This work describes the use of kinetics as a tool for rational optimization of an esterification process with down to equimolar ratios of reagents using a recyclable commercially available zirconocene complex in catalytic amounts. In contrast to previously reported group IV metal-catalyzed esterification protocols, the work presented herein circumvents the use of water scavengers and perfluorooctane sulfonate (PFOS) ligands. Insights into the operating mechanism are presented.

Poisonous weeds are a global problem since they not only hinder local economic development, but also cause ecological harm. Consolida rugulosa (family Ranunculaceae) is a weed that is widespread in Northwestern China and causes severe poisoning when ingested by livestock. In the present study, we purified the toxins in this plant and investigated their mechanism of action. Five natural diterpene alkaloids (compounds 1-5)-including two new compounds (1 and 2)-were isolated, and five semi-synthetic derivatives (6-10) were synthesised based on 4 or 5 for structure-activity analysis. The toxicity of the compounds was evaluated in vitro with lactate dehydrogenase (LDH) assay. All of the compounds-especially 1-stimulated LDH release in primary cultured rat myocardial cells, an effect that was blocked by the Na+ channel blocker lidocaine. Electrocardiography revealed that rats treated with 1 had severe arrhythmia, while heart Doppler echocardiography and analysis of serum biomarkers levels revealed that administration of 1 for 15 days induced changes in cardiac structure and myocardial enzyme levels. These effects were antagonised by lidocaine treatment. Thus, diterpene alkaloids are the main compounds responsible for the cardiotoxicity of C. rugulosa, which can be mitigated by co-administration of lidocaine.

Palladium nanoparticles (similar to 1-3 nm, 0.4 wt% Pd) were uniformly distributed over the surface of fibrous silica nanospheres (KCC-1) modified via aminopropyltriethoxysilane using a fast and cost-effective palladium (II) chloride reduction process. A wide range of substituted nitroarenes including electron withdrawing, releasing, sterically hindered and multifunctional groups have been converted to corresponding aryl formamide in quantitative yields (yields up to 98%) at moderate temperature (70 degrees C). Optimization study has proved that the 6 equivalent of formic acid is required and toluene was found to be the better solvent. The established practice is beneficial due to the use of formic acid as H-2 source and formylating agent, easiness in handling of the catalyst and simple workup procedure with efficient catalyst reusability. Endeavor to discover biorational natural products-based insecticides, two series (27) of novel 9R/S-acyloxy derivatives of cinchonidine and cinchonine were prepared and assessed for their insecticidal activity against Mythimna separata in vivo by the leaf-dipping method at 1 mg/mL. Among all the compounds, especially derivatives 6l and 6o exhibited the best insecticidal activity with final mortality rates of 75.0% and 71.4%, respectively. Catalysts that control stereochemistry are prized tools in chemical synthesis. When an effective catalyst is found, it is often explored for other types of reactions, frequently under the auspices of different mechanisms. As successes mount, a unique catalyst scaffold may become viewed as privileged. However, the mechanistic hallmarks of privileged catalysts are not easily enumerated or readily generalized to genuinely different classes of reactions or substrates. We explored the concept of scaffold uniqueness with two catalyst types for an unusual atropisomerselective cyclodehydration: (a) C-2-symmetric chiral phosphoric acids and (b) phosphothreonine-embedded, peptidic phosphoric acids. Pragmatically, both catalyst scaffolds proved fertile for enantioselective/atroposelective cyclodehydrations. Mechanistic studies revealed that the determinants of often equivalent and high atroposelectivity are different for the two catalyst classes. A data-descriptive classification of these asymmetric catalysts reveals an increasingly broad set of catalyst chemotypes, operating with different mechanistic features, that creates new opportunities for broad and complementary application of catalyst scaffolds in diverse substrate space.

A catalyst-free and solvent-free method for the hydroboration of a variety of carboxylic acids with pinacolborane was developed. The hydroboration of various aromatic and aliphatic carboxylic acids as well as dicarboxylic acids with HBpin could be completed within 6 h at room temperature or within 1 h at 60 degrees C to give the products in quantitative yields under neat conditions without the need for any solvent or metal catalyst. The possible reaction mechanism was investigated in detail based on the corresponding DFT calculations and the stoichiometric reaction of acetic acid with different equivalents of HBpin (at room temperature and 0 degrees C) and it revealed the stepwise nature of the protocol.

