In the current study, two binuclear Pt (II) complexes, containing cis, cis-[Me2Pt (mu-NN) (mu-dppm) PtMe2] (1), and cis,cis-[Me2Pt(mu-NN)(mu dppm,) Pt((CH2)(4))] (2) in which NN = phthalazine and dppm = bis (diphenylphosphino) methane were evaluated for their anticancer activities and DNA/purine nucleotide binding properties. The results of ethidium bromide/acridine orange staining and Caspase-III activity suggest that these complexes were capable to stimulate an apoptotic mechanism of cell death in the cancer cells. Using different biophysical techniques and docking simulation analysis, we indicated that these complexes were also capable to interact efficiently with DNA via a non-intercalative mechanism. According to our results, substitution of cyclopentane (in complex 2) with two methyl groups (in complex 1) results in significant improvement of the complex ability to interact with DNA and subsequently to induce the anticancer activity. Overall, these binuclear Pt (II) complexes are promising group of the non-classical potential anticancer agents which can be considered as molecular templates in designing of highly efficient platinum anticancer drugs. The transcription factor nuclear factor-kappa B (NF-kappa B) controls many physiological processes including inflammation, immunity, and apoptosis. In this study, a novel series of 6-phenoxy-[1,2,4] triazolo[3,4-a] phthalazine-3-carboxamide derivatives were synthesized as potent anti-inflammatory agents, which acted on tumor necrosis factor (TNF-alpha) as inhibitors of NF-kappa B activation. We showed that compounds 6h (6-(2,4-dichlorophenoxy)-[1,2,4] triazolo[3,4-a] phthalazine-3-carboxamide) and 6i (6-(3-tolyloxy)[1,2,4] triazolo[3,4-a]phthalazine-3-carboxamide) showed more prominent anti-inflammatory activity than other compounds, with similar activities as the reference drug dihydrotanshinone; compound 6i showed the lowest cellular toxicity among the tested compounds. In vivo evaluation of the anti-inflammatory activity showed that compound 6i exhibited excellent anti-inflammatory activity with 58.19% inhibition at 50 mg/kg intraperitoneal (i.p.), with equal efficacy as the positive control indomethacin (100 mg/kg i.p.; 59.21% inhibition). In this contribution, we study their structure and electronic states by means of a coupled-cluster method. The computed properties are compared to those of cinnoline which were obtained in our previous study [Etinski et al., Phys. Chem. Chem. Phys., 2014, 16, 4740]. The excited state features of these isomers are dependent on the position of the nitrogen atoms. We find that quinazoline and quinoxaline exhibit similarities in the ordering and character of the excited states. In contrast, a marked difference in the electronic and geometric structures of the lowest excited triplet states of cinnoline and phthalazine is noticed, although both are orthodiazanaphthalenes. Our findings suggest that the S-1 (sic) T-1 channel is responsible for the rapid intersystem crossing in quinazoline and quinoxaline, whereas the S-1 (sic) T-2 pathway is active in phthalazine.
