Synthesis of some pyridazine derivatives as antioxidants and antimicrobial agents

Purpose: To prepare some pyridazine derivatives as antioxidant and antimicrobial agents. Methods: The coupling of aryl diazonium salts with diethyl 2-cyano-3-methylglutaconate afforded pyridazine-5-carbonitrile derivatives (3a-e). The methyl function in the pyridazine derivatives (3a-e) reacted with cinnamonitriles to yield phthalazine derivatives (6a-h). Imino-pyridazine derivatives (9a-h) were obtained by reacting 2-aminoprop-1-ene-1,1,3-tricarbonitrile along with aryl diazonium salts (1a-h). Structure elucidation was done by spectral analysis. The 1,1-diphenyl-2-picryl hydrazyl (DPPH) technique was employed to assess antioxidant effect, while evaluation of antimicrobial activity was carried out by serial dilution technique. Results: Compounds, 6f (IC50 = 12.68 mu g/mL; p < 0.05), 6a (IC50 = 14.23 mu g/mL; p < 0.05), and 3c (IC50 = 14.34 mu g/mL; p < 0.05) displayed good antioxidant activity compared to ascorbic acid (IC50 = 12.45 mu g/mL; p < 0.05). Based on the zone of inhibition, compound 6c (p < 0.05) displayed higher activity than ofloxacin (p < 0.05) against E. coli. Conclusion: The presence of an oxo group and carboxylate group plays an important role in the antioxidant activity of these compounds. Compounds 6a, 6f, and 3c can potentially serve as lead compounds for the development of promising antioxidants. Efficient ZrO(NO3)(2.)2H(2)O Catalyzed Synthesis of 1H-Indazolo[1,2-b] phthalazine-1,6,11(13H)-triones and Electronic Properties Analyses, Vibrational Frequencies, NMR Chemical Shift Analysis, MEP: A DFT Study

The synthesis of 1H-indazolo[1,2-b]phthalazine-1,6,11(13H)-trione derivatives, using one-pot three-component condensation reaction of 3-nitrophthalic anhydride, hydrazine monohydrate, dimedone, and aromatic aldehydes in the presence of ZrO(NO3)(2).2H(2)O as the novel catalyst and in reflux conditions in EtOH was reported. Quantum theoretical calculations for three structures of compounds (5a, 5b, and 5c) were performed using the Hartree-Fock (HF) and density functional theory (DFT). From the optimized structure, geometric parameters were obtained and experimental measurements were compared with the calculated data. The structures of the products were confirmed by IR, H-1 NMR, C-13 NMR, mass spectra, and elemental analyses. The IR spectra data and H-1 NMR and C-13 NMR chemical shift computations of the 1H-indazolo[1,2-b]phthalazine-1,6,11(13H)-trione derivatives in the ground state were calculated. Frontier molecular orbitals (FMOs), total density of states (DOS), thermodynamic parameters, and molecular electrostatic potential (MEP) of the title compounds were investigated by theoretical calculations. Molecular properties such as the ionization potential (I), electron affinity (A), chemical hardness (eta), electronic chemical potential (mu), and electrophilicity (omega) were investigated for the structures. Thus, there was an excellent agreement between experimental and theoretical results.

A novel sulfamic acid functionalized nano-catalyst on the basis of calix[4]resorcinarene for the green one-pot synthesis of 2H-indazolo[2,1-b]phthalazine-triones under thermal solvent-free conditions

N-propyl sulfamic acid supported nano-catalyst on the basis of calix[4]resorcinarene was prepared via the efficient and facile reaction of amine functionalized polycalix[4]resorcinarene with chlorosulfonic acid. The achieved catalytic system was characterized using some spectroscopic techniques such as Fourier transform Infrared (FT-IR) spectroscopy, field emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), and CHNS elemental analysis. This newly developed acidic catalyst was employed efficiently in a one-pot three-component condensation reaction of aromatic aldehydes, dimedone and phthalhydrazide for the synthesis of 2H-indazolo[2,1-b]phthalazine-trione derivatives through an easy and eco-friendly methodology. The catalyst was easily separated from the reaction mixture by simple filtration and the desired products were achieved in good to excellent yields in short reaction times.

Finally, the dipeptide 38 was prepared by coupling of 35 with L-alanine methyl ester. Some of these compounds were screened in vitro for their antimicrobial activity. The energy gap between the highest occupied molecular orbital and lowest unoccupied molecular orbital has been calculated using the theoretical computations to reflect the chemical reactivity and kinetic stability of compounds. A theoretical study of low-lying singlet and triplet excited states of quinazoline, quinoxaline and phthalazine: insight into triplet formation

Quinazoline, quinoxaline and phthalazine are nitrogen containing heterocyclic aromatic molecules which belong to the class diazanaphthalenes. These isomers have low-lying n pi(star) and naphthalene-like pi pi(star) states that interact via spin-orbit coupling. In this contribution, we study their structure and electronic states by means of a coupled-cluster method. The computed properties are compared to those of cinnoline which were obtained in our previous study [Etinski et al., Phys. Chem. Chem. Phys., 2014, 16, 4740]. The excited state features of these isomers are dependent on the position of the nitrogen atoms. We find that quinazoline and quinoxaline exhibit similarities in the ordering and character of the excited states. In contrast, a marked difference in the electronic and geometric structures of the lowest excited triplet states of cinnoline and phthalazine is noticed, although both are orthodiazanaphthalenes. Our findings suggest that the S-1 (sic) T-1 channel is responsible for the rapid intersystem crossing in quinazoline and quinoxaline, whereas the S-1 (sic) T-2 pathway is active in phthalazine.

In this approach the dominant intermolecular interactions between acid-base pairs are identified. Such interactions appear to be crucial for the inhibition or promotion of proton transfers in the solid state. For this, the relationship between these interactions and the so-called ApKa rule has been investigated.

Highly Regioselective Carbamoylation of Electron-Deficient Nitrogen Heteroarenes with Hydrazinecarboxamides

The use of hydrazinecarboxamides as a new class of carbamoylating agents has been established through the dehydrazinative Minisci reaction of electron-deficient nitrogen heteroarenes.
