Efficient Synthesis of 2H-Indazolo[2,1-b]Phthalazine-Triones Using [PVPH]ClO4 as a Modified Polymeric Catalyst

In this work, poly(vinylpyrrolidonium) perchlorate {[PVPH]ClO4}, a newly reported modified polymeric catalyst, is used as an efficient and reusable solid acid catalyst for the promotion of the synthesis of 2H-indazolo[2,1-b]phthalazine-trione derivatives. All reactions were performed under mild conditions and the products were formed over short reaction times in excellent yields. It is important to note that this solid acid catalyst can be reused several times without any appreciable loss in its activity.

Results showed that when imidoyl chloride derivative has a fluorine atom at ortho position (3a-c), in two steps under a S(N)i mechanism and then SNAr reaction mechanism produce ay1-5-(trifluoromethyl)-1H-benzo [el [1,2,4]triazolo[1,2-a] [1,2,4]triazine-1,3(2H)-diones (4a-c) and 6-(trifluoromethyl)benzo[5,6][1,2,4]triazino[1,2-b]phthalazine-8,13-diones (4i,4j) in good to excellent yields. In the other imidoyl derivatives (3d-h and 3i,3j) under these conditions cyclization reaction does not occur and therefore produce 1-aryl-2,2,2-trifluorornethyl)-4-phenyl-1,2,4-triazolidine-3,5-diones (4d-h) and 2-(2,2,2-trifluoro-1-(arylimino)ethyl)-2,3-dihydrophthalazine-1,4-diones(4k,41) (non-cyclic products) in good to excellent yields. Experimental and theoretical study on one-pot, three-component route to 2H-indazolo[2,1-b]phthalazine-triones catalyzed by nano-alumina sulforic acid

A simple, efficient, and high yielding one-pot protocol for the synthesis of 2H-indazolo[2,1-b]phthalazine-trione derivatives has been developed by three-component coupling of phthalhydrazide, dimedone and some aromatic aldehydes in ecofriendly neat conditions promoted by nano-gamma-alumina sulforic acid. This protocol avoids the use of expensive catalysts, toxic solvents and chromatographic separation. Also the proposed protocol was extended to the linear beta-diketone (acetylaceton) instead of cyclic diketone (dimedone). The generality and functional tolerance of this convergent and environmentally benign method is demonstrated. The nature of each step of the suggested mechanism for this condensation reaction is also identified through Density Functional Theory (DFT) calculations using B3LYP/6-31G** level of theory. It is shown that the first step is charge controlled; whereas the second one is controlled by frontier molecular orbitals. The H-1 and C-13 chemical shift values together with the structural parameters of the title compound have been also computed and the scaled values have been compared with the corresponding experimental NMR and X-ray spectra. As a result, the calculated spectroscopic data and optimized geometry show a good agreement with the experimental results. (C) 2012 Elsevier B.V. All rights reserved.

Synthesis and evaluation of in vitro and in vivo trypanocidal properties of a new imidazole-containing nitrophthalazine derivative

A series of new phthalazine derivatives (1-4) containing imidazole rings and functionalized with nitro groups in the benzene ring of the phthalazine moiety were prepared and identified on the basis of their MS, elemental analyses and bidimensional H-1 and C-13 NMR data, and their trypanocidal activity was tested. The 8-nitrosubstituted compound (3) was more active in vitro against Trypanosoma cruzi and less toxic against Vero cells than the reference drug benznidazole, and showed a SI value that was 47-fold better than the reference drug in amastigote forms. It also remarkably reduced the infectivity rate in Vero cells and decreased the reactivation of parasitemia in immunodeficient mice. Ultrastructural alterations found in epimastigotes treated with 3 confirmed extensive cytoplasm destruction in the parasites, whereas histopathological analysis of the hearts of mice infected and treated with 3 resulted in a decrease in cardiac damage. Biochemical markers showed that livers, hearts, and kidneys of treated mice were substantially unaffected by the administration of 3, despite the presence of the potentially toxic nitro group. It was also found that this compound selectively inhibited the antioxidant parasite enzyme Fe-superoxide dismutase (Fe-SOD) in comparison with human CuZn-SOD, and molecular modeling suggested interaction with the H-bonding system of the iron-based moiety as a feasible mechanism of action against the enzyme. Several compounds were developed and showed favorable activities compared to the standard drug milrinone, with (4-([1,2,4]triazolo[3,4-a]phthalazin-6-yl) piperazin-1-yl)(p-tolyl) methanone (5g) being identified as the most potent with an increased stroke volume of 19.15 +/- 0.22% (milrinone: 2.46 +/- 0.07%) at a concentration of 3 x 10(-5) M. A preliminary study of mechanism of action revealed that 5g displayed its positive inotropic effect may be related to the PDE-cAMP-PKA signaling pathway. Aromatic azaheterocycle-cored luminogens with tunable physical properties via nitrogen atoms for sensing strong acids

A series of luminogens comprising one pyridine, 1,3-diazine, 1,4-diazine, 1,2-diazine and phthalazine moiety as the central core and two AIE-active tetraphenylethene units in lateral positions have been readily synthesized by Suzuki cross-coupling. They exhibited remarkably different photophysical and electrochemical properties, as well as solid packing and fine controllability via the number and position of the nitrogen atoms in the aromatic azaheterocycle core. Among them, the pyridine, 1,3-diazine and 1,4-diazine-cored luminogens displayed strong AIE activities, whereas the 1,2-diazine and phthalazine-cored luminogens exhibited almost no AIE effect. The intrinsic Lewis basicity of the as-prepared luminogens endowed them with the ability to fluorometrically detect acids with different pK(a) values. When protonated by a strong acid such as trifluoroacetic acid, the pyridine, 1,3-diazine and 1,4-diazine-cored luminogens displayed relatively weak AIE effects. In contrast, the 1,2-diazine and phthalazine-cored luminogens exhibited highly sensitive responses to strong acids within a precise pKa range by displaying turn-on fluorescence emissions in the low-energy region, which was probably owing to the synergetic effect of AIE and the constraint of the intersystem crossing effect upon protonation of the 1,2-diazine segment. They displayed reversible acidochromism in response to protonation and deprotonation in the solid state. Such unique properties of the as-prepared luminogens could be used for the selective discrimination of some organic acids, which is highly valuable in the study of biological metabolism.
