This paper reports the tandem reaction strategy of the Passerini/Wittig reaction based on the in situ capture of isocyanides. According to this strategy, plenty of isocyanides have been synthesized, which is immediately used for the tandem reaction of Passerini/Wittig reaction in one pot. Compared to the previous work, this strategy avoids the separation, purification, and storage of isocyanides, which prominently solves the problems of isocyanide-based multicomponent reaction such as: (a) The environmentally unfriendly (strong foul odor), (a) the labile of isocyanides, (c) high toxicity of isocyanides. In the meantime, in order to expand the application scope of our strategy, 1H-isochromenes and 3H-2-benzoxepin-1-ones have also been synthesized, which undergoes four-step transformations in one-pot. In addition, a relatively credible reaction mechanism has also been proposed, based on a series of control experiments. Furthermore, preliminary testing was performed on biological activity of some obtained compounds; These results showed that the synthesized compounds exhibited certain activity over P. digitatum and P. italicum. Caseinolytic protease P (ClpP) is considered as a promising target for the treatment of Staphylococcus aureus infections. In an unbiased screen of 2632 molecules, a peptidomimetic boronate, MLN9708, was found to be a potent suppressor of SaClpP function. A time-saving and cost-efficient strategy integrating in silico position scanning, multistep miniaturized synthesis, and bioactivity testing was deployed for optimization of this hit compound and led to fast exploration of structure-activity relationships. Five of 150 compounds from the miniaturized synthesis exhibited improved inhibitory activity. Compound 43Hf was the most active inhibitor and showed reversible covalent binding to SaClpP while did not destabilize the tetradecameric structure of SaClpP. The crystal structure of 43Hf-SaClpP complex provided mechanistic insight into the covalent binding mode of peptidomimetic boronate and SaClpP. Furthermore, 43Hf could bind endogenous ClpP in S. aureus cells and exhibited significant efficacy in attenuating S. aureus virulence in vitro and in vivo.

We describe a methodology for the amidation of carboxylic acids by generating phosphonium salts in situ from N-chlorophthalimide and triphenylphosphine. Aliphatic, benzylic, and aromatic carboxylic acids can be transformed into their amide counter parts using primary and secondary amines. This functional group interconversion is achieved at room temperature in good to excellent yields. Mechanistic work shows the in situ formation of chloro- and imido-phosphonium salts that react as activating agents for carboxylic acids and generate an acyloxyphosphonium species.

In this study, a mild and efficient method is developed for the silylation of diverse functional groups using CeO2 nanoparticles (n-CeO2) as solid catalysts with hexamethyldisilazane (HMDS) as silylating agent at room temperature. The TEMPO-catalyzed oxidation of alcohols to aldehydes has emerged to one of the most widely applied methodologies for such transformations. Advantages are the utilization of sodium hypochlorite, a component of household bleach, as an oxidation agent and the use of water as a co-solvent. However, a major drawback of this method is the often occurring strict limitation to use dichloromethane as an organic solvent in a biphasic reaction medium with water. Previous studies show that dichloromethane cannot easily be substituted because a decrease of selectivity or inhibition of the reaction is observed by using alternative organic solvents. Thus, up to now, only a few examples are known in which after a tedious optimization of the reaction dichloromethane could be replaced. In order to overcome the current limitations, we were interested in finding a TEMPO-oxidation method in alternative organic solvents, which is applicable for various alcohol oxidations. As a result, we found a method for N-oxyl radical-catalyzed oxidation using sodium hypochlorite as an oxidation agent in nitriles as an organic solvent component instead of dichloromethane. Besides the oxidation of aromatic primary alcohols also aliphatic primary alcohols, secondary alcohols as well as dialcohols were successfully converted when using this method, showing high selectivity towards the carbonyl compound and low amounts of the acid side-product.
