In the present work, MnO2@Mg-Al layered double hydroxide (LDH) has been prepared via a simple coprecipitation method and characterized successfully. Two products are formed from benzyl alcohol oxidation over MnO2@Mg-Al layered double hydroxide (LDH): benzaldehyde and benzoic acid. This oxidation was carried out with three different parameters: oxidant, solvent, and temperature. In addition, MnO2@Mg-Al LDHs could be easily recovered and reused for five times without any loss of activity.

An organocatalytic asymmetric enantioselective domino oxa-Michael-Mannich-[1,3]-amino rearrangement reaction of N-tosylsalicylimines with a wide range of alpha,beta-unsaturated aldehydes utilizing diarylprolinol silyl ether catalysis is described. The catalytic reactions proceed with excellent enantioselectivity (up to 99% ee) to produce the corresponding chair N-tosylimines-chromenes with a yield of up to 99%, tolerating a range of functional groups. This methodology offers a new method with great potential to further extend the synthetic power and versatility of chiral aminocatalysis.

This paper reports the tandem reaction strategy of the Passerini/Wittig reaction based on the in situ capture of isocyanides. According to this strategy, plenty of isocyanides have been synthesized, which is immediately used for the tandem reaction of Passerini/Wittig reaction in one pot. Compared to the previous work, this strategy avoids the separation, purification, and storage of isocyanides, which prominently solves the problems of isocyanide-based multicomponent reaction such as: (a) The environmentally unfriendly (strong foul odor), (a) the labile of isocyanides, (c) high toxicity of isocyanides. In the meantime, in order to expand the application scope of our strategy, 1H-isochromenes and 3H-2-benzoxepin-1-ones have also been synthesized, which undergoes four-step transformations in one-pot. In addition, a relatively credible reaction mechanism has also been proposed, based on a series of control experiments. Furthermore, preliminary testing was performed on biological activity of some obtained compounds; These results showed that the synthesized compounds exhibited certain activity over P. digitatum and P. italicum. We conducted a structure-activity relationship study to explore simplified analogues of the itraconazole (ITZ) scaffold for their ability to inhibit the hedgehog (Hh) signaling pathway. These analogues were based on exploring the effects of chemical modifications to the linker and triazolone/side chain region of ITZ. Analogue 11 was identified as the most potent compound in our first generation, with an IC50 value of 81 nM in a murine Hh-dependent basal cell carcinoma. Metabolic identification studies led us to identify truncated piperazine (26) as the major metabolite in human liver microsomes (HLMs) and an improved Hh pathway inhibitor (IC50 = 22 nM). This work verifies that continued truncation of the ITZ scaffold is a practical method to maintain potent anti-Hh activity while also reducing the molecular weight for the ITZ scaffold and achieving improved pharmacokinetic properties.

In this work, the hydroboration reactions of alkynes were performed without metal catalysts using concomitant microwave irradiation that allowed a significantly reduced reaction time. The present work describes the preparation of two ionic liquid and carboxyl acid silane reagents via photo-initiated thiol-ene click chemistry that have been bonded to silica to afford two mixed-mode stationary phases (Sil-C4Im-C9Co and Sil-C9Im-C4Co). The two stationary phases provided satisfactory retention repeatability and efficiencies. The influence of acetonitrile content, salt concentration and pH of the mobile phase was investigated to clarify the retention properties of the prepared stationary phases. The results showed that the prepared Sil-C4Im-C9Co and Sil-C9Im-C4Co undergo multiple interactions with solutes under different chromatographic conditions. The retention mechanisms were further studied by the linear energy solvation relationship and Van’t Hoff plots. Finally, the stationary phases were employed to separate hydrophobic solutes (alkylbenzenes and polycyclic aromatic hydrocarbons) under reversed phase liquid chromatography (RPLC) mode, hydrophilic solutes (carboxylic acids, nucleosides and bases) under hydrophilic interaction liquid chromatography (HILIC) mode and inorganic anions under ion-exchange chromatography (IEC) mode, providing excellent performance and varying selectivity when compared with a commercial column. The bonding method in this work is feasible and the prepared stationary phases are promising when employed in RPLC/HILIC/IEC mixed-mode chromatography applications. (C) 2019 Elsevier B.V. A quick assembly of tri, tetra and penta cyclic pyrido/isoquinolo-quinoxilines are thus achieved from readily available linear substrates. The reaction is shown to be very general by accommodating a large variety of substrates in the transformation.
