Hydrolysis of the Amide Bond in L-Methionine- and L-Histidine-Containing Dipeptides in the Presence of Dinuclear Palladium(II) Complexes with Benzodiazines Bridging Ligands

H-1 NMR spectroscopy was applied to study the catalytic activity of dinuclear Pd(II)-aqua complexes with different benzodiazine bridging ligands, [{Pd(en)(H2O)}(2)(mu-qx)](4+)(Pd1), [{Pd(en)(H2O)}(2)(mu-qz)](4+)(Pd2) and [{Pd(en)(H2O)}(2)(mu-phtz)](4+)(Pd3) (qx, qz and phtz denote quinoxaline, quinazoline and phthalazine, respectively), in the hydrolytic cleavage of the amide bond inN-acetylated L-methionylglycine (Ac-L-Met-Gly) and L-histidylglycine (Ac-L-His-Gly) dipeptides. All reactions were investigated with an equimolar amount of the reactants at pH = 2.0-2.5 in D2O and at 37 degrees C. The obtained data for the catalytic activity ofPd1-Pd3complexes are compared with those previously reported for [{Pt(en)(H2O)}(2)(mu-L)](4+)(L denotes benzodiazine: qx, qz and phtz), [{Pd(en)(H2O)}(2)(mu-L)](4+)and [{Pt(en)(H2O)}(2)(mu-L)](4+)(L denotes diazine: pyrazine and pyridazine) complexes. It was found that catalytic activity of these complexes in peptide cleavage is strongly related to the position of the nitrogen atoms in the benzodiazine or diazine bridging ligand. The investigated dinuclear Pd(II) and Pt(II) complexes show catalytic activity in the selective hydrolysis of the Met-Gly amide bond of Ac-L-Met-Gly dipeptide. Moreover, all the above mentioned Pd(II) complexes were also able to catalyze the regioselective hydrolysis of the His-Gly amide bond of Ac-L-His-Gly dipeptide. However, in the reaction with Ac-L-His-Gly, only Pt(II) aqua complexes containing bridging ligands with two nitrogen atoms in thepara-position (quinoxaline and pyrazine) were able to cleave this dipeptide.

Highly efficient red iridium(III) complexes based on phthalazine derivatives for organic light-emitting diodes

Two novel bis-cyclometalated iridium(III) complexes, Ir(ECPC)(2)(pic) and Ir(TPC)(2)(pic) (HECPC, 3-(4-(9H-carbazol-9-yl)phthalazin-1-yl)-9-ethyl-9H-carbazole; HTPC, 4-(4-(9H-carbazol-9-yl)phthalazin-1-yl)-N,N-diphenylaniline, pic = 2-picolinic acid) were synthesized in moderate yield under mild synthetic conditions, and their electrical and photophysical properties were characterized with respect to their application as a phosphorescent emitter in polymer light-emitting diodes (PLEDs). The devices based on these complexes with the structure of ITO/PEDOT:PSS (40 nm)/70%PVK:30%PBD:x% iridium(III) complex (70 nm)/TPBI (30 nm)/Ba (1.5 nm)/Al (120 nm) were fabricated with highly external quantum efficiency. The device based on Ir(ECPC)(2)(pic) exhibited a maximum external quantum efficiency of 16.3% and a luminance of 1484 cd/cm(2); while the device based on Ir(TPC)(2)(pic) showed a maximum external quantum efficiency of 11.9% and a Commission International de L’Eclairage (CIE) coordinates of (0.695,0.289). The results showed that the two new bis-cyclometalated iridium(III) complexes can be used as emitters and applied in saturated red phosphorescent organic light-emitting diodes (PHOLEDs). Anticancer Evaluation and Docking Study of New Bifunctional Phthalazine Derivatives

Aims and Objective: A series of novel phthalazine derivatives was synthesized with versatile, readily accessible electrophilic and nucleophilic reagents. The newly synthesized compounds were confirmed by the results of spectroscopic measurements. Hence, their potential clinical application investigated in particular for cancer treatment. Materials and Methods: The newly synthesized compounds were characterized by spectroscopic measurements and were tested for their in vitro anticancer activity by MTT assay against human liver cancer cell line. Docking study of all the synthesized compounds was performed within the active site of the enzyme VEGFR-2 (Vascular Endothelial Growth Factor Receptor-2). Results: The quinazoline derivative 12 emerged as the most potent compound in this study with an IC50 value of 5.4 mu M. Docking study showed that the synthesized compounds were fit in the VEGFR-2 active site almost at the same position of sorafenib and vatalanib with comparable docking scores (-15.20 to -8.92 was kcal/mol). Conclusion: we have synthesized a novel series of phthalazine derivatives and evaluated their potential anticancer activity against HEPG2 cell line. The quinazoline derivative 12 emerged as the most potent compound in this study with an IC50 value of 5.4 mu M. The SAR and docking studies pointed out that rigidification of the structure resulted in better activity and better binding within the active site of VEGFR-2 as in compounds 3, 5, 6 and 12. These results introduced new phthalazine derivatives having promising activity which could lead to the development of more potent anticancer agents.

