We designed and synthesized new atpenin A5 analogs for SAR study. Most of the analogs lacked one or several functional groups in the side chain of atpenin A5, and the stereoisomers proved to be weak nematode complex 11 inhibitors. However, we determined that 4-epi-atpenin A5 was a potent nematode complex II inhibitor comparable to atpenin A5. Tandem catalysis (i.e., a process in which a desirable product is synthesized by a one-step process consisting of sequential reactions) has attracted intensive attention owing to its sustainable green and atom-economical characteristics. In this process, the utilization of a high-efficiency multifunctional catalyst is key. However, different functional sites integrated within the catalyst are required to be rationally designed and precisely engineered to guarantee the synergy between the catalytic reactions. Herein, a novel kind of hydrogenation-esterification tandem catalyst with metal/acid (alloy/polyoxometalates) active sites integrated within the metal-organic frameworks (MOFs) was prepared by a facile self-sacrificial template route. In this tandem catalyst, the MOF cavities served as tandem reactors, the PtNi alloy sites encapsulated within the MOF material acted as hydrogenation sites, and the solid phosphotungstic acid embedded in the MOF cavities provided esterification sites. This well-designed tandem catalyst showed outstanding activity and selectivity towards the one-step synthesis of amino-ester-type anesthetics (e.g., benzocaine) owing to the synergistic catalysis of the metal and acid sites. Clearly, this novel tandem catalyst simplifies the traditional industry process and provides a new method to rationally construct new tandem catalysts.

Assessment of ocular irritation risk is an international regulatory requirement in the safety evaluation of products. In response to this need, L’Oreal developed the SkinEthie Human Corneal Epithelium (HCE) Eye Irritation Test (Err) that has been included in OECD Test Guideline 492. SkinEthic (TM) HCE EIT is able to correctly and reliably identify chemicals not requiring classification versus labelling for eye irritation or serious eye damage according to UN GHS. In an effort to promote its global use, the performance of the method was evaluated after long-distance shipment and compared to European shipment conditions. Results obtained by Cosmos Technical Center (Japan) after extended tissues transit were compared to results obtained in L’Oreal (France). Thirty-nine out of 40 blinded chemicals, representing different functional chemical classes, were consistently classified in both laboratories. The SkinEthic (TM) HCE EIT test method was also evaluated for its performance after extended storage of the tissues. The performance was in agreement with the values reported in OECD TG 492, with an overall accuracy of 87.1% (based on 119 chemicals), sensitivity of 95.5% and specificity of 73.5%. The reliability and relevance of SkinEthic (TM) HCE Err test method after long-distance shipment and extended storage remain in agreement with regulatory validation criteria.

This research focuses on retention mechanisms in a LC column with C18 stationary phase when novel eluent additives (HFIP, HFTB and TFE as well as NFTB and perfluoropinacol) are used. The retention factors between novel eluent additives and conventional ones like ammonium acetate and ammonium bicarbonate at different eluent pH values were compared. A simple set of drug-like molecules, widely spread over different logP values, containing protonated and deprotonated acids and bases was selected for this investigation. HFIP, HFTB, NFTB and PP demonstrated strong influence on basic polar analytes in basic medium. These additives drastically increased retention. A decrease in retention was observed for acidic analytes when novel eluent additives were used. This paper reports the tandem reaction strategy of the Passerini/Wittig reaction based on the in situ capture of isocyanides. BACKGROUND: Transitional care programs (TCPs) coordinate care to improve safety and efficiency surrounding hospital discharge. While TCPs have the potential to reduce hospital length of stay and readmissions, their financial implications are less well understood. OBJECTIVE: To perform a cost-benefit analysis of a previously published neurosurgical TCP implemented at an urban academic hospital from 2013 to 2015. METHODS: Patients received intensive