Then, the formulated [Simp](3)PW12O40 was changed to the nano-form via solvothermal processing in tetralin at 220 degrees C. After that, the provided powdered solid nanomaterial was characterized by FTIR, H-1 NMR, XRD, SEM, EDX and TGA analyses. Then, the focused nanomaterial was handled as a productive and encouraging nanocatalyst for the establishment of 2H-indazolo[2,1-b]phthalazine-1,6,11(13H)-trione attendants through one-pot, multi-component contraction of aldehydes, phthalhydrazide, and dimedone under solventless status and the generality and practical tolerance of this useful and environmentally benign method are illustrated. The simple work-up, smooth reaction conditions, good to excellent yields, and reasonably short reaction times are the particular benefits of this protocol. Besides, the new nano-powdered ionic liquid was a successful and modest catalyst that could definitely be recycled and reused several times without apparent decrease in its catalytic activity. Synthesis and evaluation of anti-proliferative activity of 1,4-disubstituted phthalazines

A series of new phthalazine derivatives 3a-i and 4a-c were synthesized via the reaction of 1-chlorophthalazine derivative 2 with either N-substituted-piperazines, primary or their secondary amines. Nanoscale liposomal formulation of a SYK P-site inhibitor against B-precursor leukemia

We report preclinical proof of principle for effective treatment of B-precursor acute lymphoblastic leukemia (ALL) by targeting the spleen tyrosine kinase (SYK)-dependent antiapoptotic blast cell survival machinery with a unique nanoscale pharmaceutical composition. This nanoscale liposomal formulation (NLF) contains the pentapeptide mimic 1,4-Bis (9-O dihydroquinidinyl) phthalazine/hydroquinidine 1,4-phathalazinediyl diether (C61) as the first and only selective inhibitor of the substrate binding P-site of SYK. The C61 NLF exhibited a very favorable pharmacokinetic and safety profile in mice, induced apoptosis in primary B-precursor ALL blast cells taken directly from patients as well as in vivo clonogenic ALL xenograft cells, destroyed the in vivo clonogenic fraction of ALL blast cells, and, at nontoxic dose levels, exhibited potent in vivo antileukemic activity against patient-derived ALL cells in xenograft models of aggressive B-precursor ALL. Our findings establish SYK as an attractive molecular target for therapy of B-precursor ALL. Further development of the C61 NLF may provide the foundation for therapeutic innovation against therapy-refractory B-precursor ALL.

Thermal stability of selenium, sulfur and nitrogen analogous phthalazine derivatives

Differential scanning calorimetry (DSC) and thermogravimetry (TG) are analytical and quantitative methods capable of providing reliable, fast and reproducible results. These data allow establishing the thermal stability, purity degree and the polymorphic behavior of organic compounds. Thermal analysis of fusion and degradation processes was carried out on organonitrogen, organosulfur and organoselenium phthalazine derivatives to establish thermal stability criteria. Decomposition and fusion temperatures of 27 biological active compounds, synthesized by our research group were determined using TG and DSC. Analysis of the thermal data indicated that: (a) in general, nitrogen compounds are more stable than sulfur and selenium compounds; (b) thioderivatives possess degradation temperatures higher than selenium compounds; (c) the presence of selenium atoms in molecular structure has associated a minor thermal stability; (d) sulfide derivatives decomposition process have higher T-onset values than disulfide compounds; (e) there are differences in the stability due to groups selenol, methylseleno, and cyanoseleno; (f) the nature of the substituent located on the benzyl ring has no effects on selenophthalazines thermal stability.

Synthesis and evaluation of novel dimethylpyridazine derivatives as hedgehog signaling pathway inhibitors

We report herein the design and synthesis of a series of structural modified dimethylpyridazine compounds as novel hedgehog signaling pathway inhibitors. The bicyclic phthalazine core and 4-methylamino- piperidine moiety of Taladegib were replaced with dimethylpyridazine and different azacycle building blocks, respectively. The in vitro Gli-luciferase assay results demonstrate that the new scaffold still retained potent inhibitory potency. Piperidin-4-amine moiety was found to be the best linker between pharmacophores dimethylpyridazine and fluorine substituted benzoyl group. Furthermore, the optimization of 1-methyl-1H-pyrazol and 4-fluoro-2-(trifluoromethyl) benzamide by different aliphatic or aromatic rings were also investigated and the SAR were described. Several new derivatives were found to show potent Hh signaling inhibitory activity with nanomolar IC50 values. Among these compounds, compound 11c showed the highest inhibitory potency with an IC50 value of 2.33 nM, which was comparable to the lead compound Taladegib. Formation kinetics and characterization of polyphthalazinone ether ketone hollow fiber ultrafiltration membranes

This paper reports an evaluation of the effect of both organic and inorganic additives on the gelation kinetics, morphology and separation properties of asymmetric poly(phthalazine ether ketone) (PPEK) hollow fiber ultrafiltration membranes. Using an online optical microscope-CCD camera for observation of precipitation front movement (X), a good linear relationship between X-2 and time was observed for the whole gelation process with the correlation coefficient (R-2) higher than 0.98. It was found that the pure water flux of membranes with additives were lower than neat PPEK membranes. This observation might be attributed to the decrease in the face gelation velocity caused by the extra organic additives. The water flux of PEG600 was higher than that of Tween80 at the same additive concentration. When OA concentration increased from 3.5 to 5.0 wt.%, the pure water flux increased from 332 to 367 L/m(2) hand the rejection decreased from 98.6 to 96.8%. The PPEK hollow fiber membranes exhibited finger-like macrovoids structure and a skin layer at the outer-most region of the hollow fiber. Evaluation of the membrane chemical resistance with various chemical solutions indicated that PPEK membranes had good chemical resistance against HCI, H2SO4, HNO3, CH3COOH, OA, NaOH and H2O2, while the NaClO solution can greatly damage the membrane mechanical strength. Static sorption tests of PPEK membrane revealed that PPEK membrane adsorbed more bacitracin than BSA due to the smaller size of bacitracin. (C) 2011 Elsevier B.V. All rights reserved.
