In accordance with the significant impetus of the discovery of potent vascular endothelial growth factor receptor 2 (VEGFR-2) inhibitors, herein, we report the design, synthesis, and anticancer evaluation of 12 new N-substituted-4-phenylphthalazin-1-amine derivatives against HepG2, HCT-116, and MCF-7 cells as VEGFR-2 inhibitors. The results of the cytotoxicity investigation indicated that HCT-116 and MCF-7 were the most sensitive cell lines to the influence of the newly synthesized derivatives. In particular, compound 7a was found to be the most potent derivative among all the tested compounds against the three cancer cell lines, HepG2, HCT116, and MCF-7, with IC50 = 13.67 +/- 1.2, 5.48 +/- 0.4, and 7.34 +/- 0.6 mu M, respectively, which is nearly equipotent to that of sorafenib (IC50 = 9.18 +/- 0.6, 5.47 +/- 0.3, and 7.26 +/- 0.3 mu M, respectively). All synthesized derivatives, 4a,b-8a-c, were evaluated for their inhibitory activities against VEGFR-2. The tested compounds displayed high to low inhibitory activity, with IC50 values ranging from 0.14 +/- 0.02 to 9.54 +/- 0.85 mu M. Among them, compound 7a was found to be the most potent derivative that inhibited VEGFR-2 at an IC50 value of 0.14 +/- 0.02 mu M, which is nearly 72% of that of the sorafenib IC50 value (0.10 +/- 0.02 mu M). Compounds 7b, 8c, 8b, and 8a exhibited very good activity with IC50 values of 0.18 +/- 0.02, 0.21 +/- 0.03, 0.24 +/- 0.02, and 0.35 +/- 0.04 mu M, respectively. Molecular modeling studies were carried out for all compounds against the VEGFR-2 active site. The data obtained from biological testing highly correlated with that obtained from molecular modeling studies. However, these modifications led to new phthalazine derivatives with higher VEGFR-2 inhibitory activities than vatalanib and which are nearly equipotent to sorafenib.

Synthesis and in vitro anti-proliferative activity of some novel isatins conjugated with quinazoline/phthalazine hydrazines against triple-negative breast cancer MDA-MB-231 cells as apoptosis-inducing agents

In our endeavor to develop potent isatin-based anti-proliferative agents, we utilized the hybrid-pharmacophore approach to synthesize three series of novel isatin-based hybrids 5a-h, 10a-h and 13a-c, with the prime goal of developing potent anti-proliferative agents toward TNBC MDA-MB-231 cell line. In particular, compounds 5e and 10g were the most active hybrids against MDA-MB-231 cells (IC50 = 12.35 +/- 0.12 and 12.00 +/- 0.13 mu M), with 2.37-and 2.44-fold increased activity than 5-fluorouracil (5-FU) (IC50 = 29.38 +/- 1.24 mu M). Compounds 5e and 10g induced the intrinsic apoptotic mitochondrial pathway in MDA-MB-231; evidenced by the reduced expression of the anti-apoptotic protein Bcl-2, the enhanced expression of the pro-apoptotic protein Bax and the up-regulated active caspase-9 and caspase-3 levels. Furthermore, 10g showed significant increase in the percent of annexin V-FITC positive apoptotic cells from 3.88 to 31.21% (8.4 folds compared to control).

Thermostable PPESK/TiO2 nanocomposite ultrafiltration membrane for high temperature condensed water treatment

Poly (phthalazine ether sulfone ketone) (PPESK)/TiO2 organic-inorganic composite ultrafiltration (OF) membranes with enhanced performance for high temperature condensed water treatment were prepared using a phase-inversion method. The effects of TiO2 concentration on the membrane morphologies, physic-chemical properties, membrane permeability and antifouling performance were investigated and discussed. The SEM result reveals that the penetrated finger-like structure in sublayer is suppressed, and sponge-like structure begins to be developed. The mechanical strength and thermal stability of composite membrane are improved due to the existence of hydrogen bond between TiO2 and polymer. The permeate flux is also enlarged due to the improved hydrophilicity and porosity. Compared with neat PPESK membrane, the composite membranes present improved antifouling properties, showing lower filtration resistances, MEI values and better flux recover property during high temperature condensed water treatment. The optimal addition amount of TiO2 is 2 wt.%. The high temperature condensed water with excess oil and iron can be treated efficiently using PPESK/TiO2 composite membrane. The turbidity of feed solution is almost removed. The oil and iron concentrations in permeate solution are 0.89 mg/L and 26.4 mu g/L which meet the Quality Criterion of Water and Steam for Steam Power Equipment in China. (C) 2012 Elsevier B.V.
