Synthesis and evaluation of anti-proliferative activity of 1,4-disubstituted phthalazines

A series of new phthalazine derivatives 3a-i and 4a-c were synthesized via the reaction of 1-chlorophthalazine derivative 2 with either N-substituted-piperazines, primary or their secondary amines. The structure of the synthesized, new compounds were characterized by spectral data. The anti-proliferative activity on human breast cancer cell line MCF-7 of the synthesized compounds was determined. The results showed that six of the test compounds (3a, 3g, 3i, and 4a-c) displayed potent cytotoxic activity ranging from 1.4 to 2.3 mu mol.

A new series of phthalazine-based 1,2,3-triazole derivatives are synthesized by the reaction of 2-methyl-3-(prop-2-yn-1-yl)-2,3dihydro- phthalazine-1,4-dione or 2,3-bis(prop-2-yn-1-yl)-2,3-dihydrophthalaz- ine-1,4-dione with aromatic azides via coppercatalyzed azide-alkyne cycloaddition reactions in the presence of metformine as a ligand. These reaction procedures have the advantages of high-to-excellent yields, short reaction times, mild experimental conditions, and operational simplicity. All the synthesized compounds are screened in vitro for their antibacterial activities against the three bacterial strains Micrococcus luteus, Pseudomonas aeruginosa, and Bacillus subtilis. Furthermore, the results of anti-bacterial activity of the synthesized compounds was investigated using molecular docking calculations.

The regulating inotropic activity of 6-substituted-oxy-[1,2,4]triazolo[3,4-a]phthalazine

In a previous research, we synthesized and evaluated the anticonvulsant activity of a series of 6-substituted-oxy-[1,2,4] triazolo[3,4-a] phthalazine compounds. Reference to other studies, we deduce that the compounds may be useful for regulating inotropic activity by effecting stroke volume, so in this study, we evaluated 20 compounds (15a-t) for their changed inotropic activity by measuring the left atrium stroke volume in heart preparations isolated from Wistar rats. Among the 20 compounds, 15j was the most potent with an increased stroke volume of 27.39 +/- 3.16 % at the concentration of 50 mu M, compound 15m was identified as the most potent with an decreased stroke volume of -95.59 +/- 0.50 % at the same concentration which is much more effective than referenced drug Nifedipine (-55.22 +/- 1.87% at the concentration of 1 mu M).

Their structures are confirmed by IR and NMR spectra as well as elemental analyses. All the compounds prepared are screened invitro for their anti-bacterial activities. This newly developed acidic catalyst was employed efficiently in a one-pot three-component condensation reaction of aromatic aldehydes, dimedone and phthalhydrazide for the synthesis of 2H-indazolo[2,1-b]phthalazine-trione derivatives through an easy and eco-friendly methodology. The catalyst was easily separated from the reaction mixture by simple filtration and the desired products were achieved in good to excellent yields in short reaction times.

Synthesis and antibacterial activity of some new 3-hydroxy-2-naphthoic acid hydrazide derivatives for dyeing polyester fabrics

Dibenzobarallene was used as a key intermediate for the synthesis of 2-(3-hydroxy-2-naphthoyl)-3,4,4a, 5,10,10a-hexahydro-1H-5,10-benzenobenzo[g]phthalazine-1,4-dione (2). The previous compound was coupled with the appropriate diazonium chloride to give the corresponding 4-arylazo-2-naphthol derivatives 3a-l. Also, nitration, nitrosation and bromination of compound 2 afforded the corresponding nitro, nitroso and bromo derivatives 4-6, respectively. The synthesized compounds were established and evaluated as antibacterial agents. The results showed clearly that compounds 2, 3a, 3c, 3d, 3g, 3i, 4, 5 and 6 exhibited interesting high activities compared with reference drugs. Also, these compounds were applied to polyester as disperse dyes in which their color measurement and fastness properties were evaluated.
