Aryl- or heteroaryl-based hydrazinylphthalazine derivatives as new potential antitrypanosomal agents

A series of twenty phthalazinyl-hydrazones were synthesized and tested as potential anti-Trypanosoma cruzi agents. The phthalazines containing 5-nitroheteroaryl moiety 3l and 3m displayed an excellent in vitro antitrypanosomal profile, exhibiting low micromolar EC50 values against proliferative epimastigote of T. cruzi and minimal toxicity toward Vero cells. These derivatives were more potent than the reference drug benznidazole against the epimastigote stage of the parasite. Structure-property analysis indicates that the highly conjugated 5-nitroheteroaryl moiety connected to the phthalazin scaffold play an important role in the antichagasic activity of these phthalazines. The decrease on the mitochondrial dehydrogenase activity and significant ROS production found for the parasites treated with 3l and 3m suggest that both nitro-derivatives can act through an oxidative stress mechanism. Tel-Cu-NPs Catalyst: Synthesis of Naphtho[2,3-g]phthalazine Derivatives as Potential Inhibiters of Tyrosinase Enzymes and Their Investigation in Kinetic, Molecular Docking, and Cytotoxicity Studies

Novel one-pot synthesis naphtho[2,3-g]phthalazine (1a-1k) of Mannich base derivatives can be achieved via grindstone chemistry using a Tel-Cu-NPs (telmisartan-copper nanoparticles) catalyst. This method offers efficient mild reaction conditions and high yields. Tyrosinase inhibitory activity was evaluated for all synthesized compounds, along with analysis of kinetic behavior and molecular docking studies. The synthesized compound, 1c was (IC50 = 11.5 mu M) more active than kojic acid (IC50 = 78.0 mu M). Lineweaver Burk plots were used to analyze the kinetic behavior of the most active compound 1c, it was reversible and competitive behavior. Compound 1c and kojic acid occurred in the presence of 2-hydroxyketone, which has the same inhibitory mechanism. The molecular docking of compound 1c and the control kojic acid were docked against 2Y9X protein via the Schrodinger Suite. The compound 1c showed a respectable dock score (-5.6 kcal/mol) compared to kojic acid with a dock score of (-5.2 kcal/mol) in the 2Y9X protein. Cytotoxicity activity was also evaluated by using HepG2 (liver), MCF-7 (breast), and HeLa (cervical) cancer cell lines, and high activity for 1c (GI(50) = 0.01, 0.03, and 0.04 mu M, respectively) against all cell lines was found compared to standard and other compounds. Therefore, this study succeeded in testing a few promising molecules as potential antityrosinase agents.

Synthesis and vasorelaxant and antiplatelet activities of a new series of (4-Benzylphthalazin-1-ylamino)alcohol derivatives

A new series of phthalazine derivatives was synthesized by reaction of phthalic anhydride and different substituted phenylacetic acids to yield the benzyliden-3H-isobenzofuran-1-one intermediates 2a-d. Synthesis and positive inotropic evaluation of [1,2,4]triazolo [3,4-a]phthalazine and tetrazolo[5,1-a]phthalazine derivatives bearing substituted piperazine moieties

Cu-II Anchored onto the Magnetic Talc: A New Magnetic Nanostructured Catalyst for the One-Pot Gram-Scale Synthesis of 1H-Pyrazolo[1,2-b]phthalazine-5,10-dione Derivatives

In the present study Cu(II)anchored onto the magnetic talc (gamma-Fe2O3/ talc /Cu(II)NPs) was successfully synthesized and characterized by FT-IR, XRD, TEM, FE-SEM, EDX, VSM, and ICP-OES techniques. Structural characterizations reveal that the gamma-Fe2O3/talc/Cu(II)NPs are superparamagnetic in nature, structured as a composite with average diameter of about 19-31 nm. The prepared nanostructured catalyst efficiently catalyzed the preparation of 1H-pyrazolo[1,2-b] phthalazine-5,10-diones at the gram scale without the use of the toxic organic solvents. The magnetically separable and environmentally friendly nanostructured catalyst remains quite stable during reaction conditions and reused for at least five recycle runs.

