New chiral regioisomeric gamma-hydroxyphosphine ligands were synthesized from alpha-pinene. The key transformation was the thermal [2,3]-sigmatropic rearrangement of allyldiphenylphosphinites, obtained from (1R,2R,4S,5R)-3-methyleneneoisoverbanol and (1R,2R,3R,5R)-4-methyleneneoisopinocampheol, to allylphosphine oxides. Hydroxy groups were introduced stereoselectively through a hydroboration-oxidation reaction proceeding from the less hindered site providing a trans relationship between the hydroxy and the phosphine substituents.

In this communication, we disclose the first example of metal-free and site-selective radical fluorination of readily available tertiary alkyl ethers, enabled by synergistic photocatalysis and organocatalysis. This catalytic combination allows for exclusive fluorination of tertiary C-O bonds under mild conditions even in the presence of competing reaction sites. The excellent functional group tolerance affords valuable access to sterically hindered alkyl fluorides through late-stage modification of complex molecules. The successful use of tertiary alkyl ethers in radical fluorination enhances the structural diversity of aliphatic fluorides that can be derived from naturally abundant alcohols.

Computational approaches have recently gained popularity in the field of read-across to automatically fill data-gaps for untested chemicals. Previously, we developed the generalized read-across (GenRA) tool, which utilizes in vitro bioactivity data in conjunction with chemical descriptor information to derive local validity domains to predict hazards observed in in vivo toxicity studies. Here, we modified GenRA to quantitatively predict point of departure (POD) values obtained from US EPA’s Toxicity Reference Database (ToxRefDB) version 2.0. To evaluate GenRA predictions, we first aggregated oral Lowest Observed Adverse Effect Levels (LOAEL) for 1,014 chemicals by systemic, developmental, reproductive, and cholinesterase effects. The mean LOAEL values for each chemical were converted to log molar equivalents. Applying GenRA to all chemicals with a minimum Jaccard similarity threshold of 0.05 for Morgan fingerprints and a maximum of 10 nearest neighbors predicted systemic, developmental, reproductive, and cholinesterase inhibition min aggregated LOAEL values with R-2 values of 0.23, 0.22, 0.14, and 0.43, respectively. However, when evaluating GenRA locally to clusters of structurally-similar chemicals (containing 2 to 362 chemicals), average R-2 values for systemic, developmental, reproductive, and cholinesterase LOAEL predictions improved to 0.73, 0.66, 0.60 and 0.79, respectively. Our findings highlight the complexity of the chemical-toxicity landscape and the importance of identifying local domains where GenRA can be used most effectively for predicting PODs.

The present work describes the preparation of two ionic liquid and carboxyl acid silane reagents via photo-initiated thiol-ene click chemistry that have been bonded to silica to afford two mixed-mode stationary phases (Sil-C4Im-C9Co and Sil-C9Im-C4Co). The two stationary phases provided satisfactory retention repeatability and efficiencies. The influence of acetonitrile content, salt concentration and pH of the mobile phase was investigated to clarify the retention properties of the prepared stationary phases. The results showed that the prepared Sil-C4Im-C9Co and Sil-C9Im-C4Co undergo multiple interactions with solutes under different chromatographic conditions. The retention mechanisms were further studied by the linear energy solvation relationship and Van’t Hoff plots. Finally, the stationary phases were employed to separate hydrophobic solutes (alkylbenzenes and polycyclic aromatic hydrocarbons) under reversed phase liquid chromatography (RPLC) mode, hydrophilic solutes (carboxylic acids, nucleosides and bases) under hydrophilic interaction liquid chromatography (HILIC) mode and inorganic anions under ion-exchange chromatography (IEC) mode, providing excellent performance and varying selectivity when compared with a commercial column. The bonding method in this work is feasible and the prepared stationary phases are promising when employed in RPLC/HILIC/IEC mixed-mode chromatography applications. (C) 2019 Elsevier B.V. A straightforward synthesis of acylating reagents such as Weinreb and MAP amides from aromatic, aliphatic carboxylic acids, and amino acids using PPh3/NBS combination is described. A chemo-selective modification of the carboxylic acid group into Weinreb amide in the presence of more reactive aldehydes and ketones is presented. All reactions were performed at ambient temperature under air using undried commercial grade solvent. Furthermore, the present methodology could be performed at a gram scale under inert-free reaction conditions. In addition, 7-azaindoline amide auxiliary (used for catalytic asymmetric aldol- and Mannich-type reactions), which behaves like Weinreb amide is also synthesized under similar reaction conditions.

A library of 1,2,4-oxadiazole functionalized quinoline derivatives (13a-j) were synthesized and their structures were confirmed by H-1 NMR, (CNMR)-C-13 and Mass Spectral analysis. Further, these compounds were evaluated for their anticancer activity against four human cancer cell lines, namely MCF-7 (breast), A549 (lung), DU-145 (prostate) and MDA MB-231 (breast) using Etoposide as the positive control. Most of these derivatives exhibited more potent activity towards the four cancer cell lines compared to Etoposide. Amongst all the compounds tested, compounds 13b, 13c, 13h, 13i and 13j exhibited promising activity. Further of these compounds 13b, 13i and 13j exhibited excellent activity, when compared with Etoposide.

The insect beta-N-acetylhexosaminidase OfHex1 from Ostrinia furnacalis (one of the most destructive agricultural pests) has been considered as a promising pesticide target. In this study, a series of novel and readily available ureido thioglycosides were designed and synthesized based on the catalytic mechanism and the co-crystal structures of OfHex1 with substrates. After evaluation via enzyme inhibition experiments, thioglycosides 11c and 15k were found to have inhibitory activities against OfHex1 with the K-i values of 25.6 mu M and 53.8 mu M, respectively. In addition, all these ureido thioglycosides exhibited high selectivity toward OfHex1 over hOGA and HsHexB (K-i > 100 mu M). Furthermore, to investigate the inhibitory mechanism, the possible binding modes of 11c and 15k with OfHex1 were deduced based on molecular docking analysis. This work may provide useful structural starting points for further rational design of potent inhibitors of OfHex1.
