A survey of existing stationary phases classified by the United States Pharmacopeia reveals that 120 groups of chromatographic supports mostly utilize silica-silane chemistry, polymeric materials along with some niche metal oxides. In this work, the synthesis and characterization of transition-metal free geopolymers as a new class of stationary phases for hydrophilic interaction liquid chromatography and normal phase separations is reported. The geopolymers were synthesized by reaction of synthetic aluminosilicate with potassium silicate (fumed silica dissolved in KOH) in a water-in-oil emulsion. For comparative purposes of peak shapes, a geopolymer from natural metakaolin was also synthesized. The geopolymers were examined by X-ray diffraction, energy dispersive spectroscopy, laser diffraction, and N-2-adsorption isotherms. This two-step approach gives spherical microparticles with surface area and pore size comparable to silica phases (150 m(2)/g and 120 angstrom respectively). Both synthetic aluminosilicates based and natural metakaolin based geopolymers occupy a unique spot in the HILIC selectivity chart when compared to 35 HILIC phases. An additional promising feature of geopolymers is high pH and temperature stability which are used to tune selectivity for small polar analytes. High pH separations are shown with carboxylic acids. Geopolymers also show mixed mode behavior in retention with ion-exchange properties in purely aqueous mobile phases. The separation of derivatized sugars is demonstrated and compared with porous graphitic carbon (Hypercarb (TM)) as another pH-stable stationary phase. (C) 2019 Elsevier B.V. All rights reserved.

The cleavage of O-benzyl and N-benzyl protecting groups was performed using an uncommonly low palladium loading (0.02-0.3 mol%; TON up to 5000) in a relatively short reaction time. The approach was used for a variety of substrates including pharmaceutically important precursors, and gram-scale deprotection reaction was shown. Transfer conditions together with easy-to-make Pd/C catalyst are the key features of this debenzylation scheme.

The present report describes an organocatalytic cascade reaction between 2-alkylidene benzo[b]thiophenone derivatives and enones in the presence of the Cinchona alkaloid amine. Spirobenzothiophenonic cyclohexane derivatives containing three stereocenters were prepared via one-step synthesis in yields ranging from 88 to 96% and in enantioselectivities (enantiomeric excess (ee)) ranging from 85 to 97%, with diastereoselectivities of approximately 14/2/1. Therefore, this method provides an efficient route for the synthesis of a new class of optically active 2-spirobenzothiophenones.

We describe here our investigation of the asymmetric Diels-Alder reaction of chiral 3-(acyloxy)acryloyl oxazolidinones as dienophiles in various Lewis-acid promoted reactions with cyclopentadiene. The resulting highly functionalized cycloadducts are useful intermediates for the synthesis, particularly for the optically active synthesis of 6-5-5 tricyclic hexahydro-4H-3,5-methanofuro[2,3-b]pyranol (3) with five contiguous chiral centers. This stereochemically defined crown-like heterocyclic derivative is an important high affinity ligand for a variety of highly potent HIV-1 protease inhibitors. Among the various dienophiles and Lewis acid-mediated reactions surveyed, 3-(4-methoxybenzoyl)acryloyl oxazolidinone as the dienophile and diethylaluminum chloride as the Lewis-acid provided the desired endo product with excellent diastereoselectivity. The cycloaddition was carried out in multi-gram scale and the cycloadduct was efficiently converted to alcohol 3 with high enantiomeric purity. The optically active ligand was then transformed into potent HIV-1 protease inhibitor 2.

In this study, a mild and efficient method is developed for the silylation of diverse functional groups using CeO2 nanoparticles (n-CeO2) as solid catalysts with hexamethyldisilazane (HMDS) as silylating agent at room temperature. Alcohols, phenols and acids are silylated to their respective silyl derivatives with faster reaction rate while amines and thiols required relatively longer reaction time. Moreover, the solid catalyst is easily be separated from the reaction mixture and recycled more than five times without any obvious decay in its activity. Powder X-ray diffraction (XRD), transmission electron microscope (TEM), UV-vis diffuse reflectance spectra (UV-DRS) and Raman analyses revealed identical structural integrity, particle size, absorption edge and valence state for the reused solid compared to the fresh solid catalyst.

A novel structural series of tetrahydroisoquinoline-based compounds that incorporate the diaryl urea moiety was designed, synthesized, and biologically evaluated as suppressors of VEFGR-2 signaling. As a consequence, compounds 9k and 9s exhibited comparable or superior cytotoxic activity to that of gefitinib against the tested three cell lines, including A549, MCF-7, and PC-3. Importantly, both of them downregulated the expression of VEGFR-2, and inhibited VEGFR-2 phosphorylation at the concentration of 0.5 or 1.0 mu mol/ l. Computational approaches have recently gained popularity in the field of read-across to automatically fill data-gaps for untested chemicals. Previously, we developed the generalized read-across (GenRA) tool, which utilizes in vitro bioactivity data in conjunction with chemical descriptor information to derive local validity domains to predict hazards observed in in vivo toxicity studies. Here, we modified GenRA to quantitatively predict point of departure (POD) values obtained from US EPA’s Toxicity Reference Database (ToxRefDB) version 2.0. To evaluate GenRA predictions, we first aggregated oral Lowest Observed Adverse Effect Levels (LOAEL) for 1,014 chemicals by systemic, developmental, reproductive, and cholinesterase effects. The mean LOAEL values for each chemical were converted to log molar equivalents. Applying GenRA to all chemicals with a minimum Jaccard similarity threshold of 0.05 for Morgan fingerprints and a maximum of 10 nearest neighbors predicted systemic, developmental, reproductive, and cholinesterase inhibition min aggregated LOAEL values with R-2 values of 0.23, 0.22, 0.14, and 0.43, respectively. However, when evaluating GenRA locally to clusters of structurally-similar chemicals (containing 2 to 362 chemicals), average R-2 values for systemic, developmental, reproductive, and cholinesterase LOAEL predictions improved to 0.73, 0.66, 0.60 and 0.79, respectively. Our findings highlight the complexity of the chemical-toxicity landscape and the importance of identifying local domains where GenRA can be used most effectively for predicting PODs.
