Despite numerous advances in spectroscopic methods through the latter part of the 20th century, the unequivocal structure determination of natural products can remain challenging, and inevitably, incorrect structures appear in the literature. Computational methods that allow the accurate prediction of NMR chemical shifts have emerged as a powerful addition to the toolbox of methods available for the structure determination of small organic molecules. Herein, we report the structure determination of a small, stereochemically rich natural product from Laurencia majuscula using the powerful combination of computational methods and total synthesis, along with the structure confirmation of notoryne, using the same approach. Additionally, we synthesized three further diastereomers of the L. majuscula enyne and have demonstrated that computations are able to distinguish each of the four synthetic diastereomers from the 32 possible diastereomers of the natural product. Key to the success of this work is to analyze the computational data to provide the greatest distinction between each diastereomer, by identifying chemical shifts that are most sensitive to changes in relative stereochemistry. The success of the computational methods in the structure determination of stereochemically rich, flexible organic molecules will allow all involved in structure determination to use these methods with confidence.

A chemoenzymatic synthesis of the title compound has been developed using an efficient and highly enantioselective lipase-catalyzed acylation in a hydrophobic ionic liquid, [bmim][PF6], followed by a diastereoselective asymmetric dihydroxylation as the key steps for incorporating the stereogenic centers. The further conversion to the appropriate intermediates and subsequent acylation with lauric acid furnished the target compound.

Design of chemically novel, biologically potent small heterocyclic molecules with anticancer activities, which targets the enzyme heparanase has gained prominent clinical interest. We have synthesized a novel class of carboxamide derivatives by coupling various substituted aromatic acid hydrazides and triazoleamine with pyrrolidine carboxylic acid by using coupling agents. The synthesized compounds are characterized by spectroscopic techniques such as FT-IR, DIMS and NMR. These compounds arc investigated for cytotoxicity on different cancer cell lines and heparanase inhibitory activity. Most of them showed moderate heparanase inhibitory activity and good cytotoxicity.

The effect of topology on the structure, self-assembly, and selective Fe3+ binding of delta-peptides has been investigated. A series of delta-peptides with an amino acid containing dihydropyrimidinone and o-, m-, and p-aminobenzoic acids have been designed to study the structure-function relationship. A new amino acid containing dihydropyrimidinone was synthesized by the Biginelli reaction of ethyl acetoacetate, urea, and o-nitrobenzaldehyde followed by reduction with iron powder and acetic acid. X-ray crystallography sheds some light on the conformations, self-assembly, and the diverse degrees of pi-pi stacking of adjacent delta-peptide molecules. Peptides with o- or m-aminobenzoic acid form eight-membered intramolecular hydrogen-bonded turn conformations and self-assemble through intermolecular hydrogen bonds between dihydropyrimidinone units to form a butterfly-like structure. However, the delta-peptide containing p-aminobenzoic acid forms a water-mediated cage-like structure. Irrespective of the presence of the same functional groups, only the delta-peptide with o-aminobenzoic acid can selectively bind Fe3+ in methanol as well as in water. The topology plays a crucial role in the selective Fe3+ ion binding by the delta-peptide.

An asymmetric [3 + 2] formal cycloaddition reaction between oxindole alpha-keto esters and enals has been developed. This alpha,gamma-regioselective reaction afforded multifunctional carbocyclic oxindoles via dienamine-enamine cascade reaction followed by isomerization. Furthermore, a highly enantioselective Diels-Alder reaction was demonstrated between 2,4-dienals and oxindole alpha-keto esters via trienamine intermediates. Chiral secondary amine catalyzed the both reactions under mild conditions and the cyclized products were isolated in moderate to good yields with good diastereoselectivities (>20:1 dr) and enantioselectivities (up to 99 %).

Carbohydrate-based low-molecular-weight gelators are useful and versatile compounds for the preparation of soft materials. Using N-acetyl-D-glucosamine as the starting material, we synthesized and characterized 15 glycolipids containing an amide with different ester functional groups. These include aliphatic derivatives with varying chain lengths and aromatic derivatives. Most of the hybrid amide-esters have molecular weights less than 500 D. These glycolipids were found to be effective gelators for several organic solvents, water, and aqueous solutions. Two efficient hydrogelators were also obtained at low concentrations. A few representative gels were characterized using optical microscopy, atomic force microscopy, and rheology to obtain information on their morphology and gel stability. Three gelators were also used to encapsulate naproxen sodium and toluidine blue. The sustained release of the drug from the gel to the aqueous phase was monitored by UV-vis spectroscopy. These gelators have structural flexibility that can be stimuli responsive. The esters can be hydrolyzed and several gels were converted to solutions under basic conditions. These rationally designed gelators could be utilized as stimuli-responsive smart materials with controlled release properties.

A simple, efficient, sustainable and economical method for the oxidation of alcohols and amines has been developed based on chloride, a sea abundant anionic catalyst for the practical synthesis of a wide range of carboxylic acids, ketones and imines. Oxidation of aromatic alcohols was carried out using NaCl (20 mol%) as the catalyst, NaOH (50 mol%) and aq. TBHP (4 equiv.) as the oxidant in 55-92% isolated yields. Oxidation of aromatic amines to imines was achieved by using only 20 mol% of NaCl and aq. TBHP (4 equiv.) in 32-93% isolated yields. The chlorine species formed during the reaction as the active oxidation catalyst has been identified as ClO2- for alcohols and ClO-/ClO2- for amines by control experiments. This method is mostly free from chromatographic purification, which makes it suitable for large-scale synthesis. We have scaled up to 30 gram scale the synthesis of carboxylic acids and imines in good yields and have also carried out efficiently this new method using filtered sea water as the solvent and catalyst.

Assessment of ocular irritation risk is an international regulatory requirement in the safety evaluation of products. In response to this need, L’Oreal developed the SkinEthie Human Corneal Epithelium (HCE) Eye Irritation Test (Err) that has been included in OECD Test Guideline 492. SkinEthic (TM) HCE EIT is able to correctly and reliably identify chemicals not requiring classification versus labelling for eye irritation or serious eye damage according to UN GHS. In an effort to promote its global use, the performance of the method was evaluated after long-distance shipment and compared to European shipment conditions. Results obtained by Cosmos Technical Center (Japan) after extended tissues transit were compared to results obtained in L’Oreal (France). Thirty-nine out of 40 blinded chemicals, representing different functional chemical classes, were consistently classified in both laboratories. The SkinEthic (TM) HCE EIT test method was also evaluated for its performance after extended storage of the tissues. The performance was in agreement with the values reported in OECD TG 492, with an overall accuracy of 87.1% (based on 119 chemicals), sensitivity of 95.5% and specificity of 73.5%. The reliability and relevance of SkinEthic (TM) HCE Err test method after long-distance shipment and extended storage remain in agreement with regulatory validation criteria.
