Synthesis, characterization, biological activity and equilibrium studies of metal(II) ion complexes with tridentate hydrazone ligand derived from hydralazine

In the present study, a new hydrazone ligand (2-((2-phthalazin-1-yl)hydrazono)methyl)phenol) prepared by condensation of hydralazine (1-Hydralazinophthalazine) with salicylaldehyde (SAH). The synthesized SAH-hydrazone and its metal complexes have been characterized by elemental analyses, IR, H-1 NMR, solid reflectance, magnetic moment, molar conductance, mass spectra. UV-vis and thermal analysis (TGA). The analytical data of the complexes show the formation of 1:1 [M:L] ratio, where M represents Ni(II), Co(II) and Cu(II) ions, while L represents the deprotonated hydrazone ligand. IR spectra show that SAH is coordinated to the metal ions in a tridentate manner through phthalazine-N, azomethine-N and phenolic-oxygen groups. The ligand and their metal chelates have been screened for their antimicrobial activities using the disc diffusion method against the selected bacteria and fungi. Proton-ligand association constants of (SAH) and the stepwise stability constants of its metal complexes are determined potentiometrically in 0.1 M NaNO3 at different temperatures and the corresponding thermodynamic parameters were derived and discussed. The order of -Delta G degrees and -Delta H degrees were found to obey Mn2+ < Co2+ < Ni2+ < Cu2+, in accordance with the Irving-Williams order. The complexes were stabilized by enthalpy changes and the results suggest that the complexation is an enthalpy-driven process. The concentration distribution diagrams of the complexes are evaluated. Effects of 6-(4-chlorophenoxy)-tetrazolo[5,1-a]phthalazine on Anticoagulation in Mice and the Inhibition of Experimental Thrombosis in Rats

Thrombosis is a major complication that could be fatal in acute or chronic cardio-cerebral-vascular diseases. Therefore, the development of novel agents for anticlotting and the prevention of thrombosis and cardiovascular diseases are clinically significant. This study aimed to evaluate the anticoagulant and antithrombotic effects of 6-(4-chlorophenoxy)-tetrazolo[5,1-a]phthalazine (Q808), a new phthalazine tetrazole derivative. Bleeding time, clotting time, and serum calcium ion (Ca2+) concentration were assessed in mice, whereas arteriovenous thrombus weight and plasma prothrombin time were evaluated in rats, and platelets Ca2+ influx was determined in rabbit. Daily oral administration of Q808 at 25, 50, or 100 mg/kg for 3 days significantly delayed bleeding time and clotting time in mice compared with controls. Q808 administration at 50 mg/kg significantly reduced experimental thrombus weight by 62.6% and delayed plasma prothrombin time by 58.7% in rats, whereas 50 and 100 mg/kg of Q808 daily significantly increased serum Ca2+ concentration in mice. Q808 at 0.2, 0.4, and 0.8 mg/mL significantly inhibited thrombin-induced Ca2+ influx in rabbit platelets. Our results suggest that Q808 at 25-200 mg/kg daily exerts anticoagulant and antithrombotic effects, and its mechanisms of action may involve both the intrinsic and extrinsic coagulation pathways that inhibit certain coagulation factors and platelet functions.

Synthesis and Antimicrobial Activity of Novel Heterocycles Utilizing 3-(1,4-Dioxo-3,4-dihydrophthalazin-2(1H)-yl)-3-oxopropanenitrile as Precursors

A high-strength PPESK/PVDF fibrous membrane prepared by coaxial electrospinning for lithium-ion battery separator

Electrospinning fibrous membranes have attracted a great deal of attention because of their advantages, including uniform pore size, large ratio surface area, and high porosity. For extended application in lithium-ion battery, it is essential to further improve their electrochemical, mechanical, and thermal properties. In this work, a new poly (phthalazine ether sulfone ketone) (PPESK)/polyvinyli-denefluoride (PVDF) core/shell fibrous membrane was fabricated via the coaxial electrospinning technique, followed by hot press. The PPESK/PVDF membrane hot pressed at 160 degrees C exhibits excellent comprehensive performance, including large porosity (80%), high electrolyte uptake (805%), and excellent thermal stability (at 200 degrees C). Moreover, due to the improved bonding effect derived from the solidification of the PVDF shell layer after the hot press, the mechanical property of the membrane is effectively enhanced. The electrochemical tests also indicate that the PPESK/PVDF membrane shows larger ionic conductivity and lower interfacial resistance when compared with commercial microporous polypropylene separator. In addition, simulated cells assembled with the PPESK/PVDF membrane present superior discharge capacity, stable cycle performance, and excellent rate capability. Therefore, the hot-pressed coaxial PPESK/PVDF fibrous membrane has the potential to be a promising candidate as the separator for high-performance lithium-ion battery.

Examination of Diazaspiro Cores as Piperazine Bioisosteres in the Olaparib Framework Shows Reduced DNA Damage and Cytotoxicity

Development of poly(ADP-ribose) polymerase inhibitors (PARPi’s) continues to be an attractive area of research due to synthetic lethality in DNA repair deficient cancers; however, PARPi’s also have potential as therapeutics to prevent harmful inflammation. We investigated the pharmacological impact of incorporating spirodiamine motifs into the phthalazine architecture of FDA approved PARPi olaparib. Synthesized analogues were screened for PARP-1 affinity, enzyme specificity, catalytic inhibition, DNA damage, and cytotoxicity. This work led to the identification of 10e (12.6 +/- 1.1 nM), which did not induce DNA damage at similar drug concentrations as olaparib. Interestingly, several worst in class compounds with low PARP-1 affinity, including 15b (4397 +/- 1.1 nM), induced DNA damage at micromolar concentrations, which can explain the cytotoxicity observed in vitro. This work provides further evidence that high affinity PARPi’s can be developed without DNA damaging properties offering potential new drugs for treating inflammatory related diseases.
