Chemo selective reduction of a wide range of aromatic nitro compound has been performed by using inexpensive Zn powder and CuSO4 system in water medium at room temperature. This system has high tolerance to other highly reducible groups present in nitro substance along with high conversation and selectivity. This chemo-selective reduction also provides a facile root for the synthesis of other industrially important fine chemicals or biologically important compounds where other highly reducible groups are present in close proximity to the targeted nitro groups. Calcium dependent protein kinase 1 (CDPK1) is an essential Ser/Thr kinase that controls invasion and egress by the protozoan parasite Toxoplasma gondii. The Gly gatekeeper of CDPK1 makes it exquisitely sensitive to inhibition by small molecule 1H-pyrazolo[3,4-d]pyrimidine-4-amine (PP) compounds that are bulky ATP mimetics. Here we rationally designed, synthesized, and tested a series of novel PP analogs that were evaluated for inhibition of CDPK1 enzyme activity in vitro and parasite growth in cell culture. Optimal substitution on the PP scaffold included 2-pyridyl ethers directed into the hydrophobic pocket and small carbocyclic rings accessing the ribose-binding pocket. Further optimization of the series led to identification of the lead compound 3a that displayed excellent potency, selectivity, safety profile, and efficacy in vivo. The results of these studies provide a foundation for further work to optimize CDPK1 inhibitors for the treatment of acute and chronic toxoplasmosis.

A large family of phosphite-thioether/selenoether ligands has been easily prepared from accessible L-(+)-tartaric acid and D-(+)-mannitol and applied in the M-catalyzed (M=Ir, Rh) asymmetric hydrogenation of a broad number of substrates (46 in total). The catalytic activity of two types of L-prolinamide organocatalysts was investigated for asymmetric Michael addition reaction of cyclic/acyclic ketones with beta-nitrostyrens. L-Prolinamides bearing amino groups on phenyl ring worked well, though their catalytic efficiency as well as selectivity was found to be dependent upon the position of amine group to the amide bond. Organocatalyst having -NH2 group ortho to amide bond provided the best results. Substrate scope was also studied by performing the reaction of various beta-nitrostyrenes with ketones to afford the corresponding Michael adducts in excellent yields with very high diastereoselectivities and enantioselectivities in almost all cases.

The development of sub-nanometric metal particles (<1 nm) as advanced heterogeneous catalysts has received considerable interest due to their outstanding catalytic performance, while the synthesis and stabilization of sub-nanometric catalysts (SNCs) without using additional surface capping agents remains a challenge. Herein, we report the synthesis of novel three-dimensional pyrenyl graphdiyne (Pyr-GDY) ultrafine nanofibers (3-10 nm), which can serve as an ideal substrate for electroless deposition and stabilization of Pd SNCs through the terminal uncoupled acetenyl groups in Pyr-GDY, with an average Pd particle size of only 0.83 nm. The as-synthesized Pd/Pyr-GDY composite shows extremely high catalytic activities for the reduction of nitroarenes to arylamines and Suzuki coupling reactions, 300 and 25 times higher than those of commercial Pd/C, respectively. The outstanding catalytic performance can be ascribed to the sub-nanometric Pd particles with a clean surface, and the unique three-dimensional network structure of Pyr-GDY, being favorable for rapid mass transfer. Our result provides an ideal carbon material for electroless deposition and stabilization of other SNCs with a clean surface, which will display outstanding catalytic activity for various catalytic reactions. In this study, a mild and efficient method is developed for the silylation of diverse functional groups using CeO2 nanoparticles (n-CeO2) as solid catalysts with hexamethyldisilazane (HMDS) as silylating agent at room temperature. Alcohols, phenols and acids are silylated to their respective silyl derivatives with faster reaction rate while amines and thiols required relatively longer reaction time. Moreover, the solid catalyst is easily be separated from the reaction mixture and recycled more than five times without any obvious decay in its activity. Powder X-ray diffraction (XRD), transmission electron microscope (TEM), UV-vis diffuse reflectance spectra (UV-DRS) and Raman analyses revealed identical structural integrity, particle size, absorption edge and valence state for the reused solid compared to the fresh solid catalyst.

Carbohydrate-based low-molecular-weight gelators are useful and versatile compounds for the preparation of soft materials. Using N-acetyl-D-glucosamine as the starting material, we synthesized and characterized 15 glycolipids containing an amide with different ester functional groups. These include aliphatic derivatives with varying chain lengths and aromatic derivatives. Most of the hybrid amide-esters have molecular weights less than 500 D. These glycolipids were found to be effective gelators for several organic solvents, water, and aqueous solutions. Two efficient hydrogelators were also obtained at low concentrations. A few representative gels were characterized using optical microscopy, atomic force microscopy, and rheology to obtain information on their morphology and gel stability. Three gelators were also used to encapsulate naproxen sodium and toluidine blue. The sustained release of the drug from the gel to the aqueous phase was monitored by UV-vis spectroscopy. These gelators have structural flexibility that can be stimuli responsive. The esters can be hydrolyzed and several gels were converted to solutions under basic conditions. These rationally designed gelators could be utilized as stimuli-responsive smart materials with controlled release properties.

We describe here our investigation of the asymmetric Diels-Alder reaction of chiral 3-(acyloxy)acryloyl oxazolidinones as dienophiles in various Lewis-acid promoted reactions with cyclopentadiene. The resulting highly functionalized cycloadducts are useful intermediates for the synthesis, particularly for the optically active synthesis of 6-5-5 tricyclic hexahydro-4H-3,5-methanofuro[2,3-b]pyranol (3) with five contiguous chiral centers. This stereochemically defined crown-like heterocyclic derivative is an important high affinity ligand for a variety of highly potent HIV-1 protease inhibitors. Among the various dienophiles and Lewis acid-mediated reactions surveyed, 3-(4-methoxybenzoyl)acryloyl oxazolidinone as the dienophile and diethylaluminum chloride as the Lewis-acid provided the desired endo product with excellent diastereoselectivity. The cycloaddition was carried out in multi-gram scale and the cycloadduct was efficiently converted to alcohol 3 with high enantiomeric purity. The optically active ligand was then transformed into potent HIV-1 protease inhibitor 2.
