1H-pyrazolo[1,2-b]phthalazine-5,10-diones (5) and 2H-indazolo[2,1-b]phthalazine-1,6,11(13H)-triones (7) have been synthesized by one pot, four-component method. To synthesise them phthalic anhydride (1), hydrazine hydrate (2), benzaldehydes (3) and malononitrile (4a) /ethyl cynoacetate (4b) /dimedone (6) in refluxing water for 1-1.5 h in the presence of InCl3 as a catalyst yielding 75-85%. These reactions have an easy workup, provides excellent yields, and uses water as the solvent which is considered to be relatively environmentally benign. The structures of the products were established from their spectral properties IR, H-1-NMR, C-13-NMR & Mass spectroscopy.

Phosphoramidates as Transient Precursors of Nitrogen-Centered Radical Under Visible-Light Irradiation: Application to the Synthesis of Phthalazine Derivatives

Phosphoramidates are for the first time presented as efficient N-Centered Radical (NCR) precursors under visible-light irradiation. More precisely among this class of phosphorus-derived compounds, we studied the radical reactivity of phosphonohydrazones, under mild reaction conditions, which allowed the synthesis of a wide and diversified library of the scarcely reported phthalazine scaffold. Mechanistic investigations confirmed the formation of a NCR from these brand-new phosphonohydrazones (derivatived from phosphoramidates), which were further engaged in an intramolecular 6-exo-dig cyclization to provide phthalazines. Compared to other pre-activated moieties, the phosphoramidate group is self-immolative, thus enhancing its attractiveness for the C-N bond formation.

Synthesis, characterization, X-ray structure, optical properties and theoretical calculations of condensed phthalazines

Two condensed phthalazine compounds were synthesized and characterized by FT-IR, UV-Vis, NMR spectroscopic studies. Single crystal XRD and molecular orbital calculations. Optimized geometrical structures were computed with RB3LYP method with the 6-31G(p, d) basis set. The geometrical parameters of both compounds obtained from Single Crystal XRD were found to be in accord with the calculated (DFT) values. The electronic contribution was measured using the third harmonic generation technique on thin films at 1064 nm for both compounds. Also, the values of dipole moment , the average polarizability , and the first static hyperpolarizability were computed. The theoretical and experimental results confirm the NLO behavior of both compounds.

In this paper, C-N bond formation between 2,3-dihydrophthalazine-1(4H),4-dione (phthalhydrazide) and alpha,beta-unsaturated esters was investigated and a new series of phthalazine derivatives was synthesized using an efficient and simple method under solvent-free conditions. An aza-Michael addition of phthalhydrazide to both acrylic and fumaric esters led to N-monosubstituted phthalhydrazides (as mono-Michael adduct) in the presence of tetrabutylammonium bromide as a high polar media, and 1,4-diaza-bicyclo[2,2,2] octane as an available organic base. In this reaction, the N1,N2-bis-Michael adduct was not observed at all. Also, reactions were performed at 90 degrees C and yields of products were good to excellent.

Synthesis of Pyrazolo[1,2-b]phthalazine-5,10-diones Catalyzed by Poly(ethylene glycol) Grafted 4-Dimethylaminopyridine Functionalized Ionic Liquid

A novel poly(ethylene glycol) grafted 4-dimethylaminopyridine functionalized ionic liquid ([DMAP-PEG(1000)-DIL][BF4]) was prepared and characterized by H-1 NMR, C-13 NMR, FT-IR and ESI-MS. In the presence of [DMAP-PEG(1000)-DIL][BE4], various substituted aromatic aldehyde could react with malononitrile (or ethyl cyanoacetate) and phthalhydrazide in water to obtain a series of pyrazolo[1,2-b]phthalazine-5,10-dione derivatives with 85%similar to 96% yields. The mild reaction conditions, high stable and recyclable ionic liquids, environmental friendliness are advantages of this methodology.

Application of O-sulfonic acid poly(4-vinylpyrrolidonium) chloride as an efficient polymer-supported catalyst in the rapid synthesis of 2H-indazolo[2,1-b]phthalazine-triones and Biginelli-like products

In this work, a series of 2H-indazolo[2,1-b]phthalazine-triones and Biginelli-like products were efficiently synthesized for the first time using O-Sulfonic acid poly(4-vinylpyrrolidonium) chloride ([PVP-SO3H]Cl) as an eco-friendly polymer-supported catalyst. At first, the catalyst preparation process was optimized based on the chlorosulfonic acid amount and the sulfonation time to obtain a catalyst with the maximum acidity content. The maximum acidity content for [PVP-SO3H]Cl was obtained to be 3.31 mmol g(-1). Then, the optimized [PVP-SO3H]Cl was used as a catalyst for the synthesis of a series of 2H-indazolo[2,1-b]phthalazine-triones and Biginelli-like products. It was observed that only 10 mg of the catalyst was enough to catalyze these reactions under mild conditions, and the corresponding products were obtained in high to excellent yields. Moreover, the catalyst could be repeatedly used for at least five times without a noticeable decrease in its catalytic activity.

These two dihydride complexes are shown, by NMR, to undergo both full N-heterocycle dissociation and a haptotropic shift, the rates of which are affected by both steric interactions and free ligand pK(a) values. The use of these complexes as catalysts in the transfer of polarisation from para-hydrogen to pyridazine and phthalazine via signal amplification by reversible exchange (SABRE) is explored. The possible future use of drugs which contain pyridazine and phthalazine motifs as in vivo or clinical magnetic resonance imaging probes is demonstrated; a range of NMR and phantom-based MRI measurements are reported.
