In the present study, a new synthetic strategy towardsN-acylated glycinamides was developed by the use of 1,3,5-triazinanes as formaldimine surrogates in the Ugi reaction. The targeted products were obtained in a combinatorial, diversity-oriented fashion in good yields. Further modifications allowed us to adapt this procedure for the one-pot two-step syntheses of a local anesthetic druglidocaine and several unsymmetrically substituted diketopiperazines.

Electrospray ionization (ESI) is widely used as an ionization source for the analysis of complex mixtures by mass spectrometry. However, different compounds ionize more or less effectively in the ESI source, meaning instrument responses can vary by orders of magnitude, often in hard-to-predict ways. This precludes the use of ESI for quantitative analysis where authentic standards are not available. Relative ionization efficiency (RIE) scales have been proposed as a route to predict the response of compounds in ESL In this work, a scale of RIEs was constructed for 51 carboxylic acids, spanning a wide range of additional functionalities, to produce a model for predicting the RIE of unknown compounds. While using a limited number of compounds, we explore the usefulness of building a predictor using popular supervised regression techniques, encoding the compounds as combinations of different structural features using a range of common fingerprints. It was found that Bayesian ridge regression gives the best predictive model, encoding compounds using features designed for activity coefficient models. This produced a predictive model with an R-2 score of 0.62 and a root-mean-square error (RMSE) of 0.362. Such scores are comparable to those obtained in previous studies but without the requirement to first measure or predict the physical properties of the compounds, potentially reducing the time required to make predictions.

Palladium-catalyzed cross-coupling reactions between carboxylic acids and aryl halides have several possible competitive pathways. Decarboxylative C-C bond coupling and C-H arylation are well established in the literature. However, direct C-O bond coupling between carboxylic acids and aryl halides has received little success. In this report, we describe a protocol for exclusive C-O bond formation, enabled by a bidentate N,N-ligand such as 1,10-phenanthroline. The reaction is general for a broad range of carboxylic acids and iodoarenes. Experimental evidence and computational results suggest a high energy barrier for the alternative pathway of decarboxylative carbon-carbon bond coupling.

We designed and synthesized new atpenin A5 analogs for SAR study. Most of the analogs lacked one or several functional groups in the side chain of atpenin A5, and the stereoisomers proved to be weak nematode complex 11 inhibitors. However, we determined that 4-epi-atpenin A5 was a potent nematode complex II inhibitor comparable to atpenin A5. Tandem catalysis (i.e., a process in which a desirable product is synthesized by a one-step process consisting of sequential reactions) has attracted intensive attention owing to its sustainable green and atom-economical characteristics. In this process, the utilization of a high-efficiency multifunctional catalyst is key. However, different functional sites integrated within the catalyst are required to be rationally designed and precisely engineered to guarantee the synergy between the catalytic reactions. Herein, a novel kind of hydrogenation-esterification tandem catalyst with metal/acid (alloy/polyoxometalates) active sites integrated within the metal-organic frameworks (MOFs) was prepared by a facile self-sacrificial template route. In this tandem catalyst, the MOF cavities served as tandem reactors, the PtNi alloy sites encapsulated within the MOF material acted as hydrogenation sites, and the solid phosphotungstic acid embedded in the MOF cavities provided esterification sites. This well-designed tandem catalyst showed outstanding activity and selectivity towards the one-step synthesis of amino-ester-type anesthetics (e.g., benzocaine) owing to the synergistic catalysis of the metal and acid sites. Clearly, this novel tandem catalyst simplifies the traditional industry process and provides a new method to rationally construct new tandem catalysts.

A palladium-catalyzed tandem dehydrogenative [4 + 2] annulation of terminal olefins with N-sulfonyl amides via C(sp(2))-H activation, allylic C(sp(3))-H activation, and homoallylic C(sp(3))-H elimination processes has been developed. Promoted by the DMSO ligand, various benzamides, heterocyclic arylamides, alkenyl carboxamides, and commercial olefins are found to be efficient substrates to construct important heterocyclic compounds bearing a vinyl substituent with high E stereoselectivity. Using air as the terminal oxidant also provides a great advantage regarding environmental friendliness.

Assessment of ocular irritation risk is an international regulatory requirement in the safety evaluation of products. In response to this need, L’Oreal developed the SkinEthie Human Corneal Epithelium (HCE) Eye Irritation Test (Err) that has been included in OECD Test Guideline 492. SkinEthic (TM) HCE EIT is able to correctly and reliably identify chemicals not requiring classification versus labelling for eye irritation or serious eye damage according to UN GHS. In an effort to promote its global use, the performance of the method was evaluated after long-distance shipment and compared to European shipment conditions. Results obtained by Cosmos Technical Center (Japan) after extended tissues transit were compared to results obtained in L’Oreal (France). Thirty-nine out of 40 blinded chemicals, representing different functional chemical classes, were consistently classified in both laboratories. The SkinEthic (TM) HCE EIT test method was also evaluated for its performance after extended storage of the tissues. The performance was in agreement with the values reported in OECD TG 492, with an overall accuracy of 87.1% (based on 119 chemicals), sensitivity of 95.5% and specificity of 73.5%. The reliability and relevance of SkinEthic (TM) HCE Err test method after long-distance shipment and extended storage remain in agreement with regulatory validation criteria.

This research focuses on retention mechanisms in a LC column with C18 stationary phase when novel eluent additives (HFIP, HFTB and TFE as well as NFTB and perfluoropinacol) are used. The retention factors between novel eluent additives and conventional ones like ammonium acetate and ammonium bicarbonate at different eluent pH values were compared. A simple set of drug-like molecules, widely spread over different logP values, containing protonated and deprotonated acids and bases was selected for this investigation. HFIP, HFTB, NFTB and PP demonstrated strong influence on basic polar analytes in basic medium. These additives drastically increased retention. A decrease in retention was observed for acidic analytes when novel eluent additives were used. This paper reports the tandem reaction strategy of the Passerini/Wittig reaction based on the in situ capture of isocyanides. According to this strategy, plenty of isocyanides have been synthesized, which is immediately used for the tandem reaction of Passerini/Wittig reaction in one pot. Compared to the previous work, this strategy avoids the separation, purification, and storage of isocyanides, which prominently solves the problems of isocyanide-based multicomponent reaction such as: (a) The environmentally unfriendly (strong foul odor), (a) the labile of isocyanides, (c) high toxicity of isocyanides. In the meantime, in order to expand the application scope of our strategy, 1H-isochromenes and 3H-2-benzoxepin-1-ones have also been synthesized, which undergoes four-step transformations in one-pot. In addition, a relatively credible reaction mechanism has also been proposed, based on a series of control experiments. Furthermore, preliminary testing was performed on biological activity of some obtained compounds; These results showed that the synthesized compounds exhibited certain activity over P. digitatum and P. italicum.