An Efficient and High-yielding One-pot Synthesis of 1H-pyrazolo[1,2-b]phthalazine-5,10-diones Catalyzed by Sodium Hydrogen Carbonate under Solvent-free Conditions

Ionic liquid coated sulfonated carbon@titania composites for the one-pot synthesis of indeno[1,2-b]indole-9,10-diones and 1H-pyrazolo[1,2-b]phthalazine-5,10-diones in aqueous media

A highly active and stable ionic liquid based novel solid Bronsted acid catalyst (C@TiO2-SO3H-ILs) was prepared by coating a long chain ionic liquid onto sulfonated carbon@titania composites. The ionic liquid coating resulted in efficient stability and activity of C@TiO2-SO3H-ILs under aqueous conditions by creating a hydrophobic environment on the catalyst surface. The catalytic activity of the newly designed catalyst was tested in the one-pot multicomponent synthesis of indeno[1,2-b]indole-9,10-diones and 1H-pyrazolo[1,2-b]phthalazine-5,10-diones in aqueous media. The structure of the most active catalyst, C@TiO2-SO3H-IL1, before and after the coating with the ionic liquid was investigated by FTIR, TGA, XRD, CHNS, EDX, SEM and HRTEM. The catalyst allows easy recovery and excellent reusability for up to five catalytic runs without considerable loss of activity.

Phthalazine-trione as a blue-green light-emitting moiety: crystal structures, photoluminescence and theoretical calculations

Phthalazine (2,3-diazanaphthalene) and phthalhydrazide (2,3-dihydro-1,4-phthalazinedione) compounds are heterocycles with several biological properties. Most recently, analogous phthalazine-triones also emerged as drug candidates. Herein, we introduced a phthalazine-trione moiety as a promising fluorophore in the blue and green spectral regions. This desired optical property was rationalized based on their crystal structure and time-dependent density functional theory (TD-DFT) calculations. Under ultraviolet (UV) excitation (ca. 360 nm), two light emission maxima at ca. 460 nm and 480 nm were observed for ten phthalazine-triones, regardless of the substitution pattern. This conservation in the light-emitting property was rationalized by our time-dependent DFT calculations for the three phthalazine-triones whose crystal geometries were also determined in this study (4-chlorophenyl, 3-methoxyphenyl and 4-nitrophenyl derivatives) and for those available in literature (4-fluorophenyl and 4-bromophenyl derivatives). Electronic transitions with the largest oscillator strengths were near 360 nm and were in excellent agreement with the experimental excitation energy (UV-visible and photoluminescence spectra). Lifetime values of selected samples could also be determined, which were between 1.59 ns and 3.21 ns. In all compounds, the Frontier molecular orbitals involved in these excitations were distributed over the phthalazine-trione moiety and were not localized on the changeable substituent.

By utilizing the hydrothermal in situ acylation of organic acids with N2H4, three acylhydrazidate-coordinated compounds [mn(L1)(2)(H2O)(2)] (L1 = 2,3-quinolinedicarboxylhydrazidate; HL1 = 2,3-dihydropyridazino[4,5-b]quinoline-1,4-dione) 1, [Mn-2(ox)(L-2)(2)(H2O)(6)]center dot 2H(2)O (L2 = benzimidazolate-5,6-dicarboxylhydrazide; HL2 = 6,7-dihydro-1H-imidazo[4,5-g]phthalazine-5,8-dione; ox = oxalate) 2, and [Cd(HL3)(bpy)] (L3 = 4,5-di(3′-carboxylphenyl)phthalhydrazidate; H(3)L3 = 6,7-dihydro-1H-imidazo[4,5-g]phthalazine-5,8-dione; bpy = 2,2′-bipyridine) 3, as well as two acylhydrazide molecules L4 (L4 = oxepino[2,3,4-de:7,6,5-d’e’]diphthalazine-4, 10(5H,9H)-dione) 4 and L5 (L5 = 4,5-dibromophthalhydrazide; L5 = 6,7-dibromo-2,3-dihydrophthalazine-1, 4-dione) 5 were obtained. X-ray single-crystal diffraction analysis reveals that (i) I only possesses a mononuclear structure, but it self-assembles into a 2-D supramolecular network via the N-hydrazine-H center dot center dot center dot N-hydrazine and Ow-H center dot center dot center dot O-hydroxylimino interactions; (ii) 2 exhibits a dinuclear structure. Ox acts as the linker, while L2 just serves as a terminal ligand; (iii) In 3,13 acts as a 3-connected node to propagate the 7-coordinated Cd2+ centers into a 1-D double-chain structure; (iv) 4 is a special acylhydrazide molecule. Two-OH groups for the intermediates 3,3′-biphthalhydrazide further lose one water molecule to form 4; (v) 5 is a common monoacylhydrazide molecule. Via the N-hydrazine-H center dot center dot center dot O-hydrazine, O-hydroxylimino-H center dot center dot center dot O-acylamino and the pi center dot center dot center dot pi interactions, it self-assembles into a 2-D supramolecular network.