Facile Synthesis, Characterization, and Antimicrobial Evaluation of Novel Heterocycles, Schiff Bases, and N-Nucleosides Bearing Phthalazine Moiety

The present work describes convenient synthesis of the novel Schiff bases 5a and b by reacting phthalazinones 4a and b with 4-methoxybenzaldehyde Reaction of the Schiff bases with phenylisothiocyanate afforded diazetidine derivatives 7a and b. Also, compounds 4a and b reacted with 2-bromoglucoside tetraacetate giving peracetylated N-glycosides 6a and b, which were deacetylated to afford N-glycosylated phthalazinones 8a and b. On the other hand, when compound 3 was treated with POCl3/PCl5 and/or ethyl chloroacetate, chlorophthalazine and ethyl acetate derivatives 9 and 10 were obtained, respectively. Hydrazinolysis of compounds 9 and 10 produced the hydrazino and hydrazide derivatives 11 and 12, respectively. When compound 11 reacted with 2-furanaldehyde, acetic anhydride, and/or carbon disulphide, it gave compounds 13-15, respectively. Treatment of the hydrazide 12 with aromatic aldehydes, acetic anhydride, ethyl acetoacetate, acetyl acetone, ammonium thiocyanate, and/or phthalic anhydride furnished compounds 17-21. Meanwhile, reacting Schiff base 22 with the chlorophthalazine derivative 9 produced compound 23, which on treatment with furoyl chloride produced compound 24. The structures of the novel compounds were confirmed by IR, H-1-NMR, C-13-NMR, MS, and elemental analysis. The newly synthesized compounds were tested against Bacillus subtilis and Staphylococcus aureus as Gram-positive bacteria, Escherichia coli and Pseudomonas aurignosa as Gram-negative bacteria, and Candida albicans and Aspergillus niger as fungi strains. Compounds 5a and b, 23, and 24 showed greater antimicrobial activity than the stranded compounds, suggesting that they could be considered as promising antimicrobial agents.

A magnetic inorganic-organic catalyst, PTA@Fe3O4/EN-MIL-101 (EN = ethylenediamine, PTA = phosphotungstic acid) was fabricated and characterized by XRD, HRTEM, FESEM, UV-vis, TGA-DTA, FT-IR, XPS and porosimetry. Phosphotungstic acid supported on SiO2@NHPhNH2 functionalized nanoparticles of MnFe2O4 as a recyclable catalyst for the preparation of tetrahydrobenzo[b]pyran and indazolo[2,1-b]phthalazine-triones

MnFe2O4@SiO2@NHPhNH2-phosphotungstic acid was synthesized by the reaction of diamine-modified silica coated manganese ferrite nanoparticles with H3PW12O40, the Keggin-type heteropoly acid. This is the first time that H3PW12O40 supported on diamine functionalized MnFe2O4 nanoparticles has been reported. The synthesized catalyst was characterized by various techniques, including Fourier Transform Infrared Spectroscopy (FTIR), Transmission electron-microscopy (TEM), X-ray Powder Diffraction (XRD), Scanning Electron Microscope (SEM), Energy Dispersive X-ray Spectroscopy (EDX), Inductively Coupled Plasma Atomic Emission Spectroscopy (ICP-AES), Thermogravimetric Analysis (TGA), and Vibrating Sample Magnetometer (VSM). The synthesized nanoparticle was examined as the heterogenous nanocatalyst for the synthesis of tetrahydrobenzo[b]-pyran derivatives and 2H-indazolo-[2,1-b]-phthalazine-triones under the solvent-free heating or ultrasonic irradiation conditions. This method offers several advantages including high yield, low reaction times, and the simple work up procedure due to the presence of the magnetic nanoparticles in this catalyst, so that, it can be easily recovered and reused for at least six successive times without the loss of its high catalytic activity, suggesting its great Potential for industrial applications. A mixed-ligand quinazoline-based Ni(II) Schiff base complex: Synthesis, characterization, crystal structure, antimicrobial investigation and catalytic activity for the synthesis of 2H-indazolo[2,1-b]phthalazine-triones

A tridentate Schiff base ligand, (E)-3-((2-hydroxy-3-methoxybenzylidene)amino)-2-methylquinazolin-4(3H)-one [HL], and its mixed-ligand Ni(II) complex [Ni(L)(imi)], were synthesized and fully characterized using elemental analysis, FT-IR, UV-Vis and (HNMR)-H-1 spectroscopy techniques. The structure of the synthesized ligand and complex was determined with single crystal X-ray diffraction method. In the complex, a square planner geometry was observed around the Ni(II) central atom coordinated with the donor atoms of the Schiff base ligand and one nitrogen of imidazole group. In addition, the catalytic activity of the complex on the three-component condensation of hydrazine hydrate with phthalic anhydride and dimedone to obtain 2H-indazolo[2,1-b]phthalazine-triones was investigated. Furthermore, in-vitro antimicrobial studies were performed that indicated the great antibacterial activities of the Ni(II) complex against Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Bacillus cereus bacteria.

An efficient, one-pot quantitative procedure for preparation of 1H-pyrazolo[1,2-b]phthalazine-5,10-dione derivatives from four-component condensation reaction of hydrazine monohydrate, phthalic anhydride, malononitrile or ethyl cyanoacetate, and aromatic aldehydes in the presence of magnetic Fe3O4 nanoparticles coated by (3-aminopropyl)-triethoxysilane as catalyst under mild, ambient, and solvent-free conditions is described. Simple procedure, high yield, short reaction time, and environmentally benign method are advantages of this protocol. The magnetic Fe3O4 nanoparticles coated by (3-aminopropyl)-triethoxysilane can be recovered and reused several times without loss of activity.
