ONE-POT SYNTHESES OF 1H-PYRAZOLO[1,2-B]PHTHALAZINE-5,10-DIONES IN MOLTEN TBAB

One-pot, four component syntheses for the preparation of 1-H Pyrazolo[1,2-b]phthalazine-5,10-diones (5a-5h) have been achieved from phthaloyl dichloride (1), hydrazine hydrate (2), benzaldehydes (3) and malononitrile (4a) or ethyl cynoacetate (4b) in molten Tetrabutylammonium Bromide as medium at 100-105 degrees C for 20-30 min by in-situ generation of HCl as a catalyst. These reactions have an easy workup, provide excellent yields, and use TBAB as the reaction medium.

Novel urea-functionalized silica-coated magnetic core-shell Fe3-x Ti (x) O-4 nanoparticles were prepared and fully characterized by different methods such as Fourier transform infrared spectroscopy, energy-dispersive X-ray spectroscopy, field emission scanning electron microscopy, high-resolution transmission electron microscopy, vibrating sample magnetometer, X-ray diffraction, and thermogravimetric analyses. These magnetic nanoparticles have been explored as highly efficient and recoverable heterogeneous nanocatalyst for one-pot three-component reactions under mild conditions for the synthesis of diverse range of 5-oxo-dihydropyrano[3,2-c]chromenes known as coumarins and 1H-pyrazolo[1,2-b] phthalazine-5,10-diones in high yields. Simple isolation of the products, use of green solvent, and magnetically easy isolation and reusability of the catalyst with no significant loss of activity are the main advantages of the present method.

Synthesis and Antitumor Activity Evaluation of 3,6-Substituted-1,2,4-triazolo[3,4-a]phthalazine Derivatives

With the aim of obtaining potential antitumor candidates with more efficiency and more economic value. A series of 3,6-substituted-1,2,4-triazolo[3,4-a]phthalazine derivatives were synthesized. The target products were obtained via cyclization, chlorination, substitution, cyclization, substituted with phthalic anhydride used as the starting material. The structures of target products are confirmed by H-1 NMR, C-13 NMR, HRMS. A series of 3,6-substituted-1,2,4-triazolo[3,4-a] phthalazine derivatives was evaluated for anticancer activity on four human cancer cell lines including EC-9706, HeLa and MCF-7 by MTT assay. Among them, compound 5d was more cytotoxic against EC-9706 and HeLa cell lines, with IC50 values ranging from 3.9 to 4.5 mu mol.L-1, which are superior or comparable to 5-Fuorouracil. Flow cytometry analysis indicated that compound 5d induced the cellular early apoptosis and cell cycle arrest in G2/M phase in EC-9706.

Spectroscopic characterization and quantum chemical investigation of molecular structure and vibrational spectra of phthalazine-1(2H)-one

In this study, vibrational and electronic transition analysis of phthalazine-1(211)-one have been presented using experimental techniques FT-IR, FT-Raman and density functional theory (DFT) calculation. The structural properties of the molecule in the ground state have been calculated using DFT employing B3LYP/6-311++G(d,p) basis set. Optimized geometrical parameters have been interpreted and compared with the experimental values. The complete assignments have been performed on the basis of the experimental data and potential energy distribution (PED) of the vibrational modes. The calculated HOMO and LUMO energies and energy difference (Delta EHOMO-LUMO = 4.876 eV), confirm that charge transfers occur within the molecule. The stability of the molecule arising from hyperconjugative interactions and the charge delocalization has been analyzed using natural bond orbital’s analysis (NBO). The specific heat, Gibb’s free energy, and entropy of molecule have been calculated as a function of temperature by using statistical mechanics coupled with quantum chemical calculation. Observed vibrational wave numbers have been compared with calculated values, and found to be in agreement with experimental results. The study of dielectric properties like dielectric constant at microwave frequency, static dielectric constant and dielectric constant at optical frequency of Phthalazine-1(2H)-one have been determined. The dielectric relaxation studies provide information about the molecular structure and intermolecular interaction between phthalazine-1(2H)-one and alcohol mixture.

Design, synthesis, and biological evaluation of optimized phthalazine derivatives as hedgehog signaling pathway inhibitors

We report herein the design and synthesis of a series of optimized phthalazine compounds as novel hedgehog signaling pathway inhibitors. The 4-methylamino-piperidine moiety of Taladegib was replaced by different four, five or six-membered azacycle or azaspirocycle building blocks. The in vitro Gliluciferase assay results demonstrate that the scaffold hopping in this region afforded significant influences on Hh pathway inhibition. Pyrrolidin-3-amine moiety was found to be the best linker between pharmacophores phthalazine and fluorine substituted benzoyl group. Meanwhile the optimization of 1-methyl-1H-pyrazol by different aromatic rings was also investigated and the SAR was described. Many new derivatives were found to show potent Hh signaling inhibitory activity with nanomolar IC50 values. Among these compounds, compound 23b showed the highest inhibitory potency with an IC50 value of 0.17 nM, which was 35-fold more potent than the lead compound Taladegib and 23-fold more potent than the marketed drug Vismodegib. The selected compounds 23a and 23b also possess potent antitumor activities against medulloblastoma cells proliferation in vitro. The biodegradable catalyst was characterized by XRD, TEM, FT-IR, and energy dispersive X-ray. The surface area and pore diameter of the catalyst were determined by a nitrogen adsorption-desorption isotherm experiment. A highly efficient multicomponent heteroannulation protocol for the synthesis of a library of 1H-pyrazolo[1,2-b]phthalazine-5,10-dione derivatives has been developed by applying the solid acid catalyst. This synthesis was established to follow the group-assistant-purification chemistry process avoiding traditional chromatography. The aqueous reaction medium, easy recovery of the catalyst, and high yield of the products make the protocol attractive, sustainable, and economic. This work may not only lead to environmentally benign systems, but also will provide a new aspect of organic chemistry in water.
