Piperine is a component of pepper which has earlier been reported as anticancer active compound. This work is emphasized on the design and synthesis of new hybrid piperine analogs by coupling piperine with the amine group of oxadiazoles and thiadiazoles. The new series of twelve piperine analogs was been tested for in vitro anti-proliferative activity using sulforhodamine B (SRB) assay test against MCF-7, PC-3, and HeLa cell lines. Among the twelve synthesised molecules piperine derivative with oxadiazole baring hydroxyl group (3) exhibits the higher activity against MCF-7 cell line than the reference drug Adriamycin and also displays the highest binding energy in the in silico studies. The other analogs are moderately active.

A visible-light-catalyzed radical N-acylation of amines with carboxylic acids was achieved by means of eosin Y as the photocatalyst under aqueous medium without any transition-metal-based catalytic systems. Previously we have reported that 25-OCH3-PPD could suppress the reproduction of cancer cells and cause apoptosis without obvious toxicity. Herein, we aimed to enhance its bioactivity by introducing aromatic groups to its dammarane-type skeleton. These synthesized derivatives were tested for their inhibitory activities against five cancer cell lines. Of them, compounds 3a, 14a and 18a had the strongest antiproliferative activities against tumor cells (IC50 < 15 mu M, 5-fold to 10-fold increases than 25-OCH3-PPD). Especially compound 14a displayed the most potent activity against DU145, MCF-7 and HepG2 cells (IC50 = 6.7 +/- 0.8, 4.3 +/- 0.8 and 5.8 +/- 0.6 mu M, respectively). Structure-activity relationships demonstrated that having aromatic ester at the C3 position could improve the bioactivity. Alkene carbooxygenation has attracted considerable attention over the past few decades as this approach provides an efficient access to various oxygen-containing molecules, especially the valuable O-heterocycles. However, examples of catalytic alkene carbooxygenation via a direct C-O cleavage are quite scarce, and the C-O cleavage in these cases is invariably initiated by transition metal-catalyzed oxidative addition. We report here a novel Bronsted acid-catalyzed intramolecular alkoxylation-initiated tandem sequence, which represents the first metal-free intramolecular alkoxylation/Claisen rearrangement. Significantly, an unprecedented Bronsted acid-catalyzed intramolecular alkene insertion into the C-O bond via a carbocation pathway was discovered. This method allows the stereocontrolled synthesis of valuable indole-fused bridged [4.2.1] lactones, providing ready access to biologically relevant scaffolds in a single synthetic step from an acyclic precursor. Moreover, such an asymmetric cascade cyclization has also been realized by employing a traceless chiral directing group. Control experiments favor the feasibility of a carbocation pathway for the process. In addition, biological tests showed that some of these newly synthesized indole-fused lactones exhibited their bioactivity as antitumor agents against different breast cancer cells, melanoma cells, and esophageal cancer cells.

Modifying the surface of metal catalyst is a crucial subject for improving the heterogeneous metal catalysts. It is still a great challenge to understand the structure-activity relationship (SAR) between the surface supporting groups and the metal particles. Herein, Rh NPs supported on the magnetic silica sphere with various functional groups (-NH2, -SH, -SO3H, -N=CH2, -Cl, -NHCOCH3 and -NHCH2Ph) have been prepared for catalytic hydrogenation of nitroarenes under atmospheric pressure at room temperature. We discover that the chemical state of Rh NPs depends on the supporting groups significantly. The electron-donating functionalities can effectively elevate the Rh-0/Rh3+ ratio, which increase the catalytic performance both in conversion and selectivity. The amino group decorated catalyst has such a good selectivity that no dechlorination reaction is observable in the hydrogenation of 2-chloro-3-nitropyridine. What’s more, by introducing magnetic Fe3O4 nuclei and mesoporous silica encapsulating layer, the catalyst can be recovered conveniently by an external magnet, and can be reused 10 cycles without any loss of activity.

New chiral regioisomeric gamma-hydroxyphosphine ligands were synthesized from alpha-pinene. The hydrogenation of nitroarenes is one of the most important strategies for the preparation of anilines. However, it is still a great challenge to develop mild and efficient synthetic routes toward aniline synthesis, particularly those employing both non-precious metal catalysts and low-pressure H-2. Herein, we report a highly efficient protocol for the selective hydrogenation of nitroarenes in neutral H2O using H-2(1 atm) over a heterogeneous Zn(0) catalyst under mild conditions. The nitro groups of an array of nitroarenes can be converted into -NH(2)with up to 99% conversions and a selectivity of >99%, even when functionalized with easily reducible substituents, or in the presence of aromatic ketones or styrene. This study might open an avenue for the selective hydrogenation of nitroarenes over a zinc catalyst using 1 atm H-2.

Conventional luminescent sensors relying on single emission responsive to analytes demonstrate limited sensitivity and selectivity and developing novel self-calibration probes is highly required. A series of water-stable coordination polymers (CPs) with three emitting centers for tunable emissions is reported. They are facilely synthesized by simultaneously doping red-emitting Eu3+ and green-emitting Tb3+ into the same framework. The versatile Eu0.67Tb0.33-CP is applied to conveniently distinguishing different nitroaromatics pollutants and multidimensional ratiometric sensing CrO42-/Cr2O72-. Specific nitroaromatics can cause unique changes to the ligand -> Eu3+/Tb3+ energy transfer efficiency, while CrO42-/Cr2O72- intriguingly quench the three emissions at different rates. Each nitroaromatic exhibits notably correlation to a unique combination of the two ligand -> Eu3+/Tb3+ emission intensity ratios, enabling a 2D fingerprint recognition. Furthermore, the concentration-dependent evolution of the two ratios upon exposure to CrO42-/Cr2O72- can be mapped out, achieving a 3D code recognition. As a result, the sensor demonstrates selective recognition of 4-nitrophenol and CrO42-/Cr2O72- with high sensitivity and selectivity as well as great recycling ability. Both the synthetic and sensing strategies present a promising strategy to develop facile and effective ratiometric sensors.
