Drug-resistant strains of Mycobacterium tuberculosis are on the rise and have resulted in an urgent need to develop new tuberculosis (TB) drugs. Herein, the development of chromonyl-pyrimidines as a new TB drug scaffold is presented. A library of eleven chromonyl-pyrimidines was synthesised, from which lead compound 6-((2-amino-6-methylpyrimidin-4-yl)amino)-3-benzoyl-2-phenyl-4H-chromen-4-one (MIC=36 mu M against M. tuberculosis) was identified. A second-generation library of seven aryl-substituted chromonyl-pyrimidines was then prepared, with six chromonyl-pyrimidines exhibiting an improved M. tuberculosis growth inhibition (MIC=12.5 mu M). From this work, the potential of chromonyl-pyrimidines as therapeutics for TB is demonstrated.

Eco-friendly carbocatalyst is extremely desirable in organic catalysis and wastewater treatment. Herein, we reported a one-pot synchronous hydrothermal process to synthesize a highly efficient N doped holey graphene (NHG) carbocatalyst, in which graphene oxide nanosheets are self-assembled, reduced, chemically etched, and simultaneously doped with nitrogen to form a graphene hydrogel in the presence of H2O2 and NH4OH. Benefiting from the good hydrophilicity, unique hierarchical porous structure, large specific surface area (322.1 m(2)g(-1)), and high N content (9.77 at. %), the resultant NHG carbocatalyst exhibits an excellent catalytic performance for the hydrogenation of N-containing unsaturated compounds. In 4-nitrophenol reduction reaction, the NHG carbocatalyst delivers a turnover frequency of 3.32 x 10(-2) min(-1). This catalytic activity is superior to that of the commercial Pd/C (5.0 wt%), other previously reported carbocatalysts, and many noble-metal-based catalysts. Furthermore, the molecular structure analysis combined with density functional theory calculations elucidate metal-free catalytic mechanism of hydrogenation reaction for unsaturated chromophore groups in N-containing organic dyes. Metal-organic frameworks (MOFs) have recently emerged as efficient self-sacrificial templates to fabricate porous carbon-supported metal nanoparticles (NPs). Due to observed increased activity, catalysts containing bimetallic NPs represent an active frontier for heterogeneous catalyst development. A strategy to synthesize active catalysts with highly dispersed bimetallic metal/metal oxides within a porous carbon matrixviarapid MOF decomposition using dopants is presented in this paper. A 2,4,6-trinitrotoluene (TNT) additive enhances the rapid thermolysis of the metal-doped Cu-based MOFs to minimize particle aggregation. Catalyst characterization reveals that a secondary metal increases dispersion of both metals over the carbon composite support. The catalyst preparation method influences both the metal particle size and oxidation state. Catalytic performance shows increased rates for 4-nitrophenol reduction even with <1 wt% of added secondary metal. Among the synthesized catalysts, the Ni-CuO@C bimetallic catalyst exhibits outstanding activity. This synthetic strategy is useful for creating highly efficient, robust, non-noble metal catalysts for development of sustainable chemical processes. Human DNA topoisomerase II is one of the major targets in anticancer therapy, however ATP-competitive inhibitors of this target have not yet reached their full potential. ATPase domain of human DNA topoisomerase II belongs to the GHKL ATPase superfamily and shares a very high 3D structural similarity with other superfamily members, including bacterial topoisomerases. In this work we report the discovery of a new chemotype of ATP-competitive inhibitors of human DNA topoisomerase II alpha that were discovered through screening of in-house library of ATP-competitive inhibitors of bacterial DNA gyrase and topoisomerase IV. Systematic screening of this library provided us with 20 hit compounds. 1,2,4-Substituted N-phenylpyrrolamides were selected for a further exploration which resulted in 13 new analogues, including 52 with potent activity in relaxation assay (IC50 = 3.2 mu M) and ATPase assay (IC50 = 0.43 mu M). Cytotoxic activity of all hits was determined in MCF-7 cancer cell line and the most potent compounds, 16 and 20, showed an IC50 value of 8.7 and 8.2 mu M, respectively.

Assessment of ocular irritation risk is an international regulatory requirement in the safety evaluation of products. In response to this need, L’Oreal developed the SkinEthie Human Corneal Epithelium (HCE) Eye Irritation Test (Err) that has been included in OECD Test Guideline 492. SkinEthic (TM) HCE EIT is able to correctly and reliably identify chemicals not requiring classification versus labelling for eye irritation or serious eye damage according to UN GHS. In an effort to promote its global use, the performance of the method was evaluated after long-distance shipment and compared to European shipment conditions. Results obtained by Cosmos Technical Center (Japan) after extended tissues transit were compared to results obtained in L’Oreal (France). Thirty-nine out of 40 blinded chemicals, representing different functional chemical classes, were consistently classified in both laboratories. The SkinEthic (TM) HCE EIT test method was also evaluated for its performance after extended storage of the tissues. The performance was in agreement with the values reported in OECD TG 492, with an overall accuracy of 87.1% (based on 119 chemicals), sensitivity of 95.5% and specificity of 73.5%. The reliability and relevance of SkinEthic (TM) HCE Err test method after long-distance shipment and extended storage remain in agreement with regulatory validation criteria.

A simple, efficient, sustainable and economical method for the oxidation of alcohols and amines has been developed based on chloride, a sea abundant anionic catalyst for the practical synthesis of a wide range of carboxylic acids, ketones and imines. Oxidation of aromatic alcohols was carried out using NaCl (20 mol%) as the catalyst, NaOH (50 mol%) and aq. TBHP (4 equiv.) as the oxidant in 55-92% isolated yields. Oxidation of aromatic amines to imines was achieved by using only 20 mol% of NaCl and aq. TBHP (4 equiv.) in 32-93% isolated yields. The chlorine species formed during the reaction as the active oxidation catalyst has been identified as ClO2- for alcohols and ClO-/ClO2- for amines by control experiments. This method is mostly free from chromatographic purification, which makes it suitable for large-scale synthesis. We have scaled up to 30 gram scale the synthesis of carboxylic acids and imines in good yields and have also carried out efficiently this new method using filtered sea water as the solvent and catalyst.

Carbohydrate-based low-molecular-weight gelators are useful and versatile compounds for the preparation of soft materials. Using N-acetyl-D-glucosamine as the starting material, we synthesized and characterized 15 glycolipids containing an amide with different ester functional groups. These include aliphatic derivatives with varying chain lengths and aromatic derivatives. Most of the hybrid amide-esters have molecular weights less than 500 D. These glycolipids were found to be effective gelators for several organic solvents, water, and aqueous solutions. Two efficient hydrogelators were also obtained at low concentrations. A few representative gels were characterized using optical microscopy, atomic force microscopy, and rheology to obtain information on their morphology and gel stability. Three gelators were also used to encapsulate naproxen sodium and toluidine blue. The sustained release of the drug from the gel to the aqueous phase was monitored by UV-vis spectroscopy. These gelators have structural flexibility that can be stimuli responsive. The esters can be hydrolyzed and several gels were converted to solutions under basic conditions. These rationally designed gelators could be utilized as stimuli-responsive smart materials with controlled release properties.
