Herein, a new iodide-mediated three-component annulation reaction of secondary anilines, cyclohexanones, and elemental sulfur is demonstrated, which allows access to various phenothiazines with the merits of formation of multiple chemical bonds in one single operation, high step and atom efficiency, readily available feedstocks and catalyst system, and good substrate and functional group compatibility. The developed chemistry capable of constructing novel phenothiazines with structural diversity offers a significant basis for further applications.

Lignin is a biodegradable, abundant and renewable natural polymer on earth. In this study, deep eutectic solvents (DESs)-modified lignin heterogeneous catalysts were prepared for the first time via a facile acid-base interaction between choline chloride (ChCl) and p-aminobenzoic acid (PABA) or its derivatives, which were used to efficiently catalyze the cycloaddition of CO2 and terminal epoxides under green and mild conditions. Among them, lignin-ChCl-PABA was found to exhibit the highest catalytic activity for the cycloaddition of epoxides in the presence of 1.0 MPa of CO2 while retaining high selectivity, which far outperformed those achieved on the homogeneous EDSs catalytic system. A series of cyclic carbonates could be obtained in high yields (90-99%), and lignin-ChCl-PABA was stable and reusable for the cycloaddition reaction (84% yield after 5 recycles). The attaching DES of ChCl/PABA onto the functional lignin increased the catalytic activity and also made the catalyst easier to recycle from the reaction mixture. The -OH and -NH2 groups on the surface of lignin-ChCl-PABA played a very important role in the acceleration of the cycloaddition reaction under solvent free conditions.

The present study reports the convenient synthesis, spectroscopic characterization, bio-assays and computational evaluation of a novel series of N-acyl-1H-imidazole-1-carbothioamides. The screened derivatives displayed excellent antioxidant activity, moderate antibacterial and antifungal potential. The screened derivatives were found to be highly biocompatible against hRBCs. Molecular docking ascertained the mechanism and mode of action towards the molecular target delineating that ligands and complexes were stabilized at the active site by electrostatic and hydrophobic forces in accordance to the corresponding experimental results. Docking simulation provided additional information about the possibilities of inhibitory potential of the compounds against RNA. Computational evaluation predicted that N-acyl-1H-imidazole-1-carbothioamides 5c and 5g can serve as potential surrogates for hit to lead generation and design of novel antioxidant and antibacterial agents.

The present report describes an organocatalytic cascade reaction between 2-alkylidene benzo[b]thiophenone derivatives and enones in the presence of the Cinchona alkaloid amine. Spirobenzothiophenonic cyclohexane derivatives containing three stereocenters were prepared via one-step synthesis in yields ranging from 88 to 96% and in enantioselectivities (enantiomeric excess (ee)) ranging from 85 to 97%, with diastereoselectivities of approximately 14/2/1. Therefore, this method provides an efficient route for the synthesis of a new class of optically active 2-spirobenzothiophenones.

A series of 1,2,4-triazole derivatives 1-14 was synthesized to investigate their neuroprotective effects and mechanisms of action. Compounds 5-11 noticeably protected PC12 cells from the cytotoxicity of H2O2 or sodium nitroprusside (SNP). Compound 11 was the most effective derivative. Compound 11 chelated Fe (II) iron, scavenged reactive oxygen species (ROS), and restored the mitochondrial membrane potential (MMP). Moreover, it enhanced the activity of the antioxidant defense system by increasing the serum level of superoxide dismutase (SOD) and promoting the nuclear translocation of nuclear factor erythroid 2-related factor 2 (Nrf2). Compound 11 caused certain improvements in behavior, the cerebral infarction area, and serum levels of biochemical indicators (TNF-alpha, IL-10, SOD and MDA) in a rat MCAO model. The lethal dose (LD50) of compound 11 in mice receiving intraperitoneal injections was greater than 400 mg/kg. Meanwhile, pharmacokinetic experiments revealed high bioavailability of this compound after both oral and intravenous administration (F = 60.76%, CL = 0.014 mg/kg/h) and a longer half-life (4.26 and 5.11 h after oral and intravenous administration, respectively). A desirable alternative to organic solvents, very cheap, non-toxic and biodegradable deep eutectic solvent (DES) based on choline chloride and urea was introduced for 2,4,6-trichloro triazine mediated amide bond formation between carboxylic acids and amines at room temperature. Sub-stoichiometric amount of TCT was used as a cheap and readily available C-O activating reagent. Various derivatives of amines were coupled with carboxylic acids, via triacylated triazine intermediate, resulting the reaction of acids with TCT, and the corresponding amides were obtained in moderate to good yields.

With the infection rate of Bordetella pertussis at a 60-year high, there is an urgent need for new anti-pertussis vaccines. The lipopolysaccharide (LPS) of B. pertussis is an attractive antigen for vaccine development. With the presence of multiple rare sugars and unusual glycosyl linkages, the B. pertussis LPS is a highly challenging synthetic target. In this work, aided by molecular dynamics simulation and modeling, a pertussis-LPS-like pentasaccharide was chemically synthesized for the first time. The pentasaccharide was conjugated with a powerful carrier, bacteriophage Q beta, as a vaccine candidate. Immunization of mice with the conjugate induced robust anti-glycan IgG responses with IgG titers reaching several million enzyme-linked immunosorbent assay (ELISA) units. The antibodies generated were long lasting and boostable and could recognize multiple clinical strains of B. pertussis, highlighting the potential of Q beta-glycan as a new anti-pertussis vaccine.
