Anticancer activity assessment of two novel binuclear platinum (II) complexes

In the current study, two binuclear Pt (II) complexes, containing cis, cis-[Me2Pt (mu-NN) (mu-dppm) PtMe2] (1), and cis,cis-[Me2Pt(mu-NN)(mu dppm,) Pt((CH2)(4))] (2) in which NN = phthalazine and dppm = bis (diphenylphosphino) methane were evaluated for their anticancer activities and DNA/purine nucleotide binding properties. These Pt (II) complexes, with the non-classical structures, demonstrated a significant anticancer activity against Jurkat and MCF-7 cancer cell lines. The results of ethidium bromide/acridine orange staining and Caspase-III activity suggest that these complexes were capable to stimulate an apoptotic mechanism of cell death in the cancer cells. Using different biophysical techniques and docking simulation analysis, we indicated that these complexes were also capable to interact efficiently with DNA via a non-intercalative mechanism. According to our results, substitution of cyclopentane (in complex 2) with two methyl groups (in complex 1) results in significant improvement of the complex ability to interact with DNA and subsequently to induce the anticancer activity. Overall, these binuclear Pt (II) complexes are promising group of the non-classical potential anticancer agents which can be considered as molecular templates in designing of highly efficient platinum anticancer drugs. To understand the biological importance of heterocyclic cores, novel 1H-benzimidazol-2-yl thiomethyl incorporated hybrid compounds, 2-(benzimidazol-2-ylthiomethyl)-5-aryl-1,3,4-oxadizoles and 1-(2-(1H-benzo[d]imidazol-2-ylthio)acetyl)pyridazine/phthalazinediones were designed using molecular hybridization technique, synthesized and characterized. The compounds were screened for in vitro antimicrobial activity using the serial dilution technique and were found to exhibit weak antitubercular activity, excellent to moderate antibacterial and better antifungal activities against some tested organisms in comparison to the standard drugs. Thus, some of the title compounds demonstrated antimicrobial activity.

Introducing new and effective catalysts for the synthesis of pyridazino[1,2-a]indazole, indazolo[2,1-b]phthalazine and pyrazolo[1,2-b]phthalazine derivatives

In this study, a new and effective catalyst for the synthesis of pyridazino[1,2-a]indazole, indazolo[2,1-b]phthalazine and pyrazolo[1,2-b]phthalazine derivatives was introduced. Triethanolammonium acetate ([TEAH][OAc]) accelerates the reaction in a one-pot and four-component condensation of aldehydes, hydrazine hydrate, succinic/phthalic anhydride, and 1,3-dicarbonyl compounds. The yield of the products is high, and the reaction conditions are mild and solvent-free. Furthermore, the model reaction was conducted in the presence of triethanolammonium sulphate ([TEAH][HSO4] and triethanolammonium formate ([TEAH][HCOO]) under various conditions. In addition, the catalyst is recyclable, therefore, it can be reused several times. The structure of the obtained products was confirmed by comparing the M.P., IR, and H-1 NMR. Advantages of this technique are as following: Synthesis of novel, green, and one-pot and four-component condensation (4CC) under solvent-free conditions at room temperature. The catalytic reaction is performed under mild and environmentally friendly conditions in short reaction times and excellent yields. Design, Regiospecific Green Synthesis, Chemical Computational Analysis, and Antimicrobial Evaluation of Novel Phthalazine Heterocycles

Phthalazines have received considerable attention for their wide antimicrobial activity. Regiospecific nucleophilic attack of 4-benzylphthalazin-1-ol by the 1-oxo rather than the aza group on different alkyl halides gave novel phthalazine heterocyclic derivatives. Moreover, a variety of nucleosides bonded to electron-withdrawing groups were synthesized using 4-benzylphthalazine-1-ol. The density functional theory has been used to investigate the electronic structure of the synthesized compounds. All of the synthesized derivatives showed remarkable activity when tested against Gram-positive and Gram-negative bacteria, Aspergillus niger, and Candida albicans. The reactivity of these nucleosides was expected to arise from their bonding with the lone pair of N-atom of the macromolecules of bacteria. These bonding were expected to inhibit the enzyme by forming highly stable complex with lower highest occupied molecular orbital energy. The structures of these synthesized derivatives were established by Fourier transform infrared, H-1-NMR, and C-13-NMR spectroscopic evidence.

Phosphotungstic acid supported on SiO2@NHPhNH2 functionalized nanoparticles of MnFe2O4 as a recyclable catalyst for the preparation of tetrahydrobenzo[b]pyran and indazolo[2,1-b]phthalazine-triones

MnFe2O4@SiO2@NHPhNH2-phosphotungstic acid was synthesized by the reaction of diamine-modified silica coated manganese ferrite nanoparticles with H3PW12O40, the Keggin-type heteropoly acid. This is the first time that H3PW12O40 supported on diamine functionalized MnFe2O4 nanoparticles has been reported. The synthesized catalyst was characterized by various techniques, including Fourier Transform Infrared Spectroscopy (FTIR), Transmission electron-microscopy (TEM), X-ray Powder Diffraction (XRD), Scanning Electron Microscope (SEM), Energy Dispersive X-ray Spectroscopy (EDX), Inductively Coupled Plasma Atomic Emission Spectroscopy (ICP-AES), Thermogravimetric Analysis (TGA), and Vibrating Sample Magnetometer (VSM). The synthesized nanoparticle was examined as the heterogenous nanocatalyst for the synthesis of tetrahydrobenzo[b]-pyran derivatives and 2H-indazolo-[2,1-b]-phthalazine-triones under the solvent-free heating or ultrasonic irradiation conditions. This method offers several advantages including high yield, low reaction times, and the simple work up procedure due to the presence of the magnetic nanoparticles in this catalyst, so that, it can be easily recovered and reused for at least six successive times without the loss of its high catalytic activity, suggesting its great Potential for industrial applications. Structure-activity relationship for enantiomers of potent inhibitors of B. anthracis dihydrofolate reductase

Background: Bacterial resistance to antibiotic therapies is increasing and new treatment options are badly needed. There is an overlap between these resistant bacteria and organisms classified as likely bioterror weapons. For example, Bacillus anthracis is innately resistant to the anti-folate trimethoprim due to sequence changes found in the dihydrofolate reductase enzyme. Development of new inhibitors provides an opportunity to enhance the current arsenal of anti-folate antibiotics while also expanding the coverage of the anti-folate class. Methods: We have characterized inhibitors of B. anthracis dihydrofolate reductase by measuring the K-i and MIC values and calculating the energetics of binding. This series contains a core diaminopyrimidine ring, a central dimethoxybenzyl ring, and a dihydrophthalazine moiety. We have altered the chemical groups extended from a chiral center on the dihydropyridazine ring of the phthalazine moiety. The interactions for the most potent compounds were visualized by X-ray structure determination. Results: We find that the potency of individual enantiomers is divergent with clear preference for the S-enantiomer, while maintaining a high conservation of contacts within the binding site. The preference for enantiomers seems to be predicated largely by differential interactions with protein residues Leu29, Gln30 and Arg53. Conclusions: These studies have clarified the activity of modifications and of individual enantiomers, and highlighted the role of the less-active R-enantiomer in effectively diluting the more active S-enantiomer in racemic solutions. This directly contributes to the development of new antimicrobials, combating trimethoprim resistance, and treatment options for potential bioterrorism agents. Published by Elsevier B.V.
