ligand for a variety of highly potent HIV-1 protease inhibitors. Among the various dienophiles and Lewis acid-mediated reactions surveyed, 3-(4-methoxybenzoyl)acryloyl oxazolidinone as the dienophile and diethylaluminum chloride as the Lewis-acid provided the desired endo product with excellent diastereoselectivity. The cycloaddition was carried out in multi-gram scale and the cycloadduct was efficiently converted to alcohol 3 with high enantiomeric purity. The optically active ligand was then transformed into potent HIV-1 protease inhibitor 2.

The development of sub-nanometric metal particles (<1 nm) as advanced heterogeneous catalysts has received considerable interest due to their outstanding catalytic performance, while the synthesis and stabilization of sub-nanometric catalysts (SNCs) without using additional surface capping agents remains a challenge. Herein, we report the synthesis of novel three-dimensional pyrenyl graphdiyne (Pyr-GDY) ultrafine nanofibers (3-10 nm), which can serve as an ideal substrate for electroless deposition and stabilization of Pd SNCs through the terminal uncoupled acetenyl groups in Pyr-GDY, with an average Pd particle size of only 0.83 nm. The as-synthesized Pd/Pyr-GDY composite shows extremely high catalytic activities for the reduction of nitroarenes to arylamines and Suzuki coupling reactions, 300 and 25 times higher than those of commercial Pd/C, respectively. The outstanding catalytic performance can be ascribed to the sub-nanometric Pd particles with a clean surface, and the unique three-dimensional network structure of Pyr-GDY, being favorable for rapid mass transfer. Our result provides an ideal carbon material for electroless deposition and stabilization of other SNCs with a clean surface, which will display outstanding catalytic activity for various catalytic reactions. A site-selective supported palladium nanoparticle catalyzed Suzuki-Miyaura cross-coupling reaction with heteroaryl esters and arylboronic acids as coupling partners was developed. This methodology provides a heterogeneous catalytic route for aryl ketone formation via C(acyl)-O bond activation of esters by successful suppression of the undesired decarbonylation phenomenon. The catalyst can be reused and shows high activity after eight cycles. The XPS analysis of the catalyst before and after the reaction suggested that the reaction might be performed via a Pd-0/Pd-II catalytic cycle that began with Pd-0.

Eco-friendly carbocatalyst is extremely desirable in organic catalysis and wastewater treatment. Herein, we reported a one-pot synchronous hydrothermal process to synthesize a highly efficient N doped holey graphene (NHG) carbocatalyst, in which graphene oxide nanosheets are self-assembled, reduced, chemically etched, and simultaneously doped with nitrogen to form a graphene hydrogel in the presence of H2O2 and NH4OH. Benefiting from the good hydrophilicity, unique hierarchical porous structure, large specific surface area (322.1 m(2)g(-1)), and high N content (9.77 at. %), the resultant NHG carbocatalyst exhibits an excellent catalytic performance for the hydrogenation of N-containing unsaturated compounds. In 4-nitrophenol reduction reaction, the NHG carbocatalyst delivers a turnover frequency of 3.32 x 10(-2) min(-1). This catalytic activity is superior to that of the commercial Pd/C (5.0 wt%), other previously reported carbocatalysts, and many noble-metal-based catalysts. Furthermore, the molecular structure analysis combined with density functional theory calculations elucidate metal-free catalytic mechanism of hydrogenation reaction for unsaturated chromophore groups in N-containing organic dyes. A new series of Pyrazolo[3,4-d]pyrimidine containing amide derivatives (8 a-l) were designed, synthesized, and evaluated for their in vitro alpha-amylase inhibitory activity. The IC50 values of the target compounds ranged from 1.60 +/- 0.48 to 2.04 +/- 1.20 mu M as compared to the standard acarbose 1.73 +/- 0.05 mu M. All the Pyrazolo[3,4-d]pyrimidine amide derivatives displayed good inhibitory activities, while seven analogs (8 d, 8 f, 8 g, 8 h, 8 i, 8 j and 8 k) exhibited more or less equipotent activity with IC50 values 1.77 +/- 2.84, 1.65 +/- 0.45, 1.66 +/- 2.24, 1.73 +/- 0.37, 1.60 +/- 0.48, 1.75 +/- 0.36 and 1.64 +/- 0.03 mu M respectively. Further, the most potent alpha-amylase inhibitors 8 d and 8 k were also screened for their in vivo antidiabetic activity against alloxan induced diabetic rat model at the dose of 25 and 50 mg/kg. Oral administration of these tested compounds significantly reduced the fasting blood glucose levels in dose dependent manner. The hypoglycemic effects of these compounds were more evident at 3 h and 5 h after administration of tested compounds which was similar to the effect displayed by the positive control. In addition, the binding energies calculated from the docking studies with the alpha-amylase enzyme (PDB ID: 1HNY) and biological activities indicate that the compounds containing nitro moiety on the phenyl group contributed significantly towards the antidiabetic activity.

In the present study, a series of substituted 1,3,4-thiadiazole derivatives 4(a-o), 5(a-m) and 6(a-j) were synthesized and characterized by IR, H-1 NMR, C-13 NMR and mass spectroscopic technique. The synthesized compounds were evaluated for their in vitro anti-mycobacterial activity against the Mycobacterium tuberculosis H37Rv and resistance MDR-TB strain. Among the compounds tested N-(5-(4-nitrophenyl)-1,3,4-thiadiazol-2-yl)furan-2-carboxamide (4h) showed significant inhibitory activity with MIC of 9.87 mu M (H37Rv strain) and 9.87 mu M (MDR-TB strain) compared to isoniazide (MIC of 3.64 mu M (H37Rv) and >200 mu M (MDR-TB strain)] and rifampin [MIC of 0.152 mu M (H37Rv) and 128 mu M (MDR-TB strain)]. In addition, these compounds have also been assessed for their cyto-toxicity to a mammalian Vero cell line using the MTT assay. A new synthetic application of N-sulfonyl-1,2,3-triazoles acting as a 1-aza-[4C] synthon via the 1,2-shift reaction of an alpha-imine rhodium carbene was developed for the synthesis of fused pyrimidine derivatives. The high reactivity of the strained three-membered 2H-azirine ring facilitated the unusual cyclization of electron-deficient dienes with electron-deficient dienophiles. The compatibility was good with common functionalities tolerated. Excellent chemoselectivity was observed, and no reactions occurred between the rhodium carbene and 2H-azirine. The products could be converted into seven-membered multi-functionalized 1H-1,4-diazepine derivatives, illustrating the potential application of the protocol in medium-sized N-heterocycle synthesis.
