Chiral total syntheses of all six insecticidal natural pyrethrins (three pyrethrin I and three pyrethrin II compounds) contained in the chrysanthemum (pyrethrum) flower were performed. Three common alcohol components [(S)-cinerolone, (S)-jasmololone, and (S)-pyrethrolone] were synthesized: (i) straightforward Sonogashira-type cross-couplings using available (S)-4-hydroxy-3-methyl-2-(2-propynyl)cyclopent-2-en-1-ones (the prallethrin alcohol) for (S)-cinerolone (overall 52% yield, 98% ee) and (S)-pyrethrolone (overall 54% yield, 98% ee) and (ii) traditional decarboxylative-aldol condensation and lipase-catalyzed optical resolution for (S)-jasmololone (overall 16% yield, 96% ee). Two counter acid segments [(1R,3R)-chrysanthemic acid (A) and (1R,3R)-second chrysanthemic acid precursor (B)] were prepared: (i) C(1) epimerization of ethyl (+/-)-chrysanthemates and optical resolution using (S)-naphthylethylamine to afford A (96% ee) and (ii) concise derivatization of A to B (96% ee). All six pyrethrin esters (cinerin I/II, jasmolin I/II, and pyrethrin I/II) were successfully synthesized utilizing an accessible esterification reagent (TsCl/N-methylimidazole). To investigate the stereostructure-activity relationship, all four chiral stereoisomers of cinerin I were synthesized. Three alternative syntheses of (+/-)-jasmololone were investigated (methods utilizing Piancatelli rearrangement, furan transformation, and 1-nitropropene transformation). Insecticidal activity assay (KD50 and IC50) against the common mosquito (Culex pipiens pallens) revealed that (i) pyrethrin I > pyrethrin II, (ii) pyrethrin I (II) >> cinerin I (II) >> jasmolin I (II), and (iii) natural cinerin I >> three unnatural cinerin I compounds (apparent chiral discrimination).

Fluorescent silicon nanoparticles (SiNPs) displayed different emission wavelengths have been synthesized, but it has not been reported that the preparation of wavelength controllable SiNPs by adjusting the nitrogen content of reducing agents. In this paper, the wavelength-controlled fluorescent SiNPs were prepared by selecting the dopamine (DA) with nitrogen content between catechol and 2-aminophenol as the reducing agent and N-[3-(trimethoxysilyl) propyl]-ethylenediamine (DAMO) as the silicon source via one-step hydrothermal method. The emission wavelength of the prepared SiNPs was in direct proportion to the nitrogen content in the reducing agent. To the best of our knowledge, this is the first time for exploring the nitrogen content in reducing agents could affect the optical properties of SiNPs so far. In addition, the obtained SiNPs could be applied to determinate 2-nitrophenol (2-NP). Based on the combination action of inner filter effect (IFE) and static quenching effect (SQE) mechanism, a wide linear range was obtained from 0.1 to 500 mu M, and the limit of detection was 0.029 mu M for 2-NP, which was comparable to or even lower than some previous reports. This SiNPs probe was also successfully employed for sensing of 2-NP in industrial effluent with satisfactory results (98.6%-103.4%).

In patients with diabetes mellitus (DM), amputation rates exceed 30% when lower extremity osteomyelitis is present. We sought to determine the rate of osteomyelitis and any subsequent amputation in our patients with DM and lower extremity burns. We performed a single-site, retrospective review at our burn center using the institutional burn center registry, linked to clinical and administrative data. Adults (>= 18 years old) with DM admitted from January 1, 2014 to December 31, 2018 for isolated lower extremity burns were eligible for inclusion. We evaluated demographics, burn characteristics, comorbidities, presence of radiologically confirmed osteomyelitis, length of stay (LOS), inpatient hospitalization costs, and amputation rate at 3 months and 12 months after injury. We identified 103 patients with DM and isolated lower extremity burns. Of these, 88 patients did not have osteomyelitis, while 15 patients had radiologically confirmed osteomyelitis within 3 months of the burn injury. Compared to patients without osteomyelitis, patients with osteomyelitis had significantly increased LOS (average LOS 22.7 days vs 12.1 days, P = .0042), inpatient hospitalization costs (average $135,345 vs $62,237, P = .0008), amputation rate within 3 months (66.7% vs 5.70%, P < .00001), and amputation rate within 12 months (66.7% vs 9.1%, P < .0001). The two groups were otherwise similar in demographics, burn injury characteristics, access to healthcare, and preexisting comorbidities. Patients with DM and lower extremity burns incurred increased LOS, higher inpatient hospitalization costs, and increased amputation rates if radiologically confirmed osteomyelitis was present within 3 months of the burn injury. An efficient protocol was developed for the synthesis of pi-extended isocoumarins and polycyclic aromatic hydrocarbons based on the oxidative coupling of aromatic carboxylic acids with internal alkynes catalyzed by (cyclopentadienyl)rhodium complexes. Benzothiazole based oxadiazole derivatives 1-23 have been synthesized, characterized by (INMR)-I-1, (CNMR)-C-13, and MS and evaluated for alpha-glucosidase Inhibition. All analogs exhibited a varying degree of alpha-glucosidase inhibitory activity with IC50 values ranging in between 0.5 +/- 0.01-30.90 +/- 0.70 mu M when compared with the standard acarbose (IC50 = 866.30 +/- 3.20 mu M). Structure activity relationship has been established for all compounds. Molecular docking studies were performed to predict the binding interaction of the compounds with the active site of enzyme.

Currently, due to the HIV latency mechanism, the search continues for effective drugs to combat this issue and provide a cure for AIDS. Gnidimacrin activates latent HIV-1 replication and inhibits HIV-1 infection at picomolar concentrations. This natural diterpene was able to markedly reduce the latent HIV-1 DNA level and the frequency of latently infected cells. Therefore, gnidimacrin is an excellent lead compound, and its anti-HIV potential merits further investigation. The established structure activity relationship should be useful in the development of gnidimacrin or structurally related compounds as clinical trial candidates.
