An asymmetric [3 + 2] formal cycloaddition reaction between oxindole alpha-keto esters and enals has been developed. This alpha,gamma-regioselective reaction afforded multifunctional carbocyclic oxindoles via dienamine-enamine cascade reaction followed by isomerization. Furthermore, a highly enantioselective Diels-Alder reaction was demonstrated between 2,4-dienals and oxindole alpha-keto esters via trienamine intermediates. Chiral secondary amine catalyzed the both reactions under mild conditions and the cyclized products were isolated in moderate to good yields with good diastereoselectivities (>20:1 dr) and enantioselectivities (up to 99 %).

The effect of topology on the structure, self-assembly, and selective Fe3+ binding of delta-peptides has been investigated. A series of delta-peptides with an amino acid containing dihydropyrimidinone and o-, m-, and p-aminobenzoic acids have been designed to study the structure-function relationship. A new amino acid containing dihydropyrimidinone was synthesized by the Biginelli reaction of ethyl acetoacetate, urea, and o-nitrobenzaldehyde followed by reduction with iron powder and acetic acid. X-ray crystallography sheds some light on the conformations, self-assembly, and the diverse degrees of pi-pi stacking of adjacent delta-peptide molecules. Peptides with o- or m-aminobenzoic acid form eight-membered intramolecular hydrogen-bonded turn conformations and self-assemble through intermolecular hydrogen bonds between dihydropyrimidinone units to form a butterfly-like structure. However, the delta-peptide containing p-aminobenzoic acid forms a water-mediated cage-like structure. Irrespective of the presence of the same functional groups, only the delta-peptide with o-aminobenzoic acid can selectively bind Fe3+ in methanol as well as in water. The topology plays a crucial role in the selective Fe3+ ion binding by the delta-peptide.

Design of chemically novel, biologically potent small heterocyclic molecules with anticancer activities, which targets the enzyme heparanase has gained prominent clinical interest. We have synthesized a novel class of carboxamide derivatives by coupling various substituted aromatic acid hydrazides and triazoleamine with pyrrolidine carboxylic acid by using coupling agents. The synthesized compounds are characterized by spectroscopic techniques such as FT-IR, DIMS and NMR. These compounds arc investigated for cytotoxicity on different cancer cell lines and heparanase inhibitory activity. Most of them showed moderate heparanase inhibitory activity and good cytotoxicity.

A chemoenzymatic synthesis of the title compound has been developed using an efficient and highly enantioselective lipase-catalyzed acylation in a hydrophobic ionic liquid, [bmim][PF6], followed by a diastereoselective asymmetric dihydroxylation as the key steps for incorporating the stereogenic centers. The further conversion to the appropriate intermediates and subsequent acylation with lauric acid furnished the target compound.

Despite numerous advances in spectroscopic methods through the latter part of the 20th century, the unequivocal structure determination of natural products can remain challenging, and inevitably, incorrect structures appear in the literature. Computational methods that allow the accurate prediction of NMR chemical shifts have emerged as a powerful addition to the toolbox of methods available for the structure determination of small organic molecules. Herein, we report the structure determination of a small, stereochemically rich natural product from Laurencia majuscula using the powerful combination of computational methods and total synthesis, along with the structure confirmation of notoryne, using the same approach. Additionally, we synthesized three further diastereomers of the L. majuscula enyne and have demonstrated that computations are able to distinguish each of the four synthetic diastereomers from the 32 possible diastereomers of the natural product. Key to the success of this work is to analyze the computational data to provide the greatest distinction between each diastereomer, by identifying chemical shifts that are most sensitive to changes in relative stereochemistry. The success of the computational methods in the structure determination of stereochemically rich, flexible organic molecules will allow all involved in structure determination to use these methods with confidence.

Herein we describe the synthesis of imidazo [2,1-b][1,3,4]thiadiazoles from carbohydrates with D-ribo and D-xylo configuration. The antiviral activity of these compounds was tested against Junin virus (the etiological agent of Argentine hemorrhagic fever). The p-chlorophenyl derivatives showed antiviral activity in a range of micromolar concentration.

New chiral regioisomeric gamma-hydroxyphosphine ligands were synthesized from alpha-pinene. A simple and general approach towards the total syntheses of several iridolactones such as (+/-)-boschnialactone, (+/-)-7-epi-boschnialactone, (+/-)-teucriumlactone, (+/-)-iridomyrmecin, (+/-)-isoboonein, (+/-)-7-epi-argyol, (+/-)-scabrol A, (+/-)-7-epi-scabrol A, and (+/-)-patriscabrol as well as the putative structure of scholarein A is delineated. The synthetic strategy features a diastereoselective intramolecular Pauson-Khand reaction (IPKR) to construct the iridoid framework followed by some strategic synthetic manipulations to access the targeted monoterpenes including those having diverse oxy-functionalization patterns and with 3-5 contiguous stereogenic centres in a highly stereocontrolled manner. The effect of Fe, Co and Ni promoters on supported MoS2 catalysts for hydrogenation of nitroarenes were systematically investigated via experiment, characterization and DFT calculation. It was found that the addition of promoters remarkably improved the reaction activity in a sequence of Ni > Co > Fe > Mo. Meanwhile Ni promoted catalyst with the best performance showed good recyclability and chemoselectivity for a wide substrate scope. The characterization results revealed that the addition of promoters decreased the interaction between Mo and support and facilitated the reductive sulfidation of Mo species to produce more coordinated unsaturated sites (CUS). DFT calculations showed that the addition of promoters increased the formation of CUS, and enhanced the adsorption of hydrogen. The influence degree of promoters followed the sequence Ni > Co > Fe > Mo, which was consistent with those of the activities. Nitrobenzene hydrogenation and hydrogen activation occurred at the S and Mo edge, respectively. The adsorbed hydrogen diffused from the Mo edge to the S edge to participate in the hydrogenation reaction. Mechanism investigation showed that the main reason for increased activity by the addition of promoters was the increase of amounts of CUS and the secondary reason was the augmentation of intrinsic activity of CUS. The present studies give a new understanding for promoter modified MoS2 catalysts applied for hydrogenation of nitroarenes.

A series of novel 2, 5-disubstituted 1, 3, 4-Oxadiazole derivatives as a potential anti-inflammatory, and antioxidant agent were synthesized via cyclisation. Hydrazide molecule treated with substituted acids in the presence of phosphorus oxychloride (POCl3) as an efficient reagent as well as solvent by conventional method with shorter reaction time and excellent yield. The newly synthesized 1, 3, 4-oxadiazole derivatives exhibited excellent to good anti-inflammatory and anti-oxidant activities compaired to the standard drugs. Molecular docking study on the crucial anti-inflammatory target-cyclooxygenase-2 (COX-2) revealed the ability of the scaffold to correctly recognize the active site and achieve significant bonded and non-bonded interactions with key residues therein. This study could identify potential compounds which can be pertinent starting points for structure-based drug design to obtain newer anti-inflammatory agents.
