Cellulose graft copolymers are an effective way to endow new properties to cellulose substrate, as well the rigidity, regularity, and helicity of the cellulose backbone could induce the self-assembly of supramolecular structures. In this work, right-handed helical poly(phenyl isocyanide)s (PPIn) were grafted selectively onto C-6-cellulose. Alkyne-terminated PPIn was synthesized by living polymerization of right-handed phenyl isocyanide monomer using an alkyne-terminated palladium(II) complex as an initiator/catalyst, and were grafted onto the C-6 of the cellulose backbone (Cell 6 g PPIn) at various chain lengths using copper-catalyzed alkyne-azide cycloaddition (CuAAC) click chemistry. We confirmed the successful grafting by liquid H-1 NMR and C-13 NMR, as well as solid C-13 NMR, FTIR, and GPC. After grafting onto cellulose, the right-handed chirality of PPIn was significantly increased by 111.2%. Additionally, the Cell 6 g PPIn exhibited better chiral recognition of L-PheDNSP than PPIn alone. Therefore, the helical cellulose backbone has enhanced effect on preferred helix of PPIn.

The present study reports the convenient synthesis, spectroscopic characterization, bio-assays and computational evaluation of a novel series of N-acyl-1H-imidazole-1-carbothioamides. The screened derivatives displayed excellent antioxidant activity, moderate antibacterial and antifungal potential. The screened derivatives were found to be highly biocompatible against hRBCs. Molecular docking ascertained the mechanism and mode of action towards the molecular target delineating that ligands and complexes were stabilized at the active site by electrostatic and hydrophobic forces in accordance to the corresponding experimental results. Docking simulation provided additional information about the possibilities of inhibitory potential of the compounds against RNA. Computational evaluation predicted that N-acyl-1H-imidazole-1-carbothioamides 5c and 5g can serve as potential surrogates for hit to lead generation and design of novel antioxidant and antibacterial agents.

A novel strategy for the recognition of anions in water using charge-neutral a-hole halogen and chalcogen bonding acyclic hosts is demonstrated for the first time. Exploiting the intrinsic hydrophobicity of halogen and chalcogen bond donor atoms integrated into a foldamer structural molecular framework containing hydrophilic functionalities, a series of water-soluble receptors was constructed for an anion recognition investigation. Isothermal titration calorimetry (ITC) binding studies with a range of anions revealed the receptors to display very strong and selective binding of large, wealdy hydrated anions such as I- and ReO4-. This is achieved through the formation of 2:1 host-guest stoichiometric complex assemblies, resulting in an encapsulated anion stabilized by cooperative, multidentate, convergent a-hole donors, as shown by molecular dynamics simulations carried out in water. Importantly, the combination of multiple sigma-hole-anion interactions and hydrophobic collapse results in I- affinities in water that exceed all known a-hole receptors, including cationic systems (beta(2) up to 1.68 X 10(11) M-2). Furthermore, the anion binding affinities and selectivity trends of the first example of an all-chalcogen bonding anion receptor in pure water are compared with halogen bonding and hydrogen bonding receptor analogues. These results further advance and establish halogen and chalcogen bond donor functions as new tools for overcoming the challenging goal of anion recognition in pure water.

This work provides the pK(a)at the biorelevant temperature of 37 degrees C for a set of compounds proposed as internal standards for the internal standard capillary electrophoresis (IS-CE) method. This is a high throughput method that allows the determination of the acidity constants of compounds in a short time, avoiding the exact measurement of the pH of the buffers used. pH electrode calibration at 37 degrees C can be avoided too. In order to anchor the pK(a)values obtained through the IS-CE method in the pH scale, the acidity constant at 37 degrees C of some of the standards has been determined also by the reference potentiometric method. In general, a decrease in the pK(a)value is observed when changing the temperature from 25 to 37 degrees C, and the magnitude of the change depends on the nature of the compounds. Once the pK(a)values at 37 degrees C of the internal standards have been established, the method is applied to the determination of the acidity constants of seven polyprotic (5 diprotic and 2 triprotic) drugs. The obtained mobility-pH profiles show well-defined curves, and the fits provide precise pK(a)values. Due to the lack of reference data at 37 degrees C only the pK(a)values of labetalol can be compared to values from the literature, and a very good agreement is observed.

With the infection rate of Bordetella pertussis at a 60-year high, there is an urgent need for new anti-pertussis vaccines. The lipopolysaccharide (LPS) of B. pertussis is an attractive antigen for vaccine development. With the presence of multiple rare sugars and unusual glycosyl linkages, the B. pertussis LPS is a highly challenging synthetic target. In this work, aided by molecular dynamics simulation and modeling, a pertussis-LPS-like pentasaccharide was chemically synthesized for the first time. The pentasaccharide was conjugated with a powerful carrier, bacteriophage Q beta, as a vaccine candidate. Immunization of mice with the conjugate induced robust anti-glycan IgG responses with IgG titers reaching several million enzyme-linked immunosorbent assay (ELISA) units. The antibodies generated were long lasting and boostable and could recognize multiple clinical strains of B. pertussis, highlighting the potential of Q beta-glycan as a new anti-pertussis vaccine.

A convenient and efficient strategy for the synthesis of 2-acyl benzothiazoles/thiazoles has been developed. The treatment of benzothiazole/thiazole with allylic Grignard reagents readily generates the corresponding 2-Grignard reagents, which is followed by a reaction with N,N’-carbonyldiimidazole activated carboxylic acids to afford various 2-acyl benzothiazoles/thiazoles products. We report here a two-in-one strategy for the Pd(II)-catalyzed tandem C-H arylation/decarboxylative annulation between readily available cyclic diaryliodonium salts and benzoic acids. The carboxylic acid functionality can be used as both a directing group for the ortho-C-H arylation and the reactive group for the tandem decarboxylative annulation.
