Cyanobacterial pyruvate dehydrogenase multienzyme complex E1 (PDHc E1) is a potential target enzyme for finding inhibitors to control harmful cyanobacterial blooms. In this study, a series of novel triazole thiamin diphosphate (ThDP) analogs were designed and synthesized by modifying the substituent group of triazole ring and optimizing triazole-benzene linker as potential cyanobacterial PDHc E1 (Cy-PDHc E1) inhibitors. Their inhibitory activities against Cy-PDHc E1 in vitro and algicide activities in vivo were further examined. Most of these compounds exhibited prominent inhibitory activities against Cy-PDHc E1 (IC50 1.48-4.48 mu M) and good algicide activities against Synechocystis PCC6803 (EC50 0.84-2.44 mu M) and Microcystis aeruginosa FACHB905 (EC50 0.74-1.77 mu M). Especially, compound 8d showed not only the highest inhibitory activity against Cy-PDHc E1 (IC50 1.48 mu M), but also the most powerful inhibitory selectivity between Cy-PDHc E1 (inhibitory rate 98.90%) and porcine PDHc E1 (inhibitory rate only 9.54%). Furthermore, the potential interaction between compound 8d and Cy-PDHc E1 was analyzed by a molecular docking method and site-directed mutagenesis and enzymatic analysis and fluorescence spectral analysis. These results indicated that compound 8d could be used as a hit compound for further optimization and might have potential to be developed as a new algicide.

Three series of azo dyes derived from 2-amino-5-aryl-1,3,4-thiadiazoles and aniline,N,N-dimethylaniline and phenol were synthesized in high yields by a conventional diazotization-coupling sequence. The chemical structures of the prepared compounds were confirmed by(1)H-NMR,C-13-NMR, IR, UV-Vis spectroscopy, mass spectrometry and elemental analysis. In addition, the X-ray single crystal structure of a representative azo dye was presented. For explicit determination of the influence of a substituent on radiation absorption in UV-Vis range, time-dependent density functional theory calculations were performed.

An artificial flavin system has been firstly proved to employ an N(5)-adduct for a catalytic transformation. This mode of catalysis occurs in some flavoenzymes but it is unknown in chemocatalysis, still exclusively using only C(4a)-adducts. In our report, an ethylene-bridged biomimetic flavin has been shown to participate in the Mitsunobu esterification reaction as an alternative to dialkyl azodicarboxylate. The reaction occurs via a flavin N(5)-triphenylphosphane adduct and is catalytic from the point of view of the flavin, which is regenerated by oxygen. This approach distinguishes from other catalytic Mitsunobu reaction procedures which require an extra catalytic system.

The skeleton of the diterpene dehydroabietylamine was modified, and a set of 12-hydroxy-dehydroabietylamine derivatives was obtained. The compounds were screened in colorimetric Ellman’s assays to determine their ability to act as inhibitors for the enzymes acetylcholinesterase (AChE, from electric eel) and butyrylcholinesterase (BChE, from equine serum). Additional investigations concerning the enzyme kinetics were performed and showed 12-hydroxy-N-(4-nitro-benzoyl)dehydroabietylamine (13) and 12-hydroxy-N-(isonicotinoyl)dehydroabietylamine (17) as selective BChE inhibitors holding good inhibition constants K-i = 0.72 +/- 0.06 mu M and K-i = 0.86 +/- 0.19 mu M, respectively.

Overexpression of aldehyde dehydrogenase 1A1 (ALDH1A1) is associated with the occurrence and development of obesity and insulin resistance. Herein, a series of coumarin-based ALDH1A1 inhibitors were designed, synthesized and evaluated. Among them, compounds 10, 14 and 26 exhibited potent inhibitory activity against ALDH1A1 and high selectivity over ALDH1A2, ALDH1A3, ALDH2 and ALDH3A1. This study presents an efficient synthesis of 3-[6-(substituted-phenyl)-[1,2,4]triazolo[3,4-b][1,3,4] thiadiazol-3-yl]-1H-indazole via dehydrative condensation with cyclization of 4-amino-5-(1H-indazol-3-yl)-4H-[1,2,4]triazole-3-thiol and fluorinated or nonfluorinated carboxylic acids in presence of phosphorous oxychloride. The multistep reaction pathway proceeds through different compounds. Present synthesis has the advantages of easily accessible starting materials, convenient synthesis, simple reaction condition, wider substrate scope, and higher yield (75% to 90% isolated).

