Anticancer activity assessment of two novel binuclear platinum (II) complexes

In the current study, two binuclear Pt (II) complexes, containing cis, cis-[Me2Pt (mu-NN) (mu-dppm) PtMe2] (1), and cis,cis-[Me2Pt(mu-NN)(mu dppm,) Pt((CH2)(4))] (2) in which NN = phthalazine and dppm = bis (diphenylphosphino) methane were evaluated for their anticancer activities and DNA/purine nucleotide binding properties. These Pt (II) complexes, with the non-classical structures, demonstrated a significant anticancer activity against Jurkat and MCF-7 cancer cell lines. The results of ethidium bromide/acridine orange staining and Caspase-III activity suggest that these complexes were capable to stimulate an apoptotic mechanism of cell death in the cancer cells. Using different biophysical techniques and docking simulation analysis, we indicated that these complexes were also capable to interact efficiently with DNA via a non-intercalative mechanism. According to our results, substitution of cyclopentane (in complex 2) with two methyl groups (in complex 1) results in significant improvement of the complex ability to interact with DNA and subsequently to induce the anticancer activity. Overall, these binuclear Pt (II) complexes are promising group of the non-classical potential anticancer agents which can be considered as molecular templates in designing of highly efficient platinum anticancer drugs. Simultaneous Determination of Sulfamethoxazole and Phthalazine by HPLC and Multivariate Calibration Methods

Two multivariate calibration methods are compared for the simultaneous chromatographic determination and separation of Sulfamethoxazole (SMX) and Phthalazine (PHZ) by High Performance Liquid Chromatography (HPLC). Multivariate calibration techniques such as Classical Least Squares (CLS) and Inverse Least Squares (ILS) were introduced into HPLC to determine the quantification by using UV detector at 235, 250, 260 and 270 nm. Sixteen binary mixtures of SMX and PHZ as calibration set and eight binary mixtures as prediction set were used. Results show that, Relative Errors of Prediction (REP) of CLS and ILS for SMX and PHZ were 0.17%, 0.63% and 0.15%, 0.56%, respectively.

Synthesis and Reactions of Some New Benzylphthalazin-1-ylaminophenols, 2H-Chromene and 5H-Chromeno[2,3-d]pyrimidine Derivatives with Promising Antimicrobial Activities

A series of (4-benzylphthalazin-1-ylamino) phenols 4-6 were prepared. Bromination of (4-benzylphthalazin-1-ylamino) phenol (1) with bromine afforded the bromophenol derivative 7, while condensation of the isomeric (4-benzylphthalazin-1-ylamino) phenol (2) or 7-(4-benzylphthalazin-1-ylamino)naphthalene-2-ol (3) with arylidenemalononitriles 8a-c in ethanol/piperidine solution afforded chromene derivatives 9a-c and 10a,b. Treatment of 7-(4-benzylphthalazin- 1-ylamino)-4-(p-methoxyphenyl)-4H-chromene-3-carbonitrile (9a) with triethyl orthoformate/Ac2O afforded ethoxymethyleneamino derivative 11. Condensation of 9a with DMFDMA/xylene gave chromenopyrimidine derivative 13, while treatment of 9a with Ac2O afforded the N-acetylimino derivative 14. The structures of the newly synthesized derivatives were confirmed by their elemental analysis and spectral data. The antimicrobial activities of some selected compounds were also studied and some of them were found to exhibit promising effects against Gram-positive and Gram-negative bacteria and fungi.

1-Piperazinylphthalazines as potential VEGFR-2 inhibitors and anticancer agents: Synthesis and in vitro biological evaluation

In our endeavor towards the development of effective VEGFR-2 inhibitors, three novel series of phthalazine derivatives based on 1-piperazinyl-4-arylphthalazine scaffold were synthesized. All the newly prepared phthalazines 16a-k, 18a-e and 21a-g were evaluated in vitro for their inhibitory activity against VEGFR-2. In particular, compounds 16k and 21d potently inhibited VEGFR-2 at sub-micromolar IC50 values 0.35 +/- 0.03 and 0.40 +/- 0.04 mu M, respectively. Moreover, seventeen selected compounds 16c-e, 16g, 16h, 16j, 16k, 18c-e and 21a-g were evaluated for their in vitro anticancer activity according to US-NCI protocol, where compounds 16k and 21d proved to be the most potent anticancer agents. While, compound 16k exhibited potent broad spectrum anticancer activity with full panel GI(50) (MG-MID) value of 3.62 mu M, compound 21d showed high selectivity toward leukemia and prostate cancer subpanels [subpanel GI(50) (MG-MID) 3.51 and 5.15 mu M, respectively].
