The first synthesis of the pentasaccharide fragment of the angucycline antibiotic saquayamycin Z is described. By using our sulfonyl chloride mediated reagent controlled dehydrative glycosylation, we are able to assemble the glycosidic linkages with high levels of anomeric selectivity. The total synthesis was completed in 25 total steps, and in 2.5% overall yield with a longest linear sequence of 15 steps.

A chemoenzymatic synthesis of the title compound has been developed using an efficient and highly enantioselective lipase-catalyzed acylation in a hydrophobic ionic liquid, [bmim][PF6], followed by a diastereoselective asymmetric dihydroxylation as the key steps for incorporating the stereogenic centers. We report the desiging of ultrafine Pd-based nanocatalyst containing N-doped carbon structure (Pd@NC). The material was prepared by direct dehydration at 120 degrees C followed by mixing and heating with a dopant (ammonium oxalate) at 150 degrees C in a furnace. The Pd@NC nanocatalyst containing electron-rich pyridinic-N doped carbon structure, was thoroughly characterized by various techniques namely SEM, EDX, TEM, FTIR, ICP-AES, XRD, XPS, TGA and Raman spectroscopy. The utility of the Pd@NC nanocatalyst was explored for base-free Knoevenagel condensation and 4H-pyran derivatives and also in the reduction of nitroarenes under mild and greener conditions. Further, the optical property was explored using photoluminescence spectroscopy and band gap was also calculated. The heterogeneous nature and stability of the catalyst facilitated by its ease of separation for long-term performance and recycling studies showed that catalyst was robust and remained active upto six recycling experiments. Also, the leaching of metal was confirmed by ICP-AES. The superiority of the catalyst was attributed to the metal support interaction (MSI) between metallic palladium and pyridinic-N doped carbon to acquire excellent catalytic activity and changing the reducing nature of NaBH4 towards nitro functionality. The MSI between pyridinic-N dopant on the carbon structure and PdNPs produces highly active sites for catalytic performance under mild conditions.

A two-step one pot, experimentally simple protocol, based on readily available and inexpensive reagents allowed the conversion of nitro-arenes directly to N-aryl amides. A selective synthesis of hydrazoarene from nitroarene and its application are reported. Using polystyrene (PS) resins as solid supports for Au nanoparticles (AuNPs), polystyrene-supported Au nanoparticles (AuNPs@PS) were synthesized and characterized. In the presence of AuNPs@PS (1.0 mol %) as a catalyst, nitroarenes afforded corresponding hydrazoarenes (up to 99%) with high selectivity (up to 100%) under mild reaction conditions (NaBH4, 50% aq. EtOH, and room temperature). Depending on the reaction conditions (the amount of NaBH4, the substituent of nitroarenes, and the sequential addition of HCl), nitroarenes were converted to corresponding azoarenes (up to 95%), aminoarenes (up to 99%), and 4,4′-diaminobiaryls (up to 99%). Our easily recyclable catalytic system using a solid-phase reaction vessel provides an attractive synthetic method in an eco-friendly and sustainable manner.

A desirable alternative to organic solvents, very cheap, non-toxic and biodegradable deep eutectic solvent (DES) based on choline chloride and urea was introduced for 2,4,6-trichloro triazine mediated amide bond formation between carboxylic acids and amines at room temperature. A series of novel 2, 5-disubstituted 1, 3, 4-Oxadiazole derivatives as a potential anti-inflammatory, and antioxidant agent were synthesized via cyclisation. Hydrazide molecule treated with substituted acids in the presence of phosphorus oxychloride (POCl3) as an efficient reagent as well as solvent by conventional method with shorter reaction time and excellent yield. The newly synthesized 1, 3, 4-oxadiazole derivatives exhibited excellent to good anti-inflammatory and anti-oxidant activities compaired to the standard drugs. Molecular docking study on the crucial anti-inflammatory target-cyclooxygenase-2 (COX-2) revealed the ability of the scaffold to correctly recognize the active site and achieve significant bonded and non-bonded interactions with key residues therein. This study could identify potential compounds which can be pertinent starting points for structure-based drug design to obtain newer anti-inflammatory agents.

Currently, due to the HIV latency mechanism, the search continues for effective drugs to combat this issue and provide a cure for AIDS. Gnidimacrin activates latent HIV-1 replication and inhibits HIV-1 infection at picomolar concentrations. This natural diterpene was able to markedly reduce the latent HIV-1 DNA level and the frequency of latently infected cells. Therefore, gnidimacrin is an excellent lead compound, and its anti-HIV potential merits further investigation. Twenty-nine modified gnidimacrin derivatives were synthesized and evaluated in assays for HIV replication and latency activation to establish which molecular structures must be maintained and which can tolerate changes that may be needed for better pharmacological properties. The results indicated that hydroxyl substituents at C-5 and C-20 are essential, while derivatives modified at 3-OH with aromatic esters retain anti-HIV replication and latent activation activities. The half-lives of the potent GM derivatives are over 20 h, which implies that they are stable in the plasm even though they contain ester linkages. The established structure activity relationship should be useful in the development of gnidimacrin or structurally related compounds as clinical trial candidates.
