Synthesis, crystal structure and antifungal activity of a Ni(II) complex of a new hydrazone derived from antihypertensive drug hydralazine hydrochloride

A new complex of nickel(II) with 1-(2-[(5-methylfuran-2yl)methylene)] hydrazono) phthalazine HL (1) has been synthesized and characterized by elemental analysis, IR, electronic spectra, TGA, H-1 and C-13 NMR spectroscopy. The crystal structures of the free ligand and its complex have been determined by single crystal X-ray diffraction techniques. In the complex the hydrazone ligand chelates to the nickel(II) through nitrogen atoms in a bidentate manner. The antifungal activity of the ligand and its nickel(II) complex 3 was studied against Aspergillus niger, Aspergillus flavus and Candida albicans. The results revealed modest activity of the complex (3) against the tested organisms with inhibition zones of 14, 14 and 13 mm, compared to the free ligand 1 with inhibition zones of 11 mm each for A. niger, A. flavus and C. albicans respectively. Synthesis of hydralazine-N-lactose and its metabolites in vitro

Hydralazine-N-lactose is a main impurity in hydralazine tablets. Homogeneous Composite Nonwoven Support for High Temperature-Resistant Separation Membranes

High temperature-resistant separation membranes are widely considered in various fields. However, incompetent support layer limits the performance of high temperature-resistant membranes severely. In this work, a homogeneous composite nonwoven support (HCNS) is developed by wet-laid and hot-pressing technology using undrawn polyethylene terephthalate fibers as bonding fibers. The significance of hot-pressing parameters on the performances of the HCNS is investigated, and quadratic regression models are established. The HCNS exhibits a smooth surface (roughness of 118 nm) and a porous structure (porosity of 21.06% and pore size of 6.548 mu m) with a mechanical property (tensile index of 45.80 N m g(-1)). The ultrafiltration membranes supported by the HCNS are prepared from poly(phthalazine ether sulfone ketone), which demonstrates high permeance, high rejection rate, and long-term stability at raised operation temperature, thereby suggesting the potential application of HCNS in high-temperature filtration.

Magnetic nanoparticles Fe3O4-supported guanidine as an efficient nanocatalyst for the synthesis of 2H-indazolo[2,1-b]phthalazine-triones under solvent-free conditions

Here, the application of guanidine supported on magnetic nanoparticles Fe3O4 (MNPs-guanidine) as a novel magnetically separable base nanocatalyst is described. We have investigated the application of this new catalyst for the synthesis of 2H-indazolo[2,1-b]phthalazine-trione derivatives from the three-component, one-pot condensation reaction of phthalhydrazide, cyclic 1,3-dicarbonyl, and aromatic aldehydes under solvent-free conditions. The products were obtained in short reaction times with good to high yields. The supported catalyst could be simply separated and recovered from the reaction mixture with the assistance of an external magnet and reused 18 times with little loss of activity.

Increasing the binding affinity of VEGFR-2 inhibitors by extending their hydrophobic interaction with the active site: Design, synthesis and biological evaluation of 1-substituted-4-(4-methoxybenzyl) phthalazine derivatives

A series of anilinophthalazine derivatives 4a-j was initially synthesized and tested for its VEGFR-2 inhibitory activity where it showed promising activity (IC50 = 0.636-5.76 mu M). Molecular docking studies guidance was used to improve the binding affinity for series 4a-j towards VEGFR-2 active site. This improvement was achieved by increasing the hydrophobic interaction with the hydrophobic back pocket of the VEGFR-2 active site lined with the hydrophobic side chains of Ile888, Leu889, Ile892, Val898, Val899, Leu1019 and Ile1044. Increasing the hydrophobic interaction was accomplished by extending the anilinophthalazine scaffold with a substituted phenyl moiety through an uriedo linker which should give this extension the flexibility required to accommodate itself deeply into the hydrophobic back pocket. As planned, the designed uriedo-anilinophthalazines 7a-i showed superior binding affinity than their anilinophthalazine parents (IC50 = 0.083-0.473 mu M). In particular, compounds 7g-i showed IC50 of 0.086, 0.083 and 0.086 mu M, respectively, which are better than that of the reference drug sorafenib (IC50 = 0.09 mu M). (C) 2016 Elsevier Masson SAS. Experimental and theoretical study on one-pot, three-component route to 2H-indazolo[2,1-b]phthalazine-triones catalyzed by nano-alumina sulforic acid

A simple, efficient, and high yielding one-pot protocol for the synthesis of 2H-indazolo[2,1-b]phthalazine-trione derivatives has been developed by three-component coupling of phthalhydrazide, dimedone and some aromatic aldehydes in ecofriendly neat conditions promoted by nano-gamma-alumina sulforic acid. This protocol avoids the use of expensive catalysts, toxic solvents and chromatographic separation. Also the proposed protocol was extended to the linear beta-diketone (acetylaceton) instead of cyclic diketone (dimedone). The generality and functional tolerance of this convergent and environmentally benign method is demonstrated. The nature of each step of the suggested mechanism for this condensation reaction is also identified through Density Functional Theory (DFT) calculations using B3LYP/6-31G** level of theory. It is shown that the first step is charge controlled; whereas the second one is controlled by frontier molecular orbitals. The H-1 and C-13 chemical shift values together with the structural parameters of the title compound have been also computed and the scaled values have been compared with the corresponding experimental NMR and X-ray spectra. As a result, the calculated spectroscopic data and optimized geometry show a good agreement with the experimental results. (C) 2012 Elsevier B.V. All rights reserved.

Therefore, in this paper the preparation of sulfuric acid functionalized silica-coated magnetite nanoparticles with core-shell structure (Fe3O4@silica sulfuric acid) are presented by using Fe3O4 spheres as the core and silica sulfuric acid nanoparticles as the shell. The catalyst was characterized by infrared spectroscopy (FT-IR), scanning electron microscope (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD) spectroscopy, and vibrating sample magnetometer (VSM). Ability of this nano-magnetic solid acid catalyst in the one-pot three-components condensation reaction of phthalhydrazide, aromatic aldehydes and cyclic or acyclic 1,3-diketones are also described. Utilization of easy reaction conditions, catalyst with high catalytic activity and good reusability, and simple magnetically work-up, makes this methodology as an interesting option for the economic synthesis of indazolo[2,1-b]phthalazine-triones and pyrazolo[1,2-b]phthalazine-diones.
