Boronic esters are versatile building blocks extensively used in organic chemistry and essential to a variety of coupling reactions. In this work, the hydroboration reactions of alkynes were performed without metal catalysts using concomitant microwave irradiation that allowed a significantly reduced reaction time. We conducted a structure-activity relationship study to explore simplified analogues of the itraconazole (ITZ) scaffold for their ability to inhibit the hedgehog (Hh) signaling pathway. These analogues were based on exploring the effects of chemical modifications to the linker and triazolone/side chain region of ITZ. Analogue 11 was identified as the most potent compound in our first generation, with an IC50 value of 81 nM in a murine Hh-dependent basal cell carcinoma. Metabolic identification studies led us to identify truncated piperazine (26) as the major metabolite in human liver microsomes (HLMs) and an improved Hh pathway inhibitor (IC50 = 22 nM). This work verifies that continued truncation of the ITZ scaffold is a practical method to maintain potent anti-Hh activity while also reducing the molecular weight for the ITZ scaffold and achieving improved pharmacokinetic properties.

A series of C-23 modified 23-hydroxybetulinic acid (HBA) derivatives were synthesized and evaluated for their antiproliferative activity against a panel of cancer cell lines (A2780, A375, B16, MCF-7 and HepG2). The biological screening results showed that most of the derivatives exhibited more potent antiproliferative activity than HBA, and compound 6e exhibited the most potent activity with IC50 values of 2.14 mu M, 2.89 mu M, and 3.97 mu M against A2780, B16, and MCF-7 cells, respectively. Further anticancer mechanism studies revealed that compound 6e induced the generation of intracellular reactive oxygen species (ROS) and reduction of mitochondrial membrane potential (MMP) of B16 cells in a dose-dependent manner. Moreover, western blot analysis indicated that compound 6e downregulated the expression of anti-apoptotic protein Bcl-2 and upregulated the expression of proapoptotic protein Bax, activation of caspase 3 to induce cell apoptosis. Meanwhile, compound 6e significantly inhibited the phosphorylation of MEK, ERK, and Akt without affecting the expression of MEK, ERK, and Akt. Furthermore, the in vivo anti-tumor activity of 6e was validated (tumor inhibitory ratio of 68.4% at the dose of 30 mg/kg) in mice with B16 melanoma. (C) 2019 Elsevier Masson SAS. Ferroan anorthosite suite (FAS) rocks are widely interpreted to represent primordial lunar crust. Despite their importance in pinpointing the timing of lunar crust formation, robust chronological investigations for this rock type are scarce. Here, we report the Ar-Ar, Rb-Sr, and Sm-Nd isotopic systematics for the FAS troctolitic anorthosite 62237. The Ar-Ar isotopic system has been reset by a thermal event at 3710 +/- 48 Ma, and the Rb-Sr isotopic systematics has been disturbed such that a Rb-Sr isochron age cannot be determined. However, an internal isochron for the Sm-Nd isotopic system has yielded an age of 4350 +/- 73 Ma (MSWD = 2.0) with an initial epsilon Nd-143(CHUR) of -0.53 +/- 0.26. The mineral and whole-rock fractions of 62237 plot on the same internal isochron as FAS sample 60025. The combined datasets define an age of 4372 +/- 35 Ma (MSWD = 4.0) with an initial epsilon Nd-143(CHUR) of -0.17 +/- 0.22. Literature Sm-Nd data for FAS and Mg-suite whole-rocks also plot on the 60025-62237 isochron. The coherence of data from both FAS and Mg-suite rocks examined thus far suggests that both rock suites formed contemporaneously from identical, or nearly identical, sources. In addition, the concordance of FAS and Mg-suite ages suggests that primordial crust solidification either involved both magmatic suites, or that Mg-suite magmatism was contemporaneous with FAS magmatism within resolution of the Sm-Nd chronometer. The ages for FAS and Mg-suite also coincide with the formation ages of the mare basalt source regions and urKREEP. Ferroan anorthosite suite rocks and urKREEP are thought to represent primordial LMO solidification products, whereas Mg-suite and the mare basalt source regions are argued to represent mixtures of various LMO crystallization products that were formed during density-driven overturn of the LMO. The concordance of ages implies that the 4372 +/- 35 Ma Sm-Nd isochron records the age of mantle overturn, and that overturn occurred during, or shortly after, solidification of the LMO. The insect beta-N-acetylhexosaminidase OfHex1 from Ostrinia furnacalis (one of the most destructive agricultural pests) has been considered as a promising pesticide target. In this study, a series of novel and readily available ureido thioglycosides were designed and synthesized based on the catalytic mechanism and the co-crystal structures of OfHex1 with substrates. After evaluation via enzyme inhibition experiments, thioglycosides 11c and 15k were found to have inhibitory activities against OfHex1 with the K-i values of 25.6 mu M and 53.8 mu M, respectively. In addition, all these ureido thioglycosides exhibited high selectivity toward OfHex1 over hOGA and HsHexB (K-i > 100 mu M). Furthermore, to investigate the inhibitory mechanism, the possible binding modes of 11c and 15k with OfHex1 were deduced based on molecular docking analysis. This work may provide useful structural starting points for further rational design of potent inhibitors of OfHex1.
