Synthesis of new pyrrolo[2,1-a] phthalazine derivatives via multicomponent reaction of phthalazine with 1,1-dicyanoalkenes and alkyl isocyanides

Imidazole-containing phthalazine derivatives inhibit Fe-SOD performance in Leishmania species and are active in vitro against visceral and mucosal leishmaniasis

The in vitro leishmanicidal activity of a series of imidazole-containing phthalazine derivatives 1-4 was tested on Leishmania infantum, Leishmania braziliensis and Leishmania donovani parasites, and their cytotoxicity on J774.2 macrophage cells was also measured. On the other hand, the most active compounds 2 and 4 were potent inhibitors of iron superoxide dismutase enzyme (Fe-SOD) in the three species considered, whereas their impact on human CuZn-SOD was low. Molecular modelling suggests that 2 and 4 could deactivate Fe-SOD due to a sterically favoured enhanced ability to interact with the H-bonding net that supports the antioxidant features of the enzyme.

Their structures have been determined by single-crystal X-ray diffraction, confirming that all compounds have twisted configurations with torsion angles between the pyrene unit and the 2,3-diazaanthrance part as high as 21.52 degrees (for 1), 24.74 degrees (for 2), and 21.14 degrees (for 3). The optical bandgaps for all compounds corroborate the values derived from CV. The calculation done by DFT shows that the HOMO-LUMO bandgaps are in good agreement with experimental data. Interestingly, the substituted groups (phenyl, pyridyl, thienyl) in the 1,4-positions did affect their self-assembly and the optical properties of as-resulted nanostructures. Under the same conditions, compounds 1-3 could self-assemble into different morphologies such as microrods (for 1), nanoprisms (for 2), and nanobelts (for 3). Moreover, the UV-vis absorption and emission spectra of as-prepared nanostructures were largely red-shifted, indicating J-type aggregation for all materials. Surprisingly, both 1 and 2 showed aggregation-induced emission (ALE) effect, while compound 3 showed aggregation-caused quenching (ACQ) effect. Our method to approach novel twisted azaacenes through [4 + 2] reaction could offer a new tool to develop unusual twisted conjugated materials for future optoelectronic applications.

Pyridazine-Containing Diazatwistanthracene and Tetraazatwisttetracene: Synthesis, Crystal Structures and Third Order Non-linear Optical Properties

In this paper, two pyridazine-heterocycles-containing azaacenes 9,12-diphenylphenanthro[4,5-fgh]phthalazine (DPPA) and 2,9-di-tert-butyl-4,7,11,14-tetraphenylpyridazino[4′,5′:9,10] phenanthro[4,5-fgh]phthalazine (TPPA) have been synthesized and fully characterized. The single crystal analysis of TPPA showed that TPPA molecule exhibits a twisted structure with the dihedral angel of 27 degrees between pyrene and pyridazine unit, which suppresses the pi-pi stackings in the solid state. The two-photon absorption properties of DPPA and TPPA were investigated, respectively, by using the femtosecond Z-scan technique. The results showed that both DPPA and TPPA showed moderate two-photon absorption cross-section values, and the larger one was found to be 454 GM with TPPA. Obviously, the introduction of pyridazine ring into the skeleton of acenes forming a A-D-A structure could lead to a much larger two-photon absorption coefficient and two-photon absorption cross section.

L-4, a Well-Tolerated and Orally Active Inhibitor of Hedgehog Pathway, Exhibited Potent Anti-tumor Effects Against Medulloblastoma in vitro and in vivo

Inhibition of aberrant Hedgehog (Hh) pathway had been proved to be a promising therapeutic intervention in cancers like basal cell carcinoma (BCC), medulloblastoma (MB), and so on. Two drugs (Vismodegib, Sonidegib) were approved to treat BCC and more inhibitors are in clinical investigation. However, the adverse effects and drug resistance restricted the use of Hh inhibitors. In the present study, 61 synthesized compounds containing central backbone of phthalazine or dimethylpyridazine were screened as candidates of new Hh signaling inhibitors by performing dual luciferase reporter assay. Among the compounds, L-4 exhibited an IC50 value of 2.33 nM in the Shh-Light II assay. L-4 strongly inhibited the Hh pathway in vitro and blocked the Hh pathway by antagonizing the smoothened receptor (Smo). Remarkably, L-4 could significantly suppress the Hh pathway activity provoked by Smo mutant (D473H) which showed strong resistant properties to existing drugs such as Vismodegib. Orally administered L-4 exhibited prominent dose-dependent anti-tumor efficacy in vivo in Ptch+/-; p53-/- MB allograft model. Furthermore, L-4 showed good tolerance in acute toxicity test using ICR mice. These evidences indicated that L-4 was a potent, well-tolerated, orally active inhibitor of Hedgehog pathway, and might be a promising candidate in development of Hh-targeted anti-cancer drugs.

Inhibitor design to target a unique feature in the folate pocket of Staphylococcus aureus dihydrofolate reductase

Staphylococcus aureus (Sa) is a serious concern due to increasing resistance to antibiotics. The bacterial dihydrofolate reductase enzyme is effectively inhibited by trimethoprim, a compound with antibacterial activity. Previously, we reported a trimethoprim derivative containing an acryloyl linker and a dihydophthalazine moiety demonstrating increased potency against S. aureus. We have expanded this series and assessed in vitro enzyme inhibition (K-i) and whole cell growth inhibition properties (MIC). Modifications were focused at a chiral carbon within the phthalazine heterocycle, as well as simultaneous modification at positions on the dihydrophthalazine. MIC values increased from 0.0626-0.5 mu g/mL into the 0.5-1 mu g/mL range when the edge positions were modified with either methyl or methoxy groups. Changes at the chiral carbon affected K-i measurements but with little impact on MIC values. Our structural data revealed accommodation of predominantly the S-enantiomer of the inhibitors within the folate-binding pocket. Longer modifications at the chiral carbon, such as p-methylbenzyl, protrude from the pocket into solvent and result in poorer K-i values, as do modifications with greater torsional freedom, such as 1-ethylpropyl. The most efficacious K-i was 0.7 +/- 0.3 nM, obtained with a cyclopropyl derivative containing dimethoxy modifications at the dihydrophthalazine edge. The co-crystal structure revealed an alternative placement of the phthalazine moiety into a shallow surface at the edge of the site that can accommodate either enantiomer of the inhibitor. The current design, therefore, highlights how to engineer specific placement of the inhibitor within this alternative pocket, which in turn maximizes the enzyme inhibitory properties of racemic mixtures.
