A site-selective supported palladium nanoparticle catalyzed Suzuki-Miyaura cross-coupling reaction with heteroaryl esters and arylboronic acids as coupling partners was developed. This methodology provides a heterogeneous catalytic route for aryl ketone formation via C(acyl)-O bond activation of esters by successful suppression of the undesired decarbonylation phenomenon. The catalyst can be reused and shows high activity after eight cycles. A series of C-23 modified 23-hydroxybetulinic acid (HBA) derivatives were synthesized and evaluated for their antiproliferative activity against a panel of cancer cell lines (A2780, A375, B16, MCF-7 and HepG2). The biological screening results showed that most of the derivatives exhibited more potent antiproliferative activity than HBA, and compound 6e exhibited the most potent activity with IC50 values of 2.14 mu M, 2.89 mu M, and 3.97 mu M against A2780, B16, and MCF-7 cells, respectively. Further anticancer mechanism studies revealed that compound 6e induced the generation of intracellular reactive oxygen species (ROS) and reduction of mitochondrial membrane potential (MMP) of B16 cells in a dose-dependent manner. Moreover, western blot analysis indicated that compound 6e downregulated the expression of anti-apoptotic protein Bcl-2 and upregulated the expression of proapoptotic protein Bax, activation of caspase 3 to induce cell apoptosis. Meanwhile, compound 6e significantly inhibited the phosphorylation of MEK, ERK, and Akt without affecting the expression of MEK, ERK, and Akt. Furthermore, the in vivo anti-tumor activity of 6e was validated (tumor inhibitory ratio of 68.4% at the dose of 30 mg/kg) in mice with B16 melanoma. (C) 2019 Elsevier Masson SAS. Organocatalytic asymmetric Michael addition between 3-substituted oxindoles and enals catalyzed by chiral camphor sulfonyl hydrazines (CaSHs) has been developed. The HDAC inhibitor 4-tert-butyl-N-(4-(hydroxycarbamoyl)phenyl)benzamide (AES-350, 51) was identified as a promising preclinical candidate for the treatment of acute myeloid leukemia (AML), an aggressive malignancy with a meagre 24% 5-year survival rate. The effect of topology on the structure, self-assembly, and selective Fe3+ binding of delta-peptides has been investigated. A series of delta-peptides with an amino acid containing dihydropyrimidinone and o-, m-, and p-aminobenzoic acids have been designed to study the structure-function relationship. A new amino acid containing dihydropyrimidinone was synthesized by the Biginelli reaction of ethyl acetoacetate, urea, and o-nitrobenzaldehyde followed by reduction with iron powder and acetic acid. X-ray crystallography sheds some light on the conformations, self-assembly, and the diverse degrees of pi-pi stacking of adjacent delta-peptide molecules. Peptides with o- or m-aminobenzoic acid form eight-membered intramolecular hydrogen-bonded turn conformations and self-assemble through intermolecular hydrogen bonds between dihydropyrimidinone units to form a butterfly-like structure. However, the delta-peptide containing p-aminobenzoic acid forms a water-mediated cage-like structure. Irrespective of the presence of the same functional groups, only the delta-peptide with o-aminobenzoic acid can selectively bind Fe3+ in methanol as well as in water. The topology plays a crucial role in the selective Fe3+ ion binding by the delta-peptide.

A survey of existing stationary phases classified by the United States Pharmacopeia reveals that 120 groups of chromatographic supports mostly utilize silica-silane chemistry, polymeric materials along with some niche metal oxides. In this work, the synthesis and characterization of transition-metal free geopolymers as a new class of stationary phases for hydrophilic interaction liquid chromatography and normal phase separations is reported. The geopolymers were synthesized by reaction of synthetic aluminosilicate with potassium silicate (fumed silica dissolved in KOH) in a water-in-oil emulsion. For comparative purposes of peak shapes, a geopolymer from natural metakaolin was also synthesized. The geopolymers were examined by X-ray diffraction, energy dispersive spectroscopy, laser diffraction, and N-2-adsorption isotherms. This two-step approach gives spherical microparticles with surface area and pore size comparable to silica phases (150 m(2)/g and 120 angstrom respectively). Both synthetic aluminosilicates based and natural metakaolin based geopolymers occupy a unique spot in the HILIC selectivity chart when compared to 35 HILIC phases. An additional promising feature of geopolymers is high pH and temperature stability which are used to tune selectivity for small polar analytes. High pH separations are shown with carboxylic acids. Geopolymers also show mixed mode behavior in retention with ion-exchange properties in purely aqueous mobile phases. The separation of derivatized sugars is demonstrated and compared with porous graphitic carbon (Hypercarb (TM)) as another pH-stable stationary phase. (C) 2019 Elsevier B.V. All rights reserved.

A novel series of 2-(3-methyl-1,6-diphenyl-1H-pyrazolo[3,4-b]pyridin4-yl)-5-aryl-1,3,4-oxadiazoles (4a-4h) has been synthesized from corresponding hydrazones (3a-3h) and evaluated their antiplatelet aggregation effect induced by arachidonic acid and collagen. Spectral data and elemental evaluation were used to confirm the structure of the compounds while molecular docking against cyclooxygenase 1 and 2 (COX1 & COX2) and quantitative structure-activity relationship (QSAR) were performed in describing their antiplatelet potential. All synthesized compound exhibited more than 50% platelet aggregation inhibition against both arachidonic acid and collagen. Antiplatelet activities results showed that 4b and 4f compounds have highest % inhibition against arachidonic acid. High Egap and ionization potential values showed that the compound 4d, 4e and 4f were supposed to be more active and good electron donor while 4b, 4c, 4d, 4e, 4g and 4h might be more active due to more electrophilic sites. Interaction with more than one residues in the binding pocket of COX-1 in comparison with aspirin and ligand efficacy (LE) consequences showed that compounds have excellent action potential for COX-1. Computational evaluations are in good agreement with antiplatelet activities of the compounds. All compounds might be promising antiplatelet agents especially 4b, 4f and helpful in the synthesis of new drugs for the treatment of cardiovascular diseases (CVDs).