SCM-198 (Leonurine) has attracted great attention due to its cardioprotective effects in myocardial infarction (MI). However, no systematic modifications and structure-activity relationship (SAR) studies could be traced so far. In this study, 35 analogs of SCM-198 were designed, synthesized and their cardioprotective effects were evaluated. The cell viability assay on cardiomyocyte cell line H9c2 challenged with H2O2 showed that several analogs exhibited more potent cytoprotective effects than SCM-198 at 1 mu M and 10 mu M concentrations. LDH release level in cells treated with 1 mu M 14o was comparable with cells treated with 10 mu M SCM-198. Results of Bcl-2 expression and caspase-3 activation accordingly indicated higher protective activity of 14o than SCM-198. Moreover, in a mouse model of MI, the mice pretreated with 14o had much lower infarct size compared with that of SCM-198. The mechanism study suggested that 14o improved cardiac morphology and reduced apoptosis of cardiomyocytes in the border zone of infarction, as proved by H&E and TUNEL staining. An expeditious method for the synthesis of homo and heterochiral dipeptides containing L-alanine and D/L 2-methyl allo-hydroxyl prolines was developed using direct aminolysis of bicyclic lactones derived from D/L alanine. The impact of C-2 methylation and its spatial orientation on the pyrrolidine ring puckering and prolyl amide bond configuration was ascertained by solution NMR studies. The present studies reveal that C-2 methylation causes the prolyl amide bond to exist exclusively in the trans geometry in both homo-and heterochiral dipeptides. However, the spatial orientation of the C-2 methyl group and its i + 2 position in appropriately capped model dipeptides may nucleate into a turn like structure.

This paper emphasizes the preparation of nonlinear optical (NLO) responsive chromophores and their corresponding polymers. Initially, the carboxyl acid group-based precursors of the chromophores containing strong acceptors such as nitro-substituted thiazole and thiadiazole groups were prepared and subjected to condensation reactions with a monomer to yield a series of polymers. Prior to the synthesis of the polymers, a thermally crosslinkable monomer, 2,6-di-(octy-1-nyl)-p-phenylenediamine, was synthesized by a Sonogashira coupling reaction. All the synthesized precursors, the monomer and their corresponding polymers were characterized using FTIR, H-1 NMR, and CHN analyses and UV-Vis spectrophotometry. Simultaneously, the molecular structures of the precursors were designed and optimized using the Gaussian 16 (Revision A.03) program. The dipole moments, energy band gaps between the HOMO and LUMO, polarizabilities and first order hyperpolarizabilities of the precursors were calculated theoretically using the DFT approach; the values were in the ranges of 9.4321-15.3354 D, 1.3709-1.4146 eV, 53.40-64.80 x 10(-24) esu and 3703.35-5254.90 x 10(-32) esu, respectively. The inherent viscosities (eta inh) of the polymers were measured by an Ubbelohde viscometer and were in the range of 0.2453-0.2860 dl g(-1). The thermal behavior of the polymers was investigated using DSC and TGA. The glass transition temperatures of the polymers were in the range of 190-218 degrees C. The thicknesses and refractive indices of the thin films were determined by ellipsometry, and the values were in the range of 0.100-0.127 mu m and 1.211-1.426, respectively. The molecular orientations in the thin polymer films were induced by corona poling and were ascertained using both a UV-Vis spectrophotometer and an atomic force microscope. The second harmonic generations (SHGs) of the poled polymers were determined using an Nd-YAG laser. The SHG coefficients (d(33)) ranged between 98.30 and 106.04 pm V-1 at 532 nm. Among the synthesized polymers, the polymer containing dinitro-substituted thiazole demonstrated an excellent SHG coefficient as high as 106.04 pm V-1. Furthermore, none of the polymers showed SHG decay below 100 degrees C, and they retained 95% of their SHG values even up to 600 h. Based on its enhanced NLO efficiency and long temporal stability, the polymer containing dinitro-substituted thiazole can be a particularly promising candidate for photonic devices.