preoperative education and framing of expectations for hospitalization, in-hospital discharge planning and medication reconciliation with a nurse educator, and a follow-up phone call postdischarge. The cost-benefit analysis involved program costs (nurse educator salary) and total direct hospital costs within the 30-d perioperative window including readmission costs. RESULTS: The average cost of the TCP was $435 per patient. The TCP was associated with an average total cost reduction of 17.2% (95% confidence interval [CI]: 7.3%-26.7%, P=.001). This decrease was driven by a 14.3% reduction in the average initial admission cost (95% CI: 6.2%-23.7%, P=.001), largely attributable to the 16.3% decrease in length of stay (95% CI: 9.93%-23.49%, P<.001). Thirty-day readmissions were significantly decreased in the TCP group, with a 5.5% readmission rate for controls and 2.4% for TCP enrollees (P=.04). The average cost of readmission was decreased by 71.3% (95% CI: 58.7%-74.7%, P<.01). CONCLUSION: This neurosurgical TCP was associated with decreased costs of initial admissions, 30-d readmissions, and total costs of hospitalization alongside previously published decreased length of stay and reduced 30-d readmission rates. These results underscore the clinical and financial feasibility and impact of transitional care in a surgical setting. Ferroan anorthosite suite (FAS) rocks are widely interpreted to represent primordial lunar crust. Despite their importance in pinpointing the timing of lunar crust formation, robust chronological investigations for this rock type are scarce. Here, we report the Ar-Ar, Rb-Sr, and Sm-Nd isotopic systematics for the FAS troctolitic anorthosite 62237. The Ar-Ar isotopic system has been reset by a thermal event at 3710 +/- 48 Ma, and the Rb-Sr isotopic systematics has been disturbed such that a Rb-Sr isochron age cannot be determined. However, an internal isochron for the Sm-Nd isotopic system has yielded an age of 4350 +/- 73 Ma (MSWD = 2.0) with an initial epsilon Nd-143(CHUR) of -0.53 +/- 0.26. The mineral and whole-rock fractions of 62237 plot on the same internal isochron as FAS sample 60025. The combined datasets define an age of 4372 +/- 35 Ma (MSWD = 4.0) with an initial epsilon Nd-143(CHUR) of -0.17 +/- 0.22. Literature Sm-Nd data for FAS and Mg-suite whole-rocks also plot on the 60025-62237 isochron. The coherence of data from both FAS and Mg-suite rocks examined thus far suggests that both rock suites formed contemporaneously from identical, or nearly identical, sources. In addition, the concordance of FAS and Mg-suite ages suggests that primordial crust solidification either involved both magmatic suites, or that Mg-suite magmatism was contemporaneous with FAS magmatism within resolution of the Sm-Nd chronometer. The ages for FAS and Mg-suite also coincide with the formation ages of the mare basalt source regions and urKREEP. Ferroan anorthosite suite rocks and urKREEP are thought to represent primordial LMO solidification products, whereas Mg-suite and the mare basalt source regions are argued to represent mixtures of various LMO crystallization products that were formed during density-driven overturn of the LMO. The concordance of ages implies that the 4372 +/- 35 Ma Sm-Nd isochron records the age of mantle overturn, and that overturn occurred during, or shortly after, solidification of the LMO. Caseinolytic protease P (ClpP) is considered as a promising target for the treatment of Staphylococcus aureus infections. In an unbiased screen of 2632 molecules, a peptidomimetic boronate, MLN9708, was found to be a potent suppressor of SaClpP function. A time-saving and cost-efficient strategy integrating in silico position scanning, multistep miniaturized synthesis, and bioactivity testing was deployed for optimization of this hit compound and led to fast exploration of structure-activity relationships. Five of 150 compounds from the miniaturized synthesis exhibited improved inhibitory activity. Compound 43Hf was the most active inhibitor and showed reversible covalent binding to SaClpP while did not destabilize the tetradecameric structure of SaClpP. The crystal structure of 43Hf-SaClpP complex provided mechanistic insight into the covalent binding mode of peptidomimetic boronate and SaClpP. Furthermore, 43Hf could bind endogenous ClpP in S. aureus cells and exhibited significant efficacy in attenuating S. aureus virulence in vitro and in vivo.
