A High-Temperature, High-Throughput Method for Monitoring Residual Formaldehyde in Vaccine Formulations

Formaldehyde has long been used in the chemical inactivation of viral material during vaccine production. Viral inactivation is required so that the vaccine does not infect the patient. Formaldehyde is diluted during the vaccine manufacturing process, but residual quantities of formaldehyde are still present in some current vaccines. Although formaldehyde is considered safe for use in vaccines by the Food and Drug Administration, excessive exposure to this chemical may lead to cancer or other health-related issues. An assay was developed that is capable of detecting levels of residual formaldehyde in influenza vaccine samples. The assay employs incubation of dosage formulation suspensions with hydralazine hydrochloride under mildly acidic conditions and elevated temperatures, where formaldehyde is derivatized to yield fluorescent s-triazolo-[3,4-a]-phthalazine. The assay has been traditionally run by high-performance liquid chromatography, where runtimes of 15 minutes per sample can be expected. Our laboratory has developed a plate-based version that drastically improved the throughput to a runtime of 96 samples per minute. The assay was characterized and validated with respect to reaction temperature, evaporation, stability, and selectivity to monitor residual formaldehyde in various influenza vaccine samples, including in-process samples. Heat transfer and evaporation will be especially considered in this work. Since the assay is plate based, it is automation friendly. The new assay format has attained detection limits of 0.01 mu g/mL residual formaldehyde, which is easily able to detect and quantify formaldehyde at levels used in many current vaccine formulations (<5 mu g/0.5-mL dose). Synthesis, crystal structure and antifungal activity of a Ni(II) complex of a new hydrazone derived from antihypertensive drug hydralazine hydrochloride

A new complex of nickel(II) with 1-(2-[(5-methylfuran-2yl)methylene)] hydrazono) phthalazine HL (1) has been synthesized and characterized by elemental analysis, IR, electronic spectra, TGA, H-1 and C-13 NMR spectroscopy. The crystal structures of the free ligand and its complex have been determined by single crystal X-ray diffraction techniques. In the complex the hydrazone ligand chelates to the nickel(II) through nitrogen atoms in a bidentate manner. The antifungal activity of the ligand and its nickel(II) complex 3 was studied against Aspergillus niger, Aspergillus flavus and Candida albicans. The results revealed modest activity of the complex (3) against the tested organisms with inhibition zones of 14, 14 and 13 mm, compared to the free ligand 1 with inhibition zones of 11 mm each for A. niger, A. flavus and C. albicans respectively. Green synthesis of 1,3,4-oxadiazole derivatives based on N-arylidene-2-(1-oxo-4-(4-phenoxyphenyl)phthalazin-2(1H)-yl)acetohydrazide as potential antitumor agents

Design of a new series of 1,3,4-oxadiazole derivatives was achieved in good yield via treatment of N-arylidene-2-(1-oxo-4-(4-phenoxyphenyl)phthalazin-2(1H)-yl)acetohydrazide with cerium (IV) ammonium nitrate as a catalyst. They can be formulated also using one-pot reaction of 2-(1-oxo-4-(4-phenoxyphenyl)phthalazin-2(1H)-yl)acetohydrazide, aromatic aldehyde and cerium (IV) ammonium nitrate in dichloromethane. The structural formulas of all synthesized products were elucidated based on their elemental and spectral analyses. In addition, MTT assay was used to evaluate antitumor activity of the synthesized molecules and some of them showed potent activity in comparison with Doxorubicin as a standard drug.

For cholinesterase enzymes, the K-i values were found in the range of 47.26 +/- 9.62-72.05 +/- 19.47 nM against acetylcholinesterase (AChE) and 65.03 +/- 9.88-102.83 +/- 25.04 nM against butyrylcholinesterase (BChE), respectively. The inhibition effects of these compounds against enzymes whose name are AChE, BChE, alpha-Gly, hCA I, and hCA II, were compared with control molecules like tacrine, acarbose, and acetazolamide.

Aldehyde oxidase (AOX) is a cytosolic enzyme responsible for the metabolism of some drugs and drug candidates. AOX catalyzes the oxidative hydroxylation of substrates including several aliphatic and aromatic aldehydes, and nitrogen-containing heterocyclic compounds. AOX is also reported to catalyze the reductive metabolism of nitro-compounds, N-oxides, sulfoxides, isoxazoles, isothiazoles, nitrite and hydroxamic acids. These reductive transformations are not well understood and are generally believed to only occur at low oxygen concentrations. In this study, we used 5-nitroquinoline (5NQ) as a substrate to further understand both the oxidative and the reductive transformations catalyzed by AOX. In vitro reaction of 5NQ with AOX under aerobic conditions generated the oxidized (2-oxo-5-nitroquinoline, 2-oxo-5NQ), the reduced (5-aminoquinoline, 5AQ) and the oxidized/reduced (2-oxo-5-aminoquinoline, 2-oxo-5AQ) metabolites. Interestingly, in human liver cytosol, co-incubation of 5NQ and known AOX oxidative substrates DACA and phthalazine significantly increased the yield of the reduced metabolite, while oxidized metabolites production decreased. These data indicate that 5NQ can be reduced at atmospheric oxygen concentrations and that the reductive transformation occurs at a second site that is kinetically distinct from the oxidative site.