Preparation of an ordered mesoporous polypyrrole/carbon (PPy/OMC) composite has been described through a two-step nanocasting process using KIT-6 as a template. Characterization of the PPy/OMC nanocomposite by various analysis methods such as TEM, XRD, TGA, SEM and N-2 sorption confirmed the preparation of a material with ordered mesoporous structure, uniform pore size distribution, high surface area and high stability. This nanocomposite was then used for the immobilization of palladium nanoparticles. The nanoparticles were almost uniformly distributed on the support with a narrow particle size of 20-25 nm, confirmed by various analysis methods. Performance of the Pd@PPy/OMC catalyst was evaluated in the aerobic oxidation of various primary and secondary alcohols on water as a green solvent, giving the corresponding carboxylic acids and ketones in high yields and excellent selectivity. The catalyst could also be reused for at least 10 reaction runs without losing its catalytic activity and selectivity. High catalytic efficiency of the catalyst can be attributed to a strong synergism between the PPy/OMC and that of supported Pd nanoparticles.

The effect of Fe, Co and Ni promoters on supported MoS2 catalysts for hydrogenation of nitroarenes were systematically investigated via experiment, characterization and DFT calculation. It was found that the addition of promoters remarkably improved the reaction activity in a sequence of Ni > Co > Fe > Mo. Meanwhile Ni promoted catalyst with the best performance showed good recyclability and chemoselectivity for a wide substrate scope. The characterization results revealed that the addition of promoters decreased the interaction between Mo and support and facilitated the reductive sulfidation of Mo species to produce more coordinated unsaturated sites (CUS). DFT calculations showed that the addition of promoters increased the formation of CUS, and enhanced the adsorption of hydrogen. The influence degree of promoters followed the sequence Ni > Co > Fe > Mo, which was consistent with those of the activities. Nitrobenzene hydrogenation and hydrogen activation occurred at the S and Mo edge, respectively. The adsorbed hydrogen diffused from the Mo edge to the S edge to participate in the hydrogenation reaction. Mechanism investigation showed that the main reason for increased activity by the addition of promoters was the increase of amounts of CUS and the secondary reason was the augmentation of intrinsic activity of CUS. The present studies give a new understanding for promoter modified MoS2 catalysts applied for hydrogenation of nitroarenes.

beta-Glucuronidase is responsible for the catalytic deconjugation of beta-D-glucuronides. beta-Glucuronidase has evolved to be a viable molecular target for numerous therapeutic treatments. It plays a pivotal role in the metabolism of drugs and endogenous substances. Herein, we report the inhibitory potentials of newly developed and modular benzimidazole-triazolothiadiazole hybrids spaced through a phenyl linker (1-26) and their interactions with the beta-glucuronidase. All analogues showed IC50 values in the range of 1.30 +/- 0.10 to 44.10 +/- 0.80 mu M, and hence were found to have outstanding inhibitory potential as compare to the standard D-saccharic acid 1,4-lactone (IC50 = 48.4 +/- 1.25 mu M). These modular hybrids were successfully synthesized, rigorously characterized through various spectroscopic techniques. Molecular docking studies further revealed the potential interactions between the inhibitor and active amino acid site in beta-glucuronidase. These findings helped in identifying the potential for new drug candidates. A Plausible structure activity relationship (SAR) were established which suggested that variation in the inhibitory potential was mainly based upon the substituents attached to the phenyl ring.
