Previously we have reported that 25-OCH3-PPD could suppress the reproduction of cancer cells and cause apoptosis without obvious toxicity. Herein, we aimed to enhance its bioactivity by introducing aromatic groups to its dammarane-type skeleton. These synthesized derivatives were tested for their inhibitory activities against five cancer cell lines. Of them, compounds 3a, 14a and 18a had the strongest antiproliferative activities against tumor cells (IC50 < 15 mu M, 5-fold to 10-fold increases than 25-OCH3-PPD). Especially compound 14a displayed the most potent activity against DU145, MCF-7 and HepG2 cells (IC50 = 6.7 +/- 0.8, 4.3 +/- 0.8 and 5.8 +/- 0.6 mu M, respectively). Structure-activity relationships demonstrated that having aromatic ester at the C3 position could improve the bioactivity. Alkene carbooxygenation has attracted considerable attention over the past few decades as this approach provides an efficient access to various oxygen-containing molecules, especially the valuable O-heterocycles. However, examples of catalytic alkene carbooxygenation via a direct C-O cleavage are quite scarce, and the C-O cleavage in these cases is invariably initiated by transition metal-catalyzed oxidative addition. We report here a novel Bronsted acid-catalyzed intramolecular alkoxylation-initiated tandem sequence, which represents the first metal-free intramolecular alkoxylation/Claisen rearrangement. Significantly, an unprecedented Bronsted acid-catalyzed intramolecular alkene insertion into the C-O bond via a carbocation pathway was discovered. This method allows the stereocontrolled synthesis of valuable indole-fused bridged [4.2.1] lactones, providing ready access to biologically relevant scaffolds in a single synthetic step from an acyclic precursor. Moreover, such an asymmetric cascade cyclization has also been realized by employing a traceless chiral directing group. Control experiments favor the feasibility of a carbocation pathway for the process. In addition, biological tests showed that some of these newly synthesized indole-fused lactones exhibited their bioactivity as antitumor agents against different breast cancer cells, melanoma cells, and esophageal cancer cells.

Piperine is a component of pepper which has earlier been reported as anticancer active compound. This work is emphasized on the design and synthesis of new hybrid piperine analogs by coupling piperine with the amine group of oxadiazoles and thiadiazoles. The new series of twelve piperine analogs was been tested for in vitro anti-proliferative activity using sulforhodamine B (SRB) assay test against MCF-7, PC-3, and HeLa cell lines. Among the twelve synthesised molecules piperine derivative with oxadiazole baring hydroxyl group (3) exhibits the higher activity against MCF-7 cell line than the reference drug Adriamycin and also displays the highest binding energy in the in silico studies. In patients with diabetes mellitus (DM), amputation rates exceed 30% when lower extremity osteomyelitis is present. We sought to determine the rate of osteomyelitis and any subsequent amputation in our patients with DM and lower extremity burns. We performed a single-site, retrospective review at our burn center using the institutional burn center registry, linked to clinical and administrative data. Adults (>= 18 years old) with DM admitted from January 1, 2014 to December 31, 2018 for isolated lower extremity burns were eligible for inclusion. We evaluated demographics, burn characteristics, comorbidities, presence of radiologically confirmed osteomyelitis, length of stay (LOS), inpatient hospitalization costs, and amputation rate at 3 months and 12 months after injury. We identified 103 patients with DM and isolated lower extremity burns. Of these, 88 patients did not have osteomyelitis, while 15 patients had radiologically confirmed osteomyelitis within 3 months of the burn injury. Compared to patients without osteomyelitis, patients with osteomyelitis had significantly increased LOS (average LOS 22.7 days vs 12.1 days, P = .0042), inpatient hospitalization costs (average $135,345 vs $62,237, P = .0008), amputation rate within 3 months (66.7% vs 5.70%, P < .00001), and amputation rate within 12 months (66.7% vs 9.1%, P < .0001). The two groups were otherwise similar in demographics, burn injury characteristics, access to healthcare, and preexisting comorbidities. Patients with DM and lower extremity burns incurred increased LOS, higher inpatient hospitalization costs, and increased amputation rates if radiologically confirmed osteomyelitis was present within 3 months of the burn injury. A new series of Pyrazolo[3,4-d]pyrimidine containing amide derivatives (8 a-l) were designed, synthesized, and evaluated for their in vitro alpha-amylase inhibitory activity. The IC50 values of the target compounds ranged from 1.60 +/- 0.48 to 2.04 +/- 1.20 mu M as compared to the standard acarbose 1.73 +/- 0.05 mu M. All the Pyrazolo[3,4-d]pyrimidine amide derivatives displayed good inhibitory activities, while seven analogs (8 d, 8 f, 8 g, 8 h, 8 i, 8 j and 8 k) exhibited more or less equipotent activity with IC50 values 1.77 +/- 2.84, 1.65 +/- 0.45, 1.66 +/- 2.24, 1.73 +/- 0.37, 1.60 +/- 0.48, 1.75 +/- 0.36 and 1.64 +/- 0.03 mu M respectively. Further, the most potent alpha-amylase inhibitors 8 d and 8 k were also screened for their in vivo antidiabetic activity against alloxan induced diabetic rat model at the dose of 25 and 50 mg/kg. Oral administration of these tested compounds significantly reduced the fasting blood glucose levels in dose dependent manner. The hypoglycemic effects of these compounds were more evident at 3 h and 5 h after administration of tested compounds which was similar to the effect displayed by the positive control. In addition, the binding energies calculated from the docking studies with the alpha-amylase enzyme (PDB ID: 1HNY) and biological activities indicate that the compounds containing nitro moiety on the phenyl group contributed significantly towards the antidiabetic activity.

A novel polar stationary phase for hydrophilic interaction chromatography (HILIC) is prepared by surface-initiated polymerization of hyperbranched polyglycerol onto silica. Such polymerization offers an easy way to tune the cabon load. Although there is much interest in the use of azobenzenes (ABs) as molecular switches for biological and material science applications, there is still need for systematic rationalization of their photophysical properties determined at both experimental and theoretical level. We present here the (re-) evaluation of the properties of some AB derivatives in the context of their geometrical substitution (para or meta) and nature of substituent effects (i.e. electron-donating groups, EDGs or electron-withdrawing groups, EWGs). Results of comprehensive experimental studies of their photophysical properties, thermal stabilities and functioning in reducing environments are rationalized by DFT calculations. As the associated n-pi* bands of the trans (E) and cis (Z) isomers of the studied ABs overlap, it is difficult to obtain high cis-to-trans conversion, however we have proved that by manipulation of the substituents position, the content of the cis isomer at the photostationary state (PSS) could be enhanced up to 30%. Moreover, we show that the presence of -NHCO- group at para positions contributes to reducing the effective bond order of the azo group resulting in the reduction of the height of the energy barrier for conversion and reduces the half-life of the cis isomer. Finally, electron deficient ABs are found to be more stable under reducing environments.
