An efficient protocol was developed for the synthesis of pi-extended isocoumarins and polycyclic aromatic hydrocarbons based on the oxidative coupling of aromatic carboxylic acids with internal alkynes catalyzed by (cyclopentadienyl)rhodium complexes. The coupling chemoselectivity strongly depends on whether Cp or the methylated Cp* ligands are used. The pentamethyl derivative [Cp*RhCl2](2) predominantly gives isocoumarins, while the non-methylated complex [CpRhI2](n) produces naphthalene derivatives. The polyaromatic carboxylic acids (such as 1-naphthoic acid, 1-pyrenecarboxylic acid, fluorene-1-carboxylic acid, and dibenzofuran-4-carboxylic acid) are suitable for this approach. A mixture of Cp*H/RhCl3 can be used as a catalyst instead of [Cp*RhCl2](2). The structures of 3,4-diphenylindeno[1,2-h]isochromen-1(11H)-one and 7,10-dimethyl-8,9-diphenylbenzo[pqr]tetraphene were determined by X-ray diffraction. In addition, the optical properties of the prepared compounds were studied. 7,8-Diphenyl-10H-phenaleno[1,9-gh]isochromen-10-one was employed as an emissive layer for OLED manufacturing. The OLED emits yellow-green light with a maximum intensity 1740 cd . m(-2) at 15 V.

A new, modified total synthesis of (-)-cleistenolide (1) and sixteen new analogues or derivatives was achieved starting from commercially available 1,2-O-isopropylidene-alpha-D-glucofuranose. The synthesis of 1 proceeds in six steps and 67% overall yield, using single-carbon atom degradation of a protected chiral precursor, (Z)-selective Wittig olefination, and acid catalyzed delta-lactonization. A new Lewis acid promoted procedure for one-pot O-debenzylation/O-acylation has been developed to complete the synthesis of natural product 1 and selected analogues. The synthesized compounds were tested in vitro to evaluate their cytotoxicity against K562, HL-60, Jurkat, Raji, MCF-7, MDA-MB 231, HeLa, A549, and MRC-5 cell lines. All (-)-cleistenolide analogues exhibited significantly higher cytotoxicity than lead 1 against the majority of cell lines tested. Most of the synthesized compounds are more active than doxorubicin on at least one malignant cell line, but were almost completely inactive against normal MRC-5 cells. An organocatalytic asymmetric enantioselective domino oxa-Michael-Mannich-[1,3]-amino rearrangement reaction of N-tosylsalicylimines with a wide range of alpha,beta-unsaturated aldehydes utilizing diarylprolinol silyl ether catalysis is described. The catalytic reactions proceed with excellent enantioselectivity (up to 99% ee) to produce the corresponding chair N-tosylimines-chromenes with a yield of up to 99%, tolerating a range of functional groups. This methodology offers a new method with great potential to further extend the synthetic power and versatility of chiral aminocatalysis.

Multitargeted hybrids of N-benzylpiperidine and substituted 5-phenyl-1,3,4-oxadiazoles were designed, synthesized, and evaluated against Alzheimer’s disease (AD). Tested compounds exhibited moderate to excellent inhibition against human acetylcholinesterase (hAChE), butyrylcholinesterase (hBChE), and beta-secretase-1 (hBACE-1). The potential leads 6g and 10f exhibited balanced inhibitory profiles against all the targets, with a substantial displacement of propidium iodide from the peripheral anionic site of hAChE. Hybrids 6g and 10f also elicited favorable permeation across the blood -brain barrier and were devoid of neurotoxic liability toward SH-SYSY neuroblastoma cells. Both leads remarkably disassembled A beta aggregation in thioflavin T-based selfand AChE-induced experiments. Compounds 6g and 10f ameliorated scopolamine gamma-induced cognitive dysfunctions in the Ymaze test. The ex vivo studies of rat brain homogenates established the reduced AChE levels and antioxidant activity of both compounds. Compound 6g also elicited noteworthy improvement in A beta-induced cognitive dysfunctions in the Morris water maze test with downregulation in the expression of A beta and BACE-1 proteins corroborated by Western blot and immunohistochemical analysis. The pharmacokinetic study showed excellent oral absorption characteristics of compound 6g. The in silica molecular docking and dynamics simulation studies of lead compounds affirmed their consensual binding interactions with PAS-AChE and aspartate dyad of BACE-1.
