Herein, a new iodide-mediated three-component annulation reaction of secondary anilines, cyclohexanones, and elemental sulfur is demonstrated, which allows access to various phenothiazines with the merits of formation of multiple chemical bonds in one single operation, high step and atom efficiency, readily available feedstocks and catalyst system, and good substrate and functional group compatibility. The developed chemistry capable of constructing novel phenothiazines with structural diversity offers a significant basis for further applications.

A magnetic catalyst is synthesized through growth of dendrimer of generation 3 (G3) on the surface of halloysite (Hal) nanotubes followed by decoration of dendrimer terminal groups with ionic liquid (IL) and incorporation of Fe species. The structure of the catalyst was confirmed by using several characterization techniques. Moreover, the catalytic activity of the catalyst for oxidation of benzaldehydes to the corresponding benzoic acids in the presence of hydrogen peroxide was investigated. Notably, to improve the yield of the reaction, beta-cyclodextrin (beta-CD) was employed as phase transfer agent. The contribution of dendrimer and IL to the catalysis was confirmed by comparing the catalytic activity of the catalyst with that of control the catalysts (the catalyst without IL, Fe species and dendrimer). Furthermore, the effect of dendrimer generation was elucidated by comparing the catalytic activities of the catalysts prepared by growth of dendrimers of G1, G2 and G3 on Hal. Studying the recyclability of the catalyst, it was found that the catalyst could be recovered and recycled for several reaction times with slight loss of leaching of Fe species.

Experimental mole fraction solubilities have been determined for 27 crystalline nonelectrolyte organic compounds and for one organometallic compound (ferrocene) dissolved in methyl butyrate at 298.15 K. The crystalline compounds cover a wide range of polarity and hydrogen-bonding character. Abraham model correlations were derived by combining the solubility data measured in the current study with published values retrieved from the chemical and engineering literature. In this communication, we disclose the first example of metal-free and site-selective radical fluorination of readily available tertiary alkyl ethers, enabled by synergistic photocatalysis and organocatalysis. This catalytic combination allows for exclusive fluorination of tertiary C-O bonds under mild conditions even in the presence of competing reaction sites. The excellent functional group tolerance affords valuable access to sterically hindered alkyl fluorides through late-stage modification of complex molecules. The successful use of tertiary alkyl ethers in radical fluorination enhances the structural diversity of aliphatic fluorides that can be derived from naturally abundant alcohols.

Organocatalytic asymmetric Michael addition between 3-substituted oxindoles and enals catalyzed by chiral camphor sulfonyl hydrazines (CaSHs) has been developed. Boronic esters are versatile building blocks extensively used in organic chemistry and essential to a variety of coupling reactions. In this work, the hydroboration reactions of alkynes were performed without metal catalysts using concomitant microwave irradiation that allowed a significantly reduced reaction time. Another interesting outcome of this study was that the reaction could be carried out under solvent-free conditions, highlighting the environmentally-friendly nature of our method. Objective: The aim of this study is to investigate the inhibitory effect of camptothecin derivative 3j on Non-Small Cell Lung Cancer (NSCLCs) cells and the potential anti-tumor mechanisms. Background: Camptothecin compounds are considered as the third largest natural drugs which are widely investigated in the world and they suffered restriction because of serious toxicity, such as hemorrhagic cystitis and bone marrow suppression. Methods: Using cell proliferation assay and S180 tumor mice model, a series of 20(S)-O-substituted benzoyl 7-ethylcamptothecin compounds were screened and evaluated the antitumor activities in vitro and in vivo. Camptothecin derivative 3j was selected for further study using flow cytometry in NSCLCs cells. Cell cycle related protein cyclin A2, CDK2, cyclin D and cyclin E were detected by Western Blot. Then, computer molecular docking was used to confirm the interaction between 3j and Topo L Also, DNA relaxation assay and alkaline cornet assay were used to investigate the mechanism of 3j on DNA damage. Results: Our results demonstrated that camptothecin derivative 3j showed a greater antitumor effect in eleven 20(S)-O-substituted benzoyl 7-ethylcamptothecin compounds in vitro and in vivo. The IC50 of 3j was 1.54 +/- 0.41 mu M lower than irinotecan with an IC50 of 13.86 +/- 0.80 mu M in NCI-H460 cell, which was reduced by 8 fold. In NCI-H1975 cell, the IC50 of 3j was 1.87 +/- 0.23 mu M lower than irinotecan (IC50 +/- SD, 5.35 +/- 0.38 mu M), dropped by 1.8 fold. Flow cytometry analysis revealed that 3j induced significant accumulation in a dose-dependent manner. After 24h of 3j (10 mu M)treatment, the percentage of NCI-H460 cell in S-phase significantly increased (to 93.54 +/- 4.4%) compared with control cells (31.67 +/- 3.4%). Similarly, the percentage of NCI-H1975 cell in S-phase significantly increased (to 83.99 +/- 2.4%) compared with control cells (34.45 +/- 3.9%) after treatment with 10 mu M of 3j. Moreover, increased levels of cyclin A2, CDK2, and decreased levels of cyclin D, cyclin E further confirmed that cell cycle arrest was induced by 3j. Furthermore, molecular docking studies suggested that 3j interacted with Topo I-DNA and DNA-relaxation assay simultaneously confirmed that 3j suppressed the activity of Topo I. Research on the mechanism showed that 3j exhibited anti-tumour activity via activating the DNA damage response pathway and suppressing the repair pathway in NSCLC cells. Conclusion: Novel camptothecin derivative 3j has been demonstrated as a promising antitumor agent and remains to be assessed in further studies.
